    Responding to Disinformation: Ten Recommendations for Regulatory Action and Forbearance

    This chapter elaborates on challenges and emerging best practices for state regulation of electoral disinformation throughout the electoral cycle. It is based on research for three studies during 2018–2020: into election cybersecurity for the Commonwealth; on the use of artificial intelligence (AI) to regulate disinformation for the European Parliament; and for UNESCO, the United Nations body responsible for education. The research covers more than half the world’s nations, and substantially more than half that population, and in 2019 the two largest democratic elections in history: India’s general election and the European Parliamentary elections. Regulating digital dominance in electoral disinformation presents specific challenges in three very distinctive fields: election law, media law, and mass communications regulation, and targeted online advertising, including data protection law. Implementing best practices against electoral disinformation will require action by EMBs, data protection agencies, communications and media regulators, parliamentary authorities, and ministries of justice and equivalent Neither effective implementation, nor a disinterested assessment of best practice, can be guaranteed. Electoral laws are—like much history—written by the winners, often immediately after their victory. Legal frameworks need to be updated as a response to disinformation challenges discovered during electoral processes, as well as encompassing international best practice.     Tissue resolved, gene structure refined equine transcriptome.

    BackgroundTranscriptome interpretation relies on a good-quality reference transcriptome for accurate quantification of gene expression as well as functional analysis of genetic variants. The current annotation of the horse genome lacks the specificity and sensitivity necessary to assess gene expression especially at the isoform level, and suffers from insufficient annotation of untranslated regions (UTR) usage. We built an annotation pipeline for horse and used it to integrate 1.9 billion reads from multiple RNA-seq data sets into a new refined transcriptome.ResultsThis equine transcriptome integrates eight different tissues from 59 individuals and improves gene structure and isoform resolution, while providing considerable tissue-specific information. We utilized four levels of transcript filtration in our pipeline, aimed at producing several transcriptome versions that are suitable for different downstream analyses. Our most refined transcriptome includes 36,876 genes and 76,125 isoforms, with 6474 candidate transcriptional loci novel to the equine transcriptome.ConclusionsWe have employed a variety of descriptive statistics and figures that demonstrate the quality and content of the transcriptome. The equine transcriptomes that are provided by this pipeline show the best tissue-specific resolution of any equine transcriptome to date and are flexible for several downstream analyses. We encourage the integration of further equine transcriptomes with our annotation pipeline to continue and improve the equine transcriptome

    Using Mobile Phones to Improve Vaccination Uptake in 21 Low- and Middle-Income Countries: Systematic Review

    Background: The benefits of vaccination have been comprehensively proven, however disparities in coverage persist due to poor health system management, limited resources and parental knowledge and attitudes. Evidence suggests that health interventions that engage local parties in communication strategies improve vaccination uptake. As mobile technology is widely used to improve health communication, mobile health (mHealth) interventions might be used to increase coverage. Objective: To conduct a systematic review of the available literature on the use of mHealth to improve vaccination in low and middle income countries with large numbers of unvaccinated children. Methods: In February 2017, MEDLINE, Scopus and Web of Science, and three health organization websites; Communication Initiative Network, TechNet-21, and PATH, were searched to identify mHealth intervention studies on vaccination uptake in 21 countries. Results: Ten peer-reviewed studies and eleven studies from white or grey literature were included. Nine took place in India, three in Pakistan, two each in Malawi and Nigeria, and one each in Bangladesh, Zambia, Zimbabwe and Kenya. Ten peer-reviewed studies and seven white/grey studies demonstrated improved vaccination uptake after interventions, including appointment reminders, mobile phone apps and pre-recorded messages. Conclusions: While the potential for mHealth interventions to improve vaccination coverage seems clear, the evidence for such interventions is not. The dearth of studies in countries facing the greatest barriers to immunization impedes the prospects for evidence-based policy and practice in these settings.The study was funded by the Medical Research Council, British Heart Foundation, and Homerton College, Cambridge

    TWIST1 Is Expressed in Colorectal Carcinomas and Predicts Patient Survival

    TWIST1 is a transcription factor that belongs to the family of basic helix-loop-helix proteins involved in epithelial-to-mesenchymal transition and invasion processes. The TWIST1 protein possesses oncogenic, drug-resistant, angiogenic and invasive properties, and has been related with several human tumors and other pathologies. Colorectal cancer is one of the tumors in which TWIST1 is over-expressed, but its involvement in the clinical outcome of the disease is still unclear. We tested, by RT-PCR, the expression levels of TWIST1 in normal and tumor paired-sample tissues from a series of 151 colorectal cancer patients, in order to investigate its prognostic value as a tumor marker. TWIST1 expression was restricted to tumor tissues (86.1%) and correlated with lymph node metastasis (LNM). Adjusted analysis showed that the expression levels of TWIST1 correlated with overall survival (OS) and disease-free survival (DFS). Importantly, TWIST1 expression levels predicted OS specifically at stages I and II. Moreover, patients with stage II tumors and high TWIST1 levels showed even shorter survival than patients with stage III tumors. These results suggest that TWIST1 expression levels could be a tumor indicator in stage II patients and help select patients at greater risk of poor prognosis who might benefit from adjuvant chemotherapy

