    Reduction in germination rate and elevation of peroxidase activity in Zea mays>/em> seedlings due to exposure to different microcystin analogues and toxic cell free cyanobacterial crude extract

    Agricultural crop plants may come into contact with cyanobacterial toxins via spray irrigation with water contaminated with cyanobacteria/cyanobacterial toxins. Many of the bloom forming cyanobacteria are known to produce toxins amongst those the group of the microcystins, cyclic heptapeptides are the best known once. In this study the germination of Zea mays under exposure to different microcystins and cell free cyanobacterial crude extract containing microcystin-LR was investigated. The concentration used for all microcystins in this study was 5.0 μg L-1 which is well in the environmental range. The inhibition of germination was shown as well as the inhibition of root and shoot length by toxin exposure. As a sign for the generation of oxidative stress promoted by the toxins taken up, guajacol peroxidase was measured showing in most toxin exposures an elevation of peroxidase activity. This study showed that there is a potential concern a reduction in crop yield and also to human health if agriculturally important crop plants were exposed to cyanobacterial toxins via spray irrigation

    CyanoHAB Occurrence and Water Irrigation Cyanotoxin Contamination: Ecological Impacts and Potential Health Risks

    The world-wide occurrence of harmful cyanobacteria blooms “CyanoHAB” in fresh and brackish waters creates problems for all life forms. During CyanoHAB events, toxic cyanobacteria produce cyanotoxins at high levels that can cause chronic and sub-chronic toxicities to animals, plants and humans. Cyanotoxicity in eukaryotes has been mainly focused on animals, but during these last years, data, related to cyanotoxin (mainly microcystins, MCs) impact on both aquatic and terrestrials crop plants irrigated by water containing these toxins, have become more and more available. This last cited fact is gaining importance since plants could in a direct or indirect manner contribute to cyanotoxin transfer through the food chain, and thus constitute a potent health risk source. The use of this contaminated irrigation water can also have an economical impact which appears by a reduction of the germination rate of seeds, and alteration of the quality and the productivity of crop plants. The main objective of this work was to discuss the eventual phytotoxicity of cyanotoxins (microcystins) as the major agricultural impacts induced by the use of contaminated water for plant irrigation. These investigations confirm the harmful effects (ecological, eco-physiological, socio-economical and sanitary risk) of dissolved MCs on agricultural plants. Thus, cyanotoxin phytotoxicity strongly suggests a need for the surveillance of CyanoHAB and the monitoring of water irrigation quality as well as for drinking water

    Multiscale Modeling of Influenza A Virus Infection Supports the Development of Direct-Acting Antivirals

    Influenza A viruses are respiratory pathogens that cause seasonal epidemics with up to 500,000 deaths each year. Yet there are currently only two classes of antivirals licensed for treatment and drug-resistant strains are on the rise. A major challenge for the discovery of new anti-influenza agents is the identification of drug targets that efficiently interfere with viral replication. To support this step, we developed a multiscale model of influenza A virus infection which comprises both the intracellular level where the virus synthesizes its proteins, replicates its genome, and assembles new virions and the extracellular level where it spreads to new host cells. This integrated modeling approach recapitulates a wide range of experimental data across both scales including the time course of all three viral RNA species inside an infected cell and the infection dynamics in a cell population. It also allowed us to systematically study how interfering with specific steps of the viral life cycle affects virus production. We find that inhibitors of viral transcription, replication, protein synthesis, nuclear export, and assembly/release are most effective in decreasing virus titers whereas targeting virus entry primarily delays infection. In addition, our results suggest that for some antivirals therapy success strongly depends on the lifespan of infected cells and, thus, on the dynamics of virus-induced apoptosis or the host's immune response. Hence, the proposed model provides a systems-level understanding of influenza A virus infection and therapy as well as an ideal platform to include further levels of complexity toward a comprehensive description of infectious diseases

    Effects and metabolism of the phenylurea herbicide isoproturon in the submerged macrophyte Ceratophyllum demersum L.

