    THESEUS1 and RALF34 monitor cell wall integrity

    The cell wall is a rigid network being the first barrier between a plant cell and its environment, but at the same time has to be a dynamic network whose cell growth and shape is given by deposition and remodeling of the cell wall. Maintaining cell wall integrity (CWI) is essential for correct plant development and stress response. Members of the family of Catharanthus roseus receptor-like kinase 1-like (CrRLK1L) proteins have been shown to play a role in cell wall homeostasis, mechanoperception, CWI maintenance and growth control. One of the 17 members in Arabidopsis, THESEUS1 (THE1), was identified in a suppressor screen of a cellulose deficient mutant, revealing that the reduction in growth is part of a THE1-mediated compensatory response to cell wall perturbation (Hématy et al., 2007). Interestingly, several CrRLK1L members have been shown to be receptors for Rapid Alkalinisation Factor (RALF) peptides. RALFs are on average 50 amino acids highly basic, cysteine-rich peptides, most of which are predicted to be cleaved from a highly acidic prodomain by a subtilisin protease. Recently, THE1 has been identified as a receptor for RALF34 (Gonneau et al., 2018). However, this peptide might not be the only THE1 ligand, since ralf34 loss-of-function mutants do not phenocopy all aspects of the1 mutants. RALF24 and RALF31 clustered together with RALF34, based on expression values across different tissues. We generated CRISPR/Cas9 mutants on RALF24, RALF31 and RALF34 with the aim to study which THE1 responses depend on these peptides. Our data suggest that RALF34 could not be the ligand for CWI response of THE1.     Metabolic consequences of chronic sleep restriction in rats:Changes in body weight regulation and energy expenditure

    Epidemiological studies have shown an association between short or disrupted sleep and an increased risk to develop obesity. In animal studies, however, sleep restriction leads to an attenuation of weight gain that cannot be explained by changes in energy intake. In the present study, we assessed whether the attenuated weight gain under conditions of restricted sleep is a consequence of an overall increase in energy expenditure. Adult male rats were subjected to a schedule of chronic sleep restriction (SR) for 8 days with a 4 h window of unrestricted rest per day. Electroencephalogram and electromyogram recordings were performed to quantify the effect of the sleep restriction schedule on sleep-wake patterns. In a separate experiment, we measured sleep restriction-induced changes in body weight, food intake, and regulatory hormones such as glucose, insulin, leptin and corticosterone. To investigate whether a change in energy expenditure underlies the attenuation of weight gain, energy expenditure was measured by the doubly labeled water method from day 5 until day 8 of the SR protocol. Results show a clear attenuation of weight gain during sleep restriction but no change in food intake. Baseline plasma glucose, insulin and leptin levels are decreased after sleep restriction which presumably reflects the nutritional status of the rats. The daily energy expenditure during SR was significantly increased compared to control rats. Together, we conclude that the attenuation of body weight gain in sleep restricted rats is explained by an overall increase in energy expenditure together with an unaltered energy intake.     Microbial biopesticides for integrated crop management : an assessment of environmental and regulatory sustainability

    Herbivorous insects and mites, plant diseases and weeds are major impediments to the production of food crops and are increasingly difficult to control with conventional chemicals. This paper focuses on microbial control agents with an emphasis on augmentation. There are marked differences in the availability of products in different countries which can be explained in terms of differences in their regulatory systems. Regulatory failure arises from the application of an inappropriate synthetic pesticides model. An understanding of regulatory innovation is necessary to overcome these problems. Two attempts at remedying regulatory failure in the UK and the Netherlands are assessed. Scientific advances can feed directly into the regulatory process and foster regulatory innovation

    Λ\LambdaCDM not dead yet: massive high-z Balmer break galaxies are less common than previously reported