    Individuals with unilateral transtibial amputation and lower activity levels walk more quickly when using a hydraulically articulating versus rigidly attached prosthetic ankle-foot device

    Introduction: Prosthetic ankle-foot devices incorporating a hydraulic articulation between the pylon and prosthetic foot have been shown to be beneficial to the gait of more active individuals with unilateral transtibial amputation (UTA). However, the functional benefits of using hydraulic ankle-foot devices to less active individuals with UTA are yet to be determined. The aim of the current study was to investigate the effects on gait performance of using a non-ESR foot with a hydraulic attachment, compared to an identical, rigidly attached foot during overground walking in less active individuals with UTA. Materials and Methods: Kinematic and kinetic data were recorded while five individuals with UTA, deemed K2 activity level by their prescribing physician, performed two-minute walk tests (2MWT) and ten overground gait trials, in two conditions; using a hydraulically articulating ankle foot device (HYD) and using a rigidly attached ankle foot device (RIG). Results: Walking speed during the 2MWT was increased by 6.5% on average, in the HYD (1.07 m/s) condition, compared to the RIG (1.01 m/s) condition (Cohen’s d = 0.4). Participants displayed more symmetrical inter-limb loading (d = 0.8), increased minimum forward centre of pressure velocity (d = 0.8), increased peak shank rotational velocity (d = 1.0) and decreased prosthetic energy efficiency (d = 0.7) when using the HYD compared to RIG device. Conclusions: Individuals with lower activity levels walk faster and therefore further when, using a foot with a hydraulically articulating attachment, in comparison to a rigid attachment. A reduced braking effect in early stance phase, as a result of the action of the hydraulic component present in the articulating attachment, partially explains the improvement in walking performance

    The UK needs an open data portal dedicated to coastal flood and erosion hazard risk and resilience

    In the UK, coastal flooding and erosion are two of the primary climate-related hazards to communities, businesses, and infrastructure. To better address the ramifications of those hazards, now and into the future, the UK needs to transform its scattered, fragmented coastal data resources into a systematic, integrated portal for quality-assured, publicly accessible open data. Such a portal would support analyses of coastal risk and resilience by hosting, in addition to data layers for coastal flooding and erosion, a diverse array of spatial datasets for building footprints, infrastructure networks, land use, population, and various socio-economic measures and indicators derived from survey and census data. The portal would facilitate novel combinations of spatial data layers to yield scientifically, societally, and economically beneficial insights into UK coastal systems

    Persistent T Cell Repertoire Perturbation and T Cell Activation in HIV After Long Term Treatment

    Objective: In people living with HIV (PLHIV), we sought to test the hypothesis that long term anti-retroviral therapy restores the normal T cell repertoire, and investigate the functional relationship of residual repertoire abnormalities to persistent immune system dysregulation. Methods: We conducted a case-control study in PLHIV and HIV-negative volunteers, of circulating T cell receptor repertoires and whole blood transcriptomes by RNA sequencing, complemented by metadata from routinely collected health care records. Results: T cell receptor sequencing revealed persistent abnormalities in the clonal T cell repertoire of PLHIV, characterized by reduced repertoire diversity and oligoclonal T cell expansion correlated with elevated CD8 T cell counts. We found no evidence that these expansions were driven by cytomegalovirus or another common antigen. Increased frequency of long CDR3 sequences and reduced frequency of public sequences among the expanded clones implicated abnormal thymic selection as a contributing factor. These abnormalities in the repertoire correlated with systems level evidence of persistent T cell activation in genome-wide blood transcriptomes. Conclusions: The diversity of T cell receptor repertoires in PLHIV on long term anti-retroviral therapy remains significantly depleted, and skewed by idiosyncratic clones, partly attributable to altered thymic output and associated with T cell mediated chronic immune activation. Further investigation of thymic function and the antigenic drivers of T cell clonal selection in PLHIV are critical to efforts to fully re-establish normal immune function

    Non-L\'evy mobility patterns of Mexican Me'Phaa peasants searching for fuelwood

    We measured mobility patterns that describe walking trajectories of individual Me'Phaa peasants searching and collecting fuelwood in the forests of "La Monta\~na de Guerrero" in Mexico. These one-day excursions typically follow a mixed pattern of nearly-constant steps when individuals displace from their homes towards potential collecting sites and a mixed pattern of steps of different lengths when actually searching for fallen wood in the forest. Displacements in the searching phase seem not to be compatible with L\'evy flights described by power-laws with optimal scaling exponents. These findings however can be interpreted in the light of deterministic searching on heavily degraded landscapes where the interaction of the individuals with their scarce environment produces alternative searching strategies than the expected L\'evy flights. These results have important implications for future management and restoration of degraded forests and the improvement of the ecological services they may provide to their inhabitants.Comment: 15 pages, 4 figures.
