    Monitoring environmental features using leaf stomatal characteristics

    It is recognised that atmospheric CO(_2) concentrations are increasing, with concerns raised as to the global impact continued rises may have. Plant stomatal parameters have been suggested as a means of monitoring changing CO(_2) levels. In order to assess their suitability for use, a thorough examination of leaf characteristics within a variety of surroundings was undertaken. Environments examined included both natural variations in CO(_2), which incorporated a variety of environmental influences and artificially enhanced ones where it was possible to monitor specific responses to variable CO(_2) levels. A short term study of Ranunculus ficaria, Sambucus nigra and Hedera helix revealed considerable variation in stomatal parameters, with inconsistent responses observed between and within species. These variations could not be attributed to CO(_2) changes and were assigned to a combination of other factors. R. ficaria grown in an artificial environment at ambient and elevated CO(_2) produced significant and consistent changes in stomatal parameters. Stomatal density and index were found to be reduced at elevated CO(_2) concentrations, with a decrease in guard cell dimensions. Salix herbacea, growing along a naturally reduced CO(_2) partial pressure gradient further highlighted the extent of variation in stomatal parameters in extant material within a species. No consistent stomatal trends associated with changing CO(_2) levels were observed. This inherent variation must be considered before any conclusions can be made regarding stomatal parameters obtained from fossil material. The novel application of molecular biology techniques to identify S. herbacea leaves was partially successful. However, identification of leaf macrofossils was not possible, due to the inferior preservation of DNA. However, the technique will provide a useful tool for identification if suitably preserved macrofossil fragments were available. S. herbacea leaf macrofossils revealed no consistent correlation to past changes in CO(_2) levels. The use of as an additional tool with which to monitor environmental change once again showed variation, and was not associated closely with changes in CO(_2) levels or stomatal parameters. In light of this work it would appear that the use of stomatal parameters is of limited value as a model to monitor environmental change, in the absence of information concerning other variables, and as such must be treated with caution

    Calls for Police Service: Understanding the demand profile and the UK police response

    Purpose – The purpose of this research paper is to identify and understand the demand profile of a division of Lancashire Police Constabulary in order to assist Police administrators in the evaluation of demand and understanding of Policing response. Design/methodology/approach – Police data records regarding all offences within the Central Division of Lancashire Police Constabulary over the year of 2013 were examined. Descriptive statistics were used to identify the top ten demand addresses in five categories (residential, statutory bodies, retail, nightlife, and young people’s services), and the incident classifications, time frames and response codes that were most associated with these addresses. Further content analysis was conducted on the top ten residential addresses in order to identify specific characteristics of residential demand. Findings - The majority of Lancashire Constabulary resources are being spent dealing with non-traditional police demand. The most demanding residents were found to have one or more of the following properties; (i) to be of white – Northern European origin, (ii) to be unemployed, (iii) to be associated with mental health issues, domestic violence incidents or substance abuse, and (iv) to have previously had other agency involvement. Practical Implications - These results indicate that the police could potentially reduce the demand for service by enhancing work within two key areas; partnership working and early intervention. Originality/value – In this time of austerity and police budget cuts, it is important to understand the demand on the UK police service in an attempt to reduce it

    Lymph node homing cells biologically enriched for γδ T cells express multiple genes from the T19 repertoire

    Sheep γδ T cells have been shown serologically to express T19, a membrane protein of 180-200 kDa which is a member of the scavenger receptor superfamily. Previous work from this laboratory resulted in the detection of a multigene family of T19-like genes in the sheep genome. In this study nucleotide sequences from several T19 genes were determined and are reported along with the corresponding segments of a number of expressed mRNA molecules. A segment of a single sheep T19-like gene was sequenced and these data, along with the corresponding sequences from cloned T19-like cDNA molecules from sheep and cow, were used to design an ollgonucleotide primer system suitable for amplification of corresponding segments of many T19 genes and their cDNAs. Between 30 and 40% of cloned T19 genes were amenable to amplification using the selected primers, and sequence analysis of cloned PCR products confirmed that different T19 genes encode unique amino acid sequences.     Outcomes in ANCA-Associated Vasculitis in Scotland: Validation of the Renal Risk Score in a Complete National Cohort