    Phenylurea herbicides such as isoproturon (IPU) restrain photosynthesis by connection to the D1 protein in the photosynthetic apparatus in target plants such as weeds in crop fields. Direct effects of herbicides on organisms, which are not a target of the pesticide, have been examined seldom. Since a many of agriculturally used pesticides are found in surface waters in agricultural areas, we determined the effects on the photosynthetic oxygen production of the submerged macrophyte Ceratophyllum demersum using concentrations of IPU ranging from 0.2 μg/L to 200 μg/L IPU. At environmental relevant concentrations of IPU, the photosynthetic oxygen release was impaired. A reduction of the photosynthetic oxygen release showed a time dependency with the assigned herbicide concentrations. Furthermore, this study presents the first indications for metabolism of IPU in the aquatic plant C. demersum

    Enchytraeus crypticus Avoid Soil Spiked with Microplastic.

    Microplastics (MPs) of varying sizes are widespread pollutants in our environment. The general opinion is that the smaller the size, the more dangerous the MPs are due to enhanced uptake possibilities. It would be of considerably ecological significance to understand the response of biota to microplastic contamination both physically and physiologically. Here, we report on an area choice experiment (avoidance test) using Enchytraeus crypticus, in which we mixed different amounts of high-density polyethylene microplastic particles into the soil. In all experimental scenarios, more Enchytraeids moved to the unspiked sections or chose a lower MP-concentration. Worms in contact with MP exhibited an enhanced oxidative stress status, measured as the induced activity of the antioxidative enzymes catalase and glutathione S-transferase. As plastic polymers per se are nontoxic, the exposure time employed was too short for chemicals to leach from the microplastic, and as the microplastic particles used in these experiments were too large (4 mm) to be consumed by the Enchytraeids, the likely cause for the avoidance and oxidative stress could be linked to altered soil properties

    Estimating forest aboveground biomass by low density lidar data in mixed broad-leaved forests in the Italian Pre-Alps

    Background: Estimation of forest biomass on the regional and global scale is of great importance. Many studies have demonstrated that lidar is an accurate tool for estimating forest aboveground biomass. However, results vary with forest types, terrain conditions and the quality of the lidar data. Methods: In this study, we investigated the utility of low density lidar data (<2 points∙m−2) for estimating forest aboveground biomass in the mountainous forests of northern Italy. As a study site we selected a 4 km2 area in the Valsassina mountains in Lombardy Region. The site is characterized by mixed and broad-leaved forests with variable stand densities and tree species compositions, being representative for the entire Pre-Alps region in terms of type of forest and geomorphology. We measured and determined tree height, DBH and tree species for 27 randomly located circular plots (radius =10 m) in May 2008. We used allometric equations to calculate total aboveground tree biomass and subsequently plot-level aboveground biomass (mg∙ha−1). Lidar data were collected in June 2004. Results: Our results indicate that low density lidar data can be used to estimate forest aboveground biomass with acceptable accuracies. The best height results show a R2 = 0.87 from final model and the root mean square error (RMSE) 1.02 m (8.3% of the mean). The best biomass model explained 59% of the variance in the field biomass. Leave-one-out cross validation yielded an RMSE of 30.6 mg∙ha−1 (20.9% of the mean). Conclusions: Low-density lidar data can be used to develop a forest aboveground biomass model from plot-level lidar height measurements with acceptable accuracies. In order to monitoring the National Forest Inventory, and respond to Kyoto protocol requirements, this analysis might be applied to a larger area.     Case study comparing effects of microplastic derived from bottle caps collected in two cities on Triticum aestivum (Wheat)

