    Intoxicated eyewitnesses:the effects of alcohol on eyewitness recall across repeated interviews

    The study evaluated the usefulness of repeat-interviewing of witnesses to crimes who were intoxicated by alcohol at the time of the incident and their first interview, and then re-interviewed when not intoxicated the following day. Sixty young, social drinkers were divided into three groups. One group was given a “placebo” (alcohol-like) beverage, a second was given a “low dose” of alcohol (0.2 g/kg men; 0.17 g/kg women), and a third was given a “high dose” of alcohol (0.6 g/kg men; 0.52 g/kg women) over a 15 minute period. Twenty minutes later they viewed a 4-minute video of a crime, and afterwards they were given two opportunities to recall everything that they could remember from the incident; the first opportunity was immediately after the event, and the second was 24 hours later. Analyses of the quantity and accuracy of the details recalled revealed no overall increase in the total amount of information recalled between the first and second recall opportunities. However, on average, 18% of the details recalled by participants in the second test were new and accurate. The incidence of contradictions between the first and second recall opportunities was less than 1%. Surprisingly, none of the effects were influenced by alcohol, even at the highest dose. The results imply that 1) memory for at least some incidents observed under the influence of alcohol is resilient even up to relatively high blood-alcohol levels; and 2) the repeated interviewing of witnesses who were intoxicated at the time of the crime can reveal additional, reliable information that is not present at the initial interview, just as is the case for non-intoxicated witnesses

    ISAC6+ Delivering Smarter Administration through innovation - a Benefits Realisation approach to ensuring success.

    The paper describes how the Project Management discipline of benefits realisation has been applied to an EU funded E-government initiative. It explores the benefits of using this approach, the challenges to be addressed, and suggest a framework for applying the approach to other local and national e-government initiatives. One of the key project objectives is to demonstrate through the pilot that implementation of the iSAC6+ system will provide value for money by delivering the desired benefits both to government office users and citizens. The approach described here focuses on costs and benefits generated by use of the system. There are staff costs for training, support and operation, technical costs for integrating iSAC6+ in to existing systems and websites, and more significantly organisational costs for designing and implementing new procedures and working practices. Citizens too will incur costs to access and use the service. In iSAC6+ we have created a model of costs and benefits which can be applied in the short term to the pilot, and in the longer term to a much larger number of public organisations. The aim of the Benefits Realisation model is to demonstrate that iSAC6+ is capable of delivering value for money, and thus to justify the investment needed for expanding its use. Information Technology project success or failure is traditionally judged against objectives set during initial project planning. Enterprises, both public and private sector, have found this approach inadequate because long term costs and benefits do not occur until after the project has completed. Benefits Realisation emerged in the 1990s and developed two roles: a discipline for anticipating and quantifying the expected value of a project in terms of the costs and benefits which will accrue after the project itself is complete; and an over-arching project management philosophy. The paper uses the case study experience to comment upon these two different perspectives. The model developed within the project is based upon the recognised public sector costing formula, the Standard Cost model (SCM) but goes much further by integrating it into a Benefits Realisation tool which creates an audit trail from organisational strategic aims through to detailed cost measures for both quantitative and qualitative incidences

    Survivin as a therapeutic target in Sonic hedgehog-driven medulloblastoma.

    Medulloblastoma (MB) is a highly malignant brain tumor that occurs primarily in children. Although surgery, radiation and high-dose chemotherapy have led to increased survival, many MB patients still die from their disease, and patients who survive suffer severe long-term side effects as a consequence of treatment. Thus, more effective and less toxic therapies for MB are critically important. Development of such therapies depends in part on identification of genes that are necessary for growth and survival of tumor cells. Survivin is an inhibitor of apoptosis protein that regulates cell cycle progression and resistance to apoptosis, is frequently expressed in human MB and when expressed at high levels predicts poor clinical outcome. Therefore, we hypothesized that Survivin may have a critical role in growth and survival of MB cells and that targeting it may enhance MB therapy. Here we show that Survivin is overexpressed in tumors from patched (Ptch) mutant mice, a model of Sonic hedgehog (SHH)-driven MB. Genetic deletion of survivin in Ptch mutant tumor cells significantly inhibits proliferation and causes cell cycle arrest. Treatment with small-molecule antagonists of Survivin impairs proliferation and survival of both murine and human MB cells. Finally, Survivin antagonists impede growth of MB cells in vivo. These studies highlight the importance of Survivin in SHH-driven MB, and suggest that it may represent a novel therapeutic target in patients with this disease

    Characterization of Nanoparticle Batch-To-Batch Variability

    A central challenge for the safe design of nanomaterials (NMs) is the inherent variability of NM properties, both as produced and as they interact with and evolve in, their surroundings. This has led to uncertainty in the literature regarding whether the biological and toxicological effects reported for NMs are related to specific NM properties themselves, or rather to the presence of impurities or physical effects such as agglomeration of particles. Thus, there is a strong need for systematic evaluation of the synthesis and processing parameters that lead to potential variability of different NM batches and the reproducible production of commonly utilized NMs. The work described here represents over three years of effort across 14 European laboratories to assess the reproducibility of nanoparticle properties produced by the same and modified synthesis routes for four of the OECD priority NMs (silica dioxide, zinc oxide, cerium dioxide and titanium dioxide) as well as amine-modified polystyrene NMs, which are frequently employed as positive controls for nanotoxicity studies. For 46 different batches of the selected NMs, all physicochemical descriptors as prioritized by the OECD have been fully characterized. The study represents the most complete assessment of NMs batch-to-batch variability performed to date and provides numerous important insights into the potential sources of variability of NMs and how these might be reduced