    INTRODUCTION: Antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) causes autoimmune-mediated inflammation of small blood vessels in multiple organs, including the kidneys. The ability to accurately predict kidney outcomes would enable a more personalized therapeutic approach. METHODS: We used our national renal biopsy registry to validate the ability of ANCA Renal Risk Score (ARRS) to predict end-stage kidney disease (ESKD) for individual patients. This score uses histopathological and biochemical data to stratify patients as high, medium, or low risk for developing ESKD. RESULTS: A total of 288 patients were eligible for inclusion in the study (low risk CONCLUSION: The ARRS better discriminates risk of ESKD in AAV and offers clinicians more prognostic information than the use of standard biochemical and clinical measures alone. This is the first time the ARRS has been validated in a national cohort. The proportion of patients with high-risk scores is lower in our cohort compared to others and should be noted as a limitation of this study

    Pandem-icons — exploring the characteristics of highly visible scientists during the Covid-19 pandemic

    The Covid-19 pandemic escalated demand for scientific explanations and guidance, creating opportunities for scientists to become publicly visible. In this study, we compared characteristics of visible scientists during the first year of the Covid-19 pandemic (January to December 2020) across 16 countries. We find that the scientists who became visible largely matched socio-cultural criteria that have characterised visible scientists in the past (e.g., age, gender, credibility, public image, involvement in controversies). However, there were limited tendencies that scientists commented outside their areas of expertise. We conclude that the unusual circumstances created by Covid-19 did not change the phenomenon of visible scientists in significant way

    Informing efficient randomised controlled trials: Exploration of challenges in developing progression criteria for internal pilot studies

    Objectives: Designing studies with an internal pilot phase may optimise the use of pilot work to inform more efficient randomised controlled trials (RCTs). Careful selection of preagreed decision or 'progression' criteria at the juncture between the internal pilot and main trial phases provides a valuable opportunity to evaluate the likely success of the main trial and optimise its design or, if necessary, to make the decision not to proceed with the main trial. Guidance on the appropriate selection and application of progression criteria is, however, lacking. This paper outlines the key issues to consider in the optimal development and review of operational progression criteria for RCTs with an internal pilot phase. Design: A structured literature review and exploration of stakeholders' opinions at a Medical Research Council (MRC) Hubs for Trials Methodology Research workshop. Key stakeholders included triallists, methodologists, statisticians and funders. Results: There is considerable variation in the use of progression criteria for RCTs with an internal pilot phase, although 3 common issues predominate: trial recruitment, protocol adherence and outcome data. Detailed and systematic reporting around the decisionmaking process for stopping, amending or proceeding to a main trial is uncommon, which may hamper understanding in the research community about the appropriate and optimal use of RCTs with an internal pilot phase. 10 top tips for the development, use and reporting of progression criteria for internal pilot studies are presented. Conclusions: Systematic and transparent reporting of the design, results and evaluation of internal pilot trials in the literature should be encouraged in order to facilitate understanding in the research community and to inform future trials

    Fulfilling global marine commitments; lessons learned from Gabon

    As part of the Post-2020 Biodiversity Framework, nations are assessing progress over the past decade in addressing the underlying drivers that influence direct pressures on biodiversity and formulating new policies and strategies for the decade to come. For marine conservation, global marine protected area (MPA) coverage is still falling short of the 10% target set in 2010. Here we show that while this reflects a lack of progress in many low- and middle-income countries, a few of these nations have met or exceeded international commitments. To provide an in-depth explanation of how this was achieved in Gabon, we summarize the lessons learnt by our consortium of policy makers and practitioners who helped implement a comprehensive and ecologically representative network of 20 MPAs. We show the importance of creating a national framework, building long-term stakeholder support, and focusing on research that guides implementation and policy; and outline a four-step approach that countries and donors could use as an example to help meet international commitments. By responding to calls to share lessons learned to inform future Convention on Biological Diversity targets, we show how Gabon's experiences could inform change elsewhere.Output Status: Forthcoming/Available Online Additional co-authors: Tim Collins, Philip D.
