    Future of European agriculture after the Health Check

    This paper identifies major future trends and driving factors and perspectives and challenges resulting from them for European agriculture and food sectors until the year 2020. The focus of the paper is an analysis of key driving forces and the provision of a well developed reference scenario under the assumption of continued CAP reform and taking into account the framework discussions in the Doha Development Round. To assess the impact of policies the paper also examines a liberalisation (no support) and regionalization (maximum support) scenario. In terms of policy options the paper shows that the structural change process in agriculture (measured in terms of agricultural share in GDP) is a long-term process that continues with or without policy changes. The EU is facing an increasing diversity of structure and structural adjustment. The livestock sector (especially cattle) faces important challenges and restructuring. Alternative policy settings may not produce very different effect on the overall production as factor markets adjust.     Following the paradigm for reforming the current CAP, the first objective of this study is to give insights into the economic impact of post-2013 CAP measures at different levels of aggregation (e.g. EU, Member State and region). The post-2013 CAP measures included are directed towards income for the farmers, competitiveness, valuable areas and ecosystem services. The second objective is to analyse the impact of a scenario that combines the above mentioned post-2013 CAP measures. This study can be seen as a first attempt to quantify the transition to a CAP with more targeted measures at the European level and reveals considerable methodological and data challenges.     Development of a telescope for medium-energy gamma-ray astronomy

    The Advanced Energetic Pair Telescope (AdEPT) is being developed at GSFC as a future NASA MIDEX mission to explore the medium-energy (5–200 MeV) gamma-ray range. The enabling technology for AdEPT is the Three- Dimensional Track Imager (3-DTI), a gaseous time projection chamber. The high spatial resolution 3-D electron tracking of 3-DTI enables AdEPT to achieve high angular resolution gamma-ray imaging via pair production and triplet production (pair production on electrons) in the medium-energy range. The low density and high spatial resolution of 3-DTI allows the electron positron track directions to be measured before they are dominated by Coulomb scattering. Further, the significant reduction of Coulomb scattering allows AdEPT to be the first medium-energy gamma-ray telescope to have high gamma-ray polarization sensitivity. We review the science goals that can be addressed with a medium-energy pair telescope, how these goals drive the telescope design, and the realization of this design with AdEPT. The AdEPT telescope for a future MIDEX mission is envisioned as a 8 m3 active volume filled with argon at 2 atm. The design and performance of the 3-DTI detectors for the AdEPT telescope are described as well as the outstanding instrument challenges that need to be met for the AdEPT mission

    Phosphorus and nitrogen limitation of primary production in a simulated estuarine gradient

    The transition between phosphorus limitation of primary production in freshwater and nitrogen limitation in seawater was examined along an estuarine gradient simulated in 4 large 13 m3 enclosures connected in a series and containing pelagic and benthic subsystems. Nominal salinities of 0, 5, 10 and 25 ppt were maintained by exchanging appropriate volumes of water between enclosures. River water, which served as a freshwater endmember, was naturally high in N relative to P, while the oceanic endmember (water from Narragansett Bay, RI, USA) was low in N relative to P. Production in the water column was supported by external inputs and recycled nutrients. Bioassays, inorganic nutrient concentrations and N:P ratios of the seston and inorganic nutrients indicated that phosphorus was limiting at 0, 5 and 10 ppt, while nitrogen was limiting at 25 ppt. Coincident with this shift in limiting nutrient was a shift in the N:P ratio of nutrient supply from greater than the Redfield ratio of 16 to less than 16. External inputs established relative rates of supply in each enclosure. The relative proportion of N and P in external inputs was largely a function of the hydrodynamic mixing of fresh (high N, low P) and salt water (low N, high P) endmembers. At the scale of the estuarine segment or enclosure, neither recycled inputs from the benthos and water column, nitrogen fixation nor internal losses of N and P to sedimentation and/or denitrification materially altered relative supply rates, despite a hydrodynamic residence time of 27 d

    Expression and cellular distribution of cyclin-dependent kinase 4 (Cdk4) and connexin 43 (Cx43) in porcine oocytes before and after in vitro maturation

    It is recognised that connexin 43 (Cx43) and cyclin-dependent kinase 4 (Cdk4) are involved in the cumulus cell-oocyte communication via gap junctions and the control of cell cycle progress. However, little is known about their mRNA expression pattern and encoded proteins distribution in porcine oocytes during in vitro maturation (IVM). Cumulus-oocyte complexes (COCs) were collected from 31 puberal crossbred Landrace gilts and analysed for their Cdk4 and Cx43 mRNA expression using RQ-PCR and for the respective protein expression by confocal microscopic observations. An increased Cdk4 and Cx43 mRNA expression was found in oocytes after IVM (P < 0.001 and P < 0.05, respectively). Confocal microscopic observations revealed a significant increase of Cdk4 protein expression in the cytoplasm of oocytes during the maturation process. The localisation of Cx43 changed from zona pellucida before to cytoplasm of oocytes after IVM. It is supposed that the increased expression of Cdk4 and Cx43 mRNA in oocytes after IVM is linked with the accumulation of a large amount of templates during the process of oocyte maturation. The translocation especially of Cx43 from the zona pellucida into the cytoplasm may be associated with a decrease in gap junction activity in fully grown porcine oocytes.     Decadal trends in the ocean carbon sink