    As plastic has become an integral component of daily life, microplastic has become a ubiquitous, unavoidable constituent of nearly all ecosystems. Besides monitoring the amount and distribution of microplastic in the environment, it is necessary to understand the possible direct effects, especially toxicity and how it is affected by environmental factors where it is discarded. The present study investigated how microplastic derived from high-density polyethylene bottle caps collected in two climatically different cities, i.e., Singapore (tropical rainforest climate) and Lahti, Finland (continental climate), affected the essential agricultural grain crop, Triticum aestivum (L.). Wheat seedlings were exposed to microplastic derived from these collected bottle caps, as well as new and artificially aged caps, for seven days. Morphological parameters, such as root and shoot length and oxidative stress development, were measured. Exposure to microplastic derived from the caps resulted in reduced seedling root and shoot lengths compared to the controls, as well as enhanced lipid peroxidation and catalase activity. With all parameters tested, microplastic derived from Lahti bottle caps exhibited more severe effects than Singapore, which was similar to that elicited by new microplastic. The Singapore microplastic had possibly leached its toxic substances before collection due to accelerated degradation promoted by the prevailing warmer climate conditions

    Balloon kyphoplasty in malignant spinal fractures: a systematic review and meta-analysis

    <p>Abstract</p> <p>Background</p> <p>Spinal fractures are a common source of morbidity in cancer patients. Balloon Kyphoplasty (BKP) is a minimally invasive procedure designed to stabilize fractures and correct vertebral deformities. We performed a meta-analysis to determine the efficacy and safety of BKP for spinal fractures in cancer patients.</p> <p>Methods</p> <p>We searched several electronic databases up to September 2008 and the reference lists of relevant publications for studies reporting on BKP in patients with spinal fractures secondary to osteolytic metastasis and multiple myeloma. Outcomes sought included pain relief, functional capacity, quality of life, vertebral height, kyphotic angle and adverse events. Studies were assessed for methodological bias, and estimates of effect were calculated using a random-effects model. Potential reasons for heterogeneity were explored.</p> <p>Results</p> <p>The literature search revealed seven relevant studies published from 2003 to 2008, none of which were randomized trials. Analysis of those studies indicated that BKP resulted in less pain and better functional outcomes, and that these effects were maintained up to 2 years post-procedure. While BKP also improved early vertebral height loss and spinal deformity, these effects were not long-term. No serious procedure-related complications were described. Clinically asymptomatic cement leakage occurred in 6% of all treated levels, and new vertebral fractures in 10% of patients. While there is a lack of studies comparing BKP to other interventions, some data suggested that BKP provided similar pain relief as vertebroplasty and a lower cement leakage rate.</p> <p>Conclusion</p> <p>It appears that there is level III evidence showing BKP is a well-tolerated, relatively safe and effective technique that provides early pain relief and improved functional outcomes in patients with painful neoplastic spinal fractures. BKP also provided long-term benefits in terms of pain and disability. However, the methodological quality of the original studies prevents definitive conclusions being drawn. Further investigation into the use of BKP for spinal fractures in cancer patients is warranted.</p

    Transcriptome Analysis of Silver Carp (Hypophthalmichthys molitrix) by Paired-End RNA Sequencing

    The silver carp (Hypophthalmichthys molitrix) is among the most intensively pond-cultured fish species and is used in the wild to counteract water bloom in China. However, little genomic information is available for this species, especially regarding its ability to grow rapidly in water, even water contaminated with high concentrations of poisonous microcystin. In this study, we performed de novo transcriptome assembly and analysis of the 17.10 million short-read sequences produced by the Illumina paired-end sequencing technology. Using an improved multiple k-mer contig assembly method coupled with further scaffolding, 85 759 sequences were obtained. There were 23 044 sequences annotated with 3423 gene ontology terms for 104 196 term occurrences and the three corresponding organizing principles. A total of 38 200 assembled sequences were involved in 218 predicted Kyoto Encyclopedia of Genes and Genomes metabolic pathways. We also recovered 41 of 44 genes involved in the biosynthesis of glutathione. Of these, five genes were identified as experienced positive selection between silver carp and zebrafish, as determined by the likelihood ratio test. This report is the first annotated review of the silver carp transcriptome. These data will be of interest to researchers investigating the evolution and biological processes of the silver carp. This work also provides an archive for future studies of recent speciation and evolution of Cyprinidae fishes and can be used in comparative studies of other fishes
