Orbital dependent nucleonic pairing in the lightest known isotopes of tin
By studying the 109Xe-->105Te-->101Sn superallowed alpha-decay chain, we
observe low-lying states in 101Sn, the one-neutron system outside doubly magic
100Sn. We find that the spins of the ground state (J = 7=2) and first excited
state (J = 5=2) in 101Sn are reversed with respect to the traditional level
ordering postulated for 103Sn and the heavier tin isotopes. Through simple
arguments and state-of-the-art shell model calculations we explain this
unexpected switch in terms of a transition from the single-particle regime to
the collective mode in which orbital-dependent pairing correlations, dominate.Comment: 5 pages 3 figure
Role of Engrailed-2 (EN2) as a prostate cancer detection biomarker in genetically high risk men
Controversy surrounds the use of PSA as a biomarker for prostate cancer detection, leaving an unmet need for a novel biomarker in this setting; urinary EN2 may identify individuals with clinically relevant prostate cancer. Male BRCA1 and BRCA2 mutation carriers are at increased risk of clinically significant prostate cancer and may benefit from screening. Urine samples from 413 BRCA1 and BRCA2 mutation carriers and controls were evaluated. Subjects underwent annual PSA screening with diagnostic biopsy triggered by PSA > 3.0â
ng/ml; 21 men were diagnosed with prostate cancer. Urinary EN2 levels were measured by ELISA and had a sensitivity of 66.7% and specificity of 89.3% for cancer detection. There was no statistically significant difference in EN2 levels according to genetic status or Gleason score. Urinary EN2 may be useful as a non-invasive early biomarker for prostate cancer detection in genetically high-risk individuals
Reorganization Energy for Internal Electron Transfer in Multicopper Oxidases.
We have calculated the reorganization energy for the intramolecular electron transfer between the reduced type 1 copper site and the peroxy intermediate of the trinuclear cluster in the multicopper oxidase CueO. The calculations are performed at the combined quantum mechanics and molecular mechanics (QM/MM) level, based on molecular dynamics simulations with tailored potentials for the two copper sites. We obtain a reorganization energy of 91-133 kJ/mol, depending on the theoretical treatment. The two Cu sites contribute by 12 and 22 kJ/mol to this energy, whereas the solvent contribution is 34 kJ/mol. The rest comes from the protein, involving small contributions from many residues. We have also estimated the energy difference between the two electron-transfer states and show that the reduction of the peroxy intermediate is exergonic by 43-87 kJ/mol, depending on the theoretical method. Both the solvent and the protein contribute to this energy difference, especially charged residues close to the two Cu sites. We compare these estimates with energies obtained from QM/MM optimizations and QM calculations in a vacuum and discuss differences between the results obtained at various levels of theory
Psychosocial impact of undergoing prostate cancer screening for men with BRCA1 or BRCA2 mutations.
OBJECTIVES: To report the baseline results of a longitudinal psychosocial study that forms part of the IMPACT study, a multi-national investigation of targeted prostate cancer (PCa) screening among men with a known pathogenic germline mutation in the BRCA1 or BRCA2 genes. PARTICPANTS AND METHODS: Men enrolled in the IMPACT study were invited to complete a questionnaire at collaborating sites prior to each annual screening visit. The questionnaire included sociodemographic characteristics and the following measures: the Hospital Anxiety and Depression Scale (HADS), Impact of Event Scale (IES), 36-item short-form health survey (SF-36), Memorial Anxiety Scale for Prostate Cancer, Cancer Worry Scale-Revised, risk perception and knowledge. The results of the baseline questionnaire are presented. RESULTS: A total of 432 men completed questionnaires: 98 and 160 had mutations in BRCA1 and BRCA2 genes, respectively, and 174 were controls (familial mutation negative). Participants' perception of PCa risk was influenced by genetic status. Knowledge levels were high and unrelated to genetic status. Mean scores for the HADS and SF-36 were within reported general population norms and mean IES scores were within normal range. IES mean intrusion and avoidance scores were significantly higher in BRCA1/BRCA2 carriers than in controls and were higher in men with increased PCa risk perception. At the multivariate level, risk perception contributed more significantly to variance in IES scores than genetic status. CONCLUSION: This is the first study to report the psychosocial profile of men with BRCA1/BRCA2 mutations undergoing PCa screening. No clinically concerning levels of general or cancer-specific distress or poor quality of life were detected in the cohort as a whole. A small subset of participants reported higher levels of distress, suggesting the need for healthcare professionals offering PCa screening to identify these risk factors and offer additional information and support to men seeking PCa screening

α2-Macroglobulin-Kallikrein Complexes Detect Contact System Activation in Hereditary Angioedema and Human Sepsis
Activation of the contact system has been documented in severe sepsis and hereditary angioedema, but a sensitive, specific, and quantitative assay for assessing the degree of involvement of this proteolytic enzyme cascade is not yet available. We have developed a quantitative sandwich enzyme-linked immunosorbent assay (ELISA) for the α2- macroglobulin-kallikrein (α2M-Kal) complex using an F(abâ)2 derivative of a monospecific polyclonal antibody against α2M as the capture antibody and a unique murine monoclonal antibody, 13G11, against the heavy chain of kallikrein as the detector antibody. The assay does not detect complexes in normal plasma but reacts with complexes generated by activating normal plasma with dextran sulfate at 4°C in a range of 5 to 375 nmol/L. A close correlation of the ELISA with an amidolytic assay for α2M-Kal was documented. Patients with sepsis syndrome but negative bacterial blood cultures did not show elevated plasma complexes, whereas a majority of those with positive blood cultures did show modest elevation and a single patient with septic shock showed a very high level of α2M-Kal complex. Similarly, a patient with classic hereditary angioedema (HAE) showed increased concentration of complexes on three separate occasions during attacks but normal levels between attacks. Two other HAE patients did not show elevated levels at quiescent periods. The ELISA for α2M-Kal appears to be sensitive, specific, and quantitative, and it can be used to reflect the degree of contact system activation in human sepsis and in HAE attacks

A luminous X-ray outburst from an intermediate-mass black hole in an off-centre star cluster
International audienceA unique signature for the presence of massive black holes in very dense stellar regions is occasional giant-amplitude outbursts of multi-wavelength radiation from tidal disruption and subsequent accretion of stars that make a close approach to the black holes 1 . Previous strong tidal disruption event (TDE) candidates were all associated with the centres of largely isolated galaxies 26 . Here, we report the discovery of a luminous X-ray outburst from a massive star cluster at a projected distance of 12.5âkpc from the centre of a large lenticular galaxy. The luminosity peaked at ~10âergâs and decayed systematically over 10âyears, approximately following a trend that supports the identification of the event as a TDE. The X-ray spectra were all very soft, with emission confined to be âČ3.0âkeV, and could be described with a standard thermal disk. The disk cooled significantly as the luminosity decreasedâa key thermal-state signature often observed in accreting stellar-mass black holes. This thermal-state signature, coupled with very high luminosities, ultrasoft X-ray spectra and the characteristic power-law evolution of the light curve, provides strong evidence that the source contains an intermediate-mass black hole with a mass tens of thousand times that of the solar mass. This event demonstrates that one of the most effective means of detecting intermediate-mass black holes is through X-ray flares from TDEs in star clusters