    La troponine T ultrasensible : un nouvel outil diagnostic pour le médecin sportif?

    peer reviewedIntroduction : Le risque d’accidents cardiaques ou de mort subite après effort physique intense est bien connu. Ces évènements indésirables se produisent souvent chez des sujets présentant une maladie coronarienne asymptomatique et ignorée. Néanmoins, vu ce risque, l’American Heart Association recommande de réaliser un screening cardiovasculaire chez les athlètes de tout âge. Dans cette optique, le dosage de marqueurs cardiaques de nouvelle génération, plus sensibles, comme la troponine T ultrasensible (hsTnT) peut certainement apporter des informations très intéressantes par la détection de dommages myocardiques mêmes mineurs. Matériels et méthodes : Des 20 sujets masculins volontaires âgés de 22.36±2.02 années, sédentaires, 8 ont dû être exclus (abandon, malaise à l’effort...). La VO2max a été préalablement déterminée lors d’un test à l’effort sur cycloergomètre une semaine avant le test afin de ne pas interférer avec les résultats de l’effort physique intense (EPI) pour lequel les sujets ont couru sur tapis roulant durant 1 heure à 75% de la VO2max. Quatre échantillons sanguins de 5 ml (tube hépariné-lithium) ont été prélevés : juste avant (T1), directement après (T2), 4 heures après (T3) et 24 heures après l’EPI (T4). Le dosage de hsTnT (Modular de Roche Diagnostic®) est réalisé sur du plasma par une technique d’électrocheminiluminescence. Résultats : Une augmentation statistiquement significative des résultats à T3 (p<0.01) est observée. L’élévation de la hsTnT est progressive pour atteindre un pic maximum à T3 et revenir dans les normes à T4. Le seuil critique de 0.03 ng/mL a été retenu et 75% des sujets présentent un taux supérieur à ce dernier à T3 (moyenne : 0.053 ng/mL), alors que 100% des sujets se trouvent en dessous de ce seuil à T1 (0.0041 ng/mL). Discussion - Conclusions : Ces résultats, extrêmement intéressants, suggèrent que la libération de hsTnT serait due soit à un processus physiologique de remodelage, soit à un processus irréversible de lésions au niveau des cardiomyocytes (nécrose). Il est également possible que cette élévation des troponines soit due à une libération à partir du pool cytosolique mais aussi elle peut être la conséquence de dommages membranaires potentiellement induits par le stress oxydatif. A l’issue de cette étude, nous démontrons que la hsTnT peut être un nouvel outil diagnostic dans le domaine de la cardiologie du sport

    La investigación tuvo como objetivo determinar la relación entre clima laboral y estrés laboral en trabajadores de una empresa Pesquera del distrito de Chimbote, la metodología empleada fue de tipo descriptivo, enfoque cuantitativo y diseño no experimental correlacional. En los resultados se determinó que en el nivel del clima laboral que perciben los trabajadores en la empresa pesquera, que el 40% de los trabajadores calificaron un nivel alto con el clima laboral en la pesquera, debido a que se sientes augusto trabajando en sus puestos; por otro lado, mientras que el 40% de los encuestados respondieron con un nivel bajo ante el tema clima laboral debido a que no se siente cómodos; se determinó que en la variable estrés laboral se aprecia que los trabajadores se distribuyen en mayor grado en el nivel bajo y medio (60%), una distribución similar se aprecia en cada una de las dimensiones que estructuran la variable estrés laboral, donde los trabajadores se distribuyen en mayor medida en el nivel bajo y medio (54% a 60%). Como conclusión se determinó que el grado de correlación que hay entre el clima laboral y estrés laboral es de 0.847, teniendo una relación positiva muy fuerte

    Are owl pellets good estimators of prey abundance?

    Some ecologists have been skeptics about the use of owl pellets to estimate small mammal's fauna. This is due to the assumptions required by this method: (a) that owls hunt at random, and (b) that pellets represent a random sample from the environment. We performed statistical analysis to test these assumptions and to assess the effectiveness of Barn owl pellets as a useful estimator of field abundances of its preys. We used samples collected in the arid Extra-Andean Patagonia along an altitudinal environmental gradient from lower Monte ecoregion to upper Patagonian steppe ecoregion, with a mid-elevation ecotone. To test if owls hunt at random, we estimated expected pellet frequency by creating a distribution of random pellets, which we compared with data using a simulated chi-square. To test if pellets represent a random sample from the environment, differences between ecoregions were evaluated by PERMANOVAs with Bray–Curtis dissimilarities. We did not find evidence that owls foraged non-randomly. Therefore, we can assume that the proportions of the small mammal's species in the diet are representative of the proportions of the species in their communities. Only Monte is different from other ecoregions. The ecotone samples are grouped with those of Patagonian steppes. There are no real differences between localities in the small mammal's abundances in each of these ecoregions and/or Barn owl pellets cannot detect patterns at a smaller spatial scale.
