    Can they manage in a sustainable way? The role of international destination awards in beach management

    This study aims to reveal the perceptions of local beach destination managers about the level of sustainability in their management practices. This research puts into practice a comparative study assessing two beach destinations: Mugla, located in Turkey, which won the World Travel Award, and Algarve, located in Portugal, which is a member state of the European Union (EU) that has also repeatedly won the same award. In this context, in-depth interviews were held with tourism offices in selected destinations and qualitative data were obtained through semistructured interviews that were carried out between February and April 2020. As a study outcome, two hypotheses were elected and analysed: "the two regions' sustainable managerial practices differ " and "no difference exists between the regions. " Both regions won the same award, but the EU Algarve destination is still winning the award, whereas the non-EU Mugla destination is not. The findings can be shared with beach destination managers to create or increase their sustainability awareness. No studies were found that compared the sustainable management of award-winning EU and non-EU beach destinations.     Tamoxifen and estradiol interact with the flavin mononucleotide site of complex I leading to mitochondrial failure

    This study evaluated the action of tamoxifen and estradiol on the function of isolated liver mitochondria. We observed that although tamoxifen and estradiol per se did not affect mitochondrial complexes II, III, or IV, complex I is affected, this effect being more drastic (except for state 4 of respiration) when mitochondria were coincubated with both drugs. Furthermore, using two respiratory chain inhibitors, rotenone and diphenyliodonium chloride, we identified the flavin mononucleotide site of complex I as the target of tamoxifen and/or estradiol action(s). Tamoxifen (25 microm) per se induced a significant increase in hydrogen peroxide production and state 4 of respiration. Additionally, a significant decrease in respiratory control ratio, transmembrane, and depolarization potentials were observed. Estradiol per se decreased carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP)-stimulated respiration, state 3 of respiration, and respiratory control ratio and increased lag phase of repolarization. With the exception of state 4 of respiration whose increase induced by tamoxifen was reversed by the presence of estradiol, the effects of tamoxifen were highly exacerbated when estradiol was present. We observed that 10 microm tamoxifen in the presence of estradiol affected mitochondria significantly by decreasing FCCP-stimulated respiration, state 3 of respiration, respiratory control ratio, and ADP depolarization and increasing the lag phase of repolarization. All of the deleterious effects induced by 25 microm tamoxifen were highly exacerbated in the presence of estradiol. Furthermore, we observed that the effects of both compounds were independent of estrogen receptors because the pure estrogen antagonist ICI 182,780 did not interfere with tamoxifen and/or estradiol detrimental effects. Altogether, our data provide a mechanistic explanation for the multiple cytotoxic effects of tamoxifen including its capacity to destroy tamoxifen-resistant breast cancer cells in the presence of estradiol.     Construção e validação de um vídeo educativo sobre o banho de imersão do recém-nascido

    Objetivo: Construir e validar um vídeo educativo sobre o banho de imersão do recémnascido.Método: Pesquisa metodológica, de produção tecnológica, desenvolvida nas fases: préprodução, produção e pós-produção no período de dezembro de 2020 a fevereiro de 2022, validado por enfermeiros doutores,profissionais da área da comunicação social, equipe de enfermagem, puérperas e familiares. Utilizou-se o Índice de Validade de Conteúdo para avaliar concordância entre juízes e público-alvo e Alfa de Cronbrach a confiabilidade dos itens.Resultados: O vídeo tem oito minutos e 34 segundos e foi organizado emantes, durante e após o banho. Roteiro/script e storyboard avaliados quanto ao objetivo, conteúdo, relevância, ambiente, linguagem verbal e inclusão de tópicos e vídeo quanto a funcionalidade, usabilidade, eficiência, técnica audiovisual, ambiente e procedimento, todos alcançaram índices iguais ou maiores a 0,80.Conclusão: O vídeo apresenta potencial tecnológico para ser utilizado durante ações de educação emsaúde e práticas de educação permanente.     Exploring the biotechnological value of marine invertebrates: a closer look at the biochemical and antioxidant properties of Sabella spallanzanii and Microcosmus squamiger

    Sabella spallanzanii and Microcosmus squamiger were profiled for proximate composition, minerals, amino acids, fatty acids (FA), carotenoids, radical scavenging activity on the 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical, oxygen radical absorbance capacity (ORAC) and iron and copper chelating properties. Microcosmus squamiger had the highest level of moisture and crude protein, S. spallanzanii was enriched in crude fat and ash. Both species had similar levels of carbohydrates and energy. There was a prevalence of arginine and glycine in S. spallanzanii, and of taurine in M. squamiger. The most abundant minerals in both species were Na, Ca, and K. The methanol extract of S. spallanzanii had metal chelating properties towards copper and iron, while the methanol extract of M. squamiger was able to chelate copper. M. squamiger extracts had similar ORAC values. Fucoxanthinol and fucoxanthin were the major carotenoids in the M. squamiger dichloromethane extract. Saturated FA were more abundant than unsaturated ones in methanol extracts, and unsaturated FA prevailed in the dichloromethane extracts. Palmitic acid was the predominant FA in methanol extracts, whereas eicosapentaenoic (EPA) and dihomo-γ-linolenic acids were the major compounds in dichloromethane extracts. Low n-6/n-3 ratios were obtained. Our results suggests that both species could be explored as sources of bioactive ingredients with multiple applications.info:eu-repo/semantics/publishedVersio

