The role of macrophages in cancer progression
In recent years, it has been demonstrated that the tumor microenvironment is a fundamental piece in the progression of cancer and thus, it emerges as a potential therapeutic target. This microenvironment contains non-malignant cell lines, including tumor-associated macrophages (TAMs), which play a key role in the control or promotion of tumor growth, metastasis and angiogenesis. Therefore, understanding how TAMs interact with tumor cells is critical to understand cancer development.
The fruit fly Drosophila melanogaster is an excellent genetically tractable model system to address the complex cell interactions and genetic cooperation that lead to tumor formation and progression. In this respect, Drosophila embryonic macrophages (hemocytes) have emerged as a powerful system to study the innate immune response against the early stages of tumor development. Deriving from the head mesoderm, hemocytes are highly migratory cells, achieving an even distribution throughout the embryo by the end of embryogenesis.
The aim of this study is to create a tumor in an embryonic tissue and analyse the response of embryonic macrophages to the presence of tumor. To do so, we have used the Gal4/UAS system to test several of the most representative oncogenes in human cancers, such as Ras, Notch, EGFR, PI3K or Yorkie, for their ability to generate a tumor in the salivary glands. The choice of this tissue relies on the fact that salivary glands develop in the head region of the embryo, the place where embryonic macrophages emerge from. Thus, one could test whether tumors recruit macrophages by analysing if they halt at salivary glands containing a tumor. Our analysis in fixed tissue shows that indeed macrophages stop their migration at salivary glands-containing tumors and wrap them. We are now in the process of analysing this behaviour at the cellular level in vivo by confocal microscopy.
This project also proposes to compare at a genetic level the behaviour of wild type and TAMs macrophages. To perform this a protocol for the isolation of macrophages by flow cytometry is being established. Once control and experimental macrophages are isolated, RNA-seq will be carried out to analyse the gene expression profile of both types of macrophages.This will allow us to identify genes and pathways involved in the regulation of tumor progression by macrophages

Taking the pulse of Earth's tropical forests using networks of highly distributed plots
Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science. Investigating the Metabolism of Estrogens in Ligilactobacillus salivarius Strains Isolated from Human Milk and Vaginal Microbiota

Nevertheless, the role of potentially probiotic microorganisms in estrogen metabolism has not been investigated so far. In this work, we have explored the ability of six Ligilactobacillus salivarius strains isolated from human milk and vaginal samples to degrade and/or conjugate parental estrogens in vitro and under aerobic conditions. The quantification of estrogens and their derivatives was carried out in cell-free supernatants by LCQQQ-MS. All the tested L. salivarius strains achieved an average degradation rate of estrone and estriol of 98% and 55%, respectively, whereas 17β-estradiol was preferentially conjugated (up to 40%). The presence of seven out of ten genes encoding enzymes relevant for estrogen metabolism was further confirmed by PCR, highlighting their genetic potential for degrading, conjugating and/or deconjugating estrogens. The tested L. salivarius strains may be considered potential probiotics affecting the fate of endogenous estrogens. Biodiversidad 2015. Reporte de Estado y Tendencias de la Biodiversidad Continental de Colombia
El propósito de este documento es fortalecer la capacidad de agentes públicos y privados para la aplicación de la PNGIBSE, que constituye en sí misma una apuesta de interfaz entre ciencia, política y sociedad en la
perspectiva de construir sostenibilidad en el desarrollo. Además de ello, representa un insumo para el seguimiento a los compromisos del país frente a convenios e iniciativas internacionales (CDB, IPBES, OCDE), así
como un mecanismo pedagógico para generar interés, conciencia y apropiación de las diferentes dimensiones
de la biodiversidad del país.
En esta oportunidad, el reporte avanza en el desarrollo de nuevas infografías vinculadas con diferentes fuentes de información que garantizan la calidad de los datos con los que se propone y representa el estado de la biodiversidad y de su gestión en Colombia. Incluye como novedad la presentación de material en portal web dedicado (reporte.humboldt.org.co), de manera que todas las personas puedan apropiarse e interactuar con el contenido de manera más efectiva. La perspectiva es construir un modelo en tiempo real del trabajo que se desarrolla en el país para conocer, proteger, utilizar o restaurar su biodiversidad y servicios ecosistémicos, promoviendo la apropiación de todos los ciudadanos e instituciones en la tarea.
Biodiversidad 2015, es evidente, aún no está en capacidad de dar cuenta de todos los procesos de gestión de biodiversidad que se dan en Colombia, liderados por múltiples actores y a todas las escalas. Paulatinamente esperamos poder abrir el espacio para incrementar la visibilidad de todas y cada una de las iniciativas que se adelanten, a sabiendas de que la posibilidad de encontrar nuevas respuestas y mejores prácticas no depende del Instituto ni de ninguna instancia en particular, sino de la capacidad colectiva de aprendizaje. Summary: Background: Anti-COVID-19 hyperimmune immunoglobulin (hIG) can provide standardized and controlled antibody content. Data from controlled clinical trials using hIG for the prevention or treatment of COVID-19 outpatients have not been reported. We assessed the safety and efficacy of subcutaneous anti-COVID-19 hyperimmune immunoglobulin 20% (C19-IG20%) compared to placebo in preventing development of symptomatic COVID-19 in asymptomatic individuals with SARS-CoV-2 infection. Methods: We did a multicentre, randomized, double-blind, placebo-controlled trial, in asymptomatic unvaccinated adults (≥18 years of age) with confirmed SARS-CoV-2 infection within 5 days between April 28 and December 27, 2021. Participants were randomly assigned (1:1:1) to receive a blinded subcutaneous infusion of 10 mL with 1 g or 2 g of C19-IG20%, or an equivalent volume of saline as placebo. The primary endpoint was the proportion of participants who remained asymptomatic through day 14 after infusion. Secondary endpoints included the proportion of individuals who required oxygen supplementation, any medically attended visit, hospitalisation, or ICU, and viral load reduction and viral clearance in nasopharyngeal swabs. Safety was assessed as the proportion of patients with adverse events. The trial was terminated early due to a lack of potential benefit in the target population in a planned interim analysis conducted in December 2021. ClinicalTrials.gov registry: NCT04847141. Findings: 461 individuals (mean age 39.6 years [SD 12.8]) were randomized and received the intervention within a mean of 3.1 (SD 1.27) days from a positive SARS-CoV-2 test. In the prespecified modified intention-to-treat analysis that included only participants who received a subcutaneous infusion, the primary outcome occurred in 59.9% (91/152) of participants receiving 1 g C19-IG20%, 64.7% (99/153) receiving 2 g, and 63.5% (99/156) receiving placebo (difference in proportions 1 g C19-IG20% vs. placebo, −3.6%; 95% CI -14.6% to 7.3%, p = 0.53; 2 g C19-IG20% vs placebo, 1.1%; −9.6% to 11.9%, p = 0.85). None of the secondary clinical efficacy endpoints or virological endpoints were significantly different between study groups. Adverse event rate was similar between groups, and no severe or life-threatening adverse events related to investigational product infusion were reported. Interpretation: Our findings suggested that administration of subcutaneous human hyperimmune immunoglobulin C19-IG20% to asymptomatic individuals with SARS-CoV-2 infection was safe but did not prevent development of symptomatic COVID-19.
