    Mangrove microniches determine the structural and functional diversity of enriched petroleum hydrocarbon-degrading consortia

    In this study, the combination of culture enrichments and molecular tools was used to identify bacterial guilds, plasmids and functional genes potentially important in the process of petroleum hydrocarbon (PH) decontamination in mangrove microniches (rhizospheres and bulk sediment). In addition, we aimed to recover PH-degrading consortia (PHDC) for future use in remediation strategies. The PHDC were enriched with petroleum from rhizosphere and bulk sediment samples taken from a mangrove chronically polluted with oil hydrocarbons. Southern blot hybridization (SBH) assays of PCR amplicons from environmental DNA before enrichments resulted in weak positive signals for the functional gene types targeted, suggesting that PH-degrading genotypes and plasmids were in low abundance in the rhizosphere and bulk sediments. However, after enrichment, these genes were detected and strong microniche-dependent differences in the abundance and composition of hydrocarbonoclastic bacterial populations, plasmids (IncP-1 alpha, IncP-1 beta, IncP-7 and IncP-9) and functional genes (naphthalene, extradiol and intradiol dioxygenases) were revealed by in-depth molecular analyses [PCR-denaturing gradient gel electrophoresis and hybridization (SBH and microarray)].     The streaming kk-mismatch problem

    We consider the streaming complexity of a fundamental task in approximate pattern matching: the kk-mismatch problem. It asks to compute Hamming distances between a pattern of length nn and all length-nn substrings of a text for which the Hamming distance does not exceed a given threshold kk. In our problem formulation, we report not only the Hamming distance but also, on demand, the full \emph{mismatch information}, that is the list of mismatched pairs of symbols and their indices. The twin challenges of streaming pattern matching derive from the need both to achieve small working space and also to guarantee that every arriving input symbol is processed quickly. We present a streaming algorithm for the kk-mismatch problem which uses O(klognlognk)O(k\log{n}\log\frac{n}{k}) bits of space and spends \ourcomplexity time on each symbol of the input stream, which consists of the pattern followed by the text. The running time almost matches the classic offline solution and the space usage is within a logarithmic factor of optimal. Our new algorithm therefore effectively resolves and also extends an open problem first posed in FOCS'09. En route to this solution, we also give a deterministic O(k(lognk+logΣ))O( k (\log \frac{n}{k} + \log |\Sigma|) )-bit encoding of all the alignments with Hamming distance at most kk of a length-nn pattern within a text of length O(n)O(n). This secondary result provides an optimal solution to a natural communication complexity problem which may be of independent interest.Comment: 27 page

As a result, a chemistry professor who performs outreach activities at local elementary schools exploited this interest in mysteries as a method to spark curiosity in science. This was accomplished through the development of an outreach activity in which students solve a mystery using density and fingerprint analysis

    A review of one-step fluorescent cyanoacrylate techniques

    A review of recent research in the use of one-step fluorescent cyanoacrylate techniques is presented. Advantages and disadvantages of such techniques in comparison to two-step processes are discussed. Further studies and new experimental data are presented to aid this review: three one-step cyanoacrylate products (Lumicyano, PolyCyano UV and PECA Multiband) containing a fluorescent dye were tested to evaluate their effectiveness in developing latent fingermarks on polyethylene bags by means of a pseudo operational trial. The results were compared to the traditional two-step process of cyanoacrylate fuming followed by staining with ethanol-based basic yellow 40 (BY40). The study was conducted using sequential treatments of an initial fuming cycle, a second cycle and finally BY40 staining. LumicyanoTM and PolyCyano UV performed similarly before BY40 staining, with both providing good contrast and visibility under fluorescence. PECA Multiband, however, did not develop as many fingermarks and proved to be problematic for the fuming cabinet. Subsequent BY40 staining of fingermarks developed by all three one-step processes enabled the visualisation of new fingermarks