    Early JWST observations that targeted so-called double-break sources (attributed to Lyman and Balmer breaks at z>7z>7), reported a previously unknown population of very massive, evolved high-redshift galaxies. This surprising discovery led to a flurry of attempts to explain these objects' unexpected existence including invoking alternatives to the standard Λ\LambdaCDM cosmological paradigm. To test these early results, we adopted the same double-break candidate galaxy selection criteria to search for such objects in the JWST images of the CAnadian NIRISS Unbiased Cluster Survey (CANUCS), and found a sample of 19 sources over five independent CANUCS fields that cover a total effective area of 60\sim60\,arcmin2^2 at z8z\sim8. However, (1) our SED fits do not yield exceptionally high stellar masses for our candidates, while (2) spectroscopy of five of the candidates shows that while all five are at high redshifts, their red colours are due to high-EW emission lines in star-forming galaxies rather than Balmer breaks in massive, evolved systems. Additionally, (3) field-to-field variance leads to differences of 1.5\sim 1.5 dex in the maximum stellar masses measured in the different fields, suggesting that the early single-field JWST observations may have suffered from cosmic variance and/or sample bias. Finally, (4) we show that the presence of even a single massive outlier can dominate conclusions from small samples such as those in early JWST observations. In conclusion, we find that the double-break sources in CANUCS are not sufficiently massive or numerous to warrant questioning the standard Λ\LambdaCDM paradigm.Comment: V2: correction of display problem of Fig.1 in Chrome browser.     A mode-of-action ontology model for safety evaluation of chemicals: outcome of a series of workshops on repeated dose toxicity

    Repeated dose toxicity evaluation aims at assessing the occurrence of adverse effects following chronic or repeated exposure to chemicals. Non-animal approaches have gained importance in the last decades because of ethical considerations as well as due to scientific reasons calling for more human-based strategies. A critical aspect of this challenge is linked to the capacity to cover a comprehensive set of interdependent mechanisms of action, link them to adverse effects and interpret their probability to be triggered in the light of the exposure at the (sub)cellular level. Inherent to its structured nature, an ontology addressing repeated dose toxicity could be a scientific and transparent way to achieve this goal. Additionally, repeated dose toxicity evaluation through the use of a harmonized ontology should be performed in a reproducible and consistent manner, while mimicking as accurately as possible human physiology and adaptivity. In this paper, the outcome of a series of workshops organized by Cosmetics Europe on this topic is reported. As such, this manuscript shows how experts set critical elements and ways of establishing a mode-of-action ontology model as a support to risk assessors aiming to perform animal-free safety evaluation of chemicals based on repeated dose toxicity data

    The Sparkler: Evolved High-Redshift Globular Clusters Captured by JWST

    Using data from JWST, we analyze the compact sources ("sparkles") located around a remarkable zspec=1.378z_{\rm spec}=1.378 galaxy (the "Sparkler") that is strongly gravitationally lensed by the z=0.39z=0.39 galaxy cluster SMACS J0723.3-7327. Several of these compact sources can be cross-identified in multiple images, making it clear that they are associated with the host galaxy. Combining data from JWST's {\em Near-Infrared Camera} (NIRCam) with archival data from the {\em Hubble Space Telescope} (HST), we perform 0.4-4.4μ\mum photometry on these objects, finding several of them to be very red and consistent with the colors of quenched, old stellar systems. Morphological fits confirm that these red sources are spatially unresolved even in strongly magnified JWST/NIRCam images, while JWST/NIRISS spectra show [OIII]5007 emission in the body of the Sparkler but no indication of star formation in the red compact sparkles. The most natural interpretation of these compact red companions to the Sparkler is that they are evolved globular clusters seen at z=1.378z=1.378. Applying \textsc{Dense Basis} SED-fitting to the sample, we infer formation redshifts of zform711z_{form} \sim 7-11 for these globular cluster candidates, corresponding to ages of 3.94.1\sim 3.9-4.1 Gyr at the epoch of observation and a formation time just \sim0.5~Gyr after the Big Bang. If confirmed with additional spectroscopy, these red, compact "sparkles" represent the first evolved globular clusters found at high redshift, could be amongst the earliest observed objects to have quenched their star formation in the Universe, and may open a new window into understanding globular cluster formation.     GeneCAT—novel webtools that combine BLAST and co-expression analyses

    The gene co-expression analysis toolbox (GeneCAT) introduces several novel microarray data analyzing tools. First, the multigene co-expression analysis, combined with co-expressed gene networks, provides a more powerful data mining technique than standard, single-gene co-expression analysis. Second, the high-throughput Map-O-Matic tool matches co-expression pattern of multiple query genes to genes present in user-defined subdatabases, and can therefore be used for gene mapping in forward genetic screens. Third, Rosetta combines co-expression analysis with BLAST and can be used to find ‘true’ gene orthologs in the plant model organisms Arabidopsis thaliana and Hordeum vulgare (Barley).