    Expression profiling in ovarian cancer reveals coordinated regulation of BRCA1/2 and homologous recombination genes

This article is an open access article distributed under the terms and conditions of the Creative Commons Attribution (CC BY) license (https://creativecommons.org/licenses/by/4.0/).Predictive biomarkers are crucial in clarifying the best strategy to use poly(ADP-ribose) polymerase inhibitors (PARPi) for the greatest benefit to ovarian cancer patients. PARPi are specifically lethal to cancer cells that cannot repair DNA damage by homologous recombination (HR), and HR deficiency is frequently associated with BRCA1/2 mutations. Genetic tests for BRCA1/2 mutations are currently used in the clinic, but results can be inconclusive due to the high prevalence of rare DNA sequence variants of unknown significance. Most tests also fail to detect epigenetic modifications and mutations located deep within introns that may alter the mRNA. The aim of this study was to investigate whether quantitation of BRCA1/2 mRNAs in ovarian cancer can provide information beyond the DNA tests. Using the nCounter assay from NanoString Technologies, we analyzed RNA isolated from 38 ovarian cancer specimens and 11 normal fallopian tube samples. We found that BRCA1/2 expression was highly variable among tumors. We further observed that tumors with lower levels of BRCA1/2 mRNA showed downregulated expression of 12 additional HR genes. Analysis of 299 ovarian cancer samples from The Cancer Genome Atlas (TCGA) confirmed the coordinated expression of BRCA1/2 and HR genes. To facilitate the routine analysis of BRCA1/2 mRNA in the clinical setting, we developed a targeted droplet digital PCR approach that can be used with FFPE samples.     Betacyanins from Gomphrena globosa L. flowers: Incorporation in cookies as natural colouring agents

    A betacyanin rich extract was obtained from the flowers of Gomphrena globosa L. by ultrasound-assisted extraction and dried either by lyophilization or spray-drying, was tested as a natural colourant in cookies and compared to a commercial colourant. The extracts were characterized in terms of betacyanin content and antioxidant potential. The effects of the colourants incorporation in the cookies were assessed through proximate composition, soluble sugars, fatty acids, color, texture and microbial load, over a shelf life of 30 days. Considering all the assays and analyzing the results through a 2-way analysis of variance, the cookies incorporated with spray-dried colourant showed the most intense pink coloration while cookies incorporated with lyophilized extract lost less color intensity over time.     O presente projeto de extensão visa a integração entre os estudantes do campusSão Bento do Sul e a comunidade em que está inserido, por meio da literatura,tornando os participantes canais de propagação do gosto literário na comunidade,que apresenta algum tipo de vulnerabilidade, mostrando que a leitura literária não sópode como deve deixar de ser algo elitizado. O projeto promove encontrosquinzenais com crianças e adolescentes, entre 10 e 16 anos, sendo trabalhadostextos curtos e reflexivos, muitas vezes, seguidos por uma dinâmica que auxilia nafruição dos textos literários

    Significado da maternidade frente às hospitalizações de filhos com doenças crônicas

    Objective: To describe the meanings of the experience of motherhood in the hospitalization of children with chronic disease. Method: exploratory field research with a qualitative approach, conducted with six mothers of children with chronic disease, in the Pediatric Intensive Care Unit of a teaching hospital. The collection took place from February to May 2017, through semi-structured interviews and we used content analysis in the thematic modality for data treatment. Results: three categories emerged: Ambivalent feelings expressed by mothers; Uncertainty of motherhood regarding hospitalized child with chronic disease; and Change in maternal routine regarding hospitalization of children with chronic disease. Conclusion: the study provided an opportunity to respond to the experience of motherhood of children with chronic illness manifested through feelings, uncertainties and changes in family routine, which allowed us to reflect on interventions that can alleviate the negative implications throughout the hospitalization period of these children.Objetivo: descrever os significados da experiência da maternidade frente às hospitalizações do filho com doença crônica. Método: pesquisa de campo, exploratória, de abordagem qualitativa, realizada com seis mães de crianças com doença crônica, na Unidade de Terapia Intensiva Pediátrica de um hospital de ensino. A coleta ocorreu nos meses de fevereiro a maio de 2017, por meio de entrevista semiestruturada e utilizou-se a análise de conteúdo na modalidade temática para tratamento dos dados. Resultados: emergiram três categorias: Sentimentos Ambivalentes manifestados pelas mães; Incertezas da maternidade diante do filho hospitalizado com doença crônica; e Mudança na rotina materna frente à hospitalização do filho com doença crônica. Conclusão: o estudo proporcionou responder a experiência da maternidade do filho com doença crônica manifestada por meio de sentimentos, incertezas e mudanças na rotina familiar, o que permitiu refletir sobre intervenções que possam amenizar as implicações negativas durante todo o período de internação dessas crianças