    Measurements show large decadal variability in the rate of [Formula: see text] accumulation in the atmosphere that is not driven by [Formula: see text] emissions. The decade of the 1990s experienced enhanced carbon accumulation in the atmosphere relative to emissions, while in the 2000s, the atmospheric growth rate slowed, even though emissions grew rapidly. These variations are driven by natural sources and sinks of [Formula: see text] due to the ocean and the terrestrial biosphere. In this study, we compare three independent methods for estimating oceanic [Formula: see text] uptake and find that the ocean carbon sink could be responsible for up to 40% of the observed decadal variability in atmospheric [Formula: see text] accumulation. Data-based estimates of the ocean carbon sink from [Formula: see text] mapping methods and decadal ocean inverse models generally agree on the magnitude and sign of decadal variability in the ocean [Formula: see text] sink at both global and regional scales. Simulations with ocean biogeochemical models confirm that climate variability drove the observed decadal trends in ocean [Formula: see text] uptake, but also demonstrate that the sensitivity of ocean [Formula: see text] uptake to climate variability may be too weak in models. Furthermore, all estimates point toward coherent decadal variability in the oceanic and terrestrial [Formula: see text] sinks, and this variability is not well-matched by current global vegetation models. Reconciling these differences will help to constrain the sensitivity of oceanic and terrestrial [Formula: see text] uptake to climate variability and lead to improved climate projections and decadal climate predictions

    Design of nucleotide-mimetic and non-nucleotide inhibitors of the translation initiation factor eIF4E: Synthesis, structural and functional characterisation.

    Eukaryotic translation initiation factor 4E (eIF4E) is considered as the corner stone in the cap-dependent translation initiation machinery. Its role is to recruit mRNA to the ribosome through recognition of the 5'-terminal mRNA cap structure (m7GpppN, where G is guanosine, N is any nucleotide). eIF4E is implicated in cell transformation, tumourigenesis, and angiogenesis by facilitating translation of oncogenic mRNAs; it is thus regarded as an attractive anticancer drug target. We have used two approaches to design cap-binding inhibitors of eIF4E by modifying the N7-substituent of m7GMP and replacing the phosphate group with isosteres such as squaramides, sulfonamides, and tetrazoles, as well as by structure-based virtual screening aimed at identifying non-nucleotide cap-binding antagonists. Phosphomimetic nucleotide derivatives and highly ranking virtual hits were evaluated in a series of in vitro and cell-based assays to identify the first non-nucleotide eIF4E cap-binding inhibitor with activities in cell-based assays, N-[(5,6-dihydro-6-oxo-1,3-dioxolo[4,5-g]quinolin-7-yl)methyl]-N'-(2-methyl-propyl)-N-(phenyl-methyl)thiourea (14), including down-regulation of oncogenic proteins and suppression of RNA incorporation into polysomes. Although we did not observe cellular activity with any of our modified m7GMP phosphate isostere compounds, we obtained X-ray crystallography structures of three such compounds in complex with eIF4E, 5'-deoxy-5'-(1,2-dioxo-3-hydroxycyclobut-3-en-4-yl)amino-N7-methyl-guanosine (4a), N7-3-chlorobenzyl-5'-deoxy-5'-(1,2-dioxo-3-hydroxy-cyclobut-3-en-4-yl)amino-guanosine (4f), and N7-benzyl-5'-deoxy-5'-(trifluoromethyl-sulfamoyl)guanosine (7a). Collectively, the data we present on structure-based design of eIF4E cap-binding inhibitors should facilitate the optimisation of such compounds as potential anticancer agents

    Maternal prenatal external locus of control and reduced mathematical and science abilities in their offspring:A Longitudinal Birth Cohort Study

    A personality scale that identifies individuals’ general attitude to what happens to them as largely a matter of luck or fate or of powerful others (externality) or whether they feel they can influence the consequences (internality) is known as locus of control (LOC). A continuous scale can distinguish those who are more external from those who are more internal. Lower scholastic achievement is associated with externality and higher achievement with internality, but little is known about the association of parental LOC on children’s academic performance. Data collected within the Avon Longitudinal Study of Parents and Children (ALSPAC) are analyzed to assess associations between mothers’ LOC orientation, measured during pregnancy, and their children’s abilities in mathematics and science reasoning. We found that maternal external LOC is associated with lower scores for her child assessed by tests measuring mental arithmetic as well as understanding of mathematical and scientific concepts. Additionally, we determined the extent to which three separate sets of factors previously found to positively influence the developing child’s ability mediate these findings: (a) perinatal and infant exposures, such as prenatal smoking, binge drinking, consumption of oily fish, and postnatal breast feeding; (b) parenting attitudes and strategies; and (c) the interface of the parents with their child’s school. The three factors identify at least 50% of the mechanism by which maternal externality is associated with poor academic outcomes in her child and may be candidates for further investigation as possible intervention targets
