    Near Miscible CO2 Application to Improve Oil Recovery

    Carbon dioxide (CO2) injection for enhanced oil recovery is a proven technology. CO2 injection is normally operated at a pressure above the minimum miscibility pressure (MMP), which is determined by crude oil composition and reservoir conditions. This is the lowest pressure at which the injected CO2 becomes dynamically miscible with the crude oil remaining in the reservoir. However, many reservoirs are located at depths or under geologic conditions such that they must operate at pressures below the MMP. When CO2 is injected at below the MMP, displacement efficiency decreases as a result of the loss of miscibility. CO2 injection is usually not considered as an enhanced oil recovery process in these reservoirs. Near miscible displacement generally refers to the process that occurs at pressures slightly below the MMP, but the actual pressure range has never been clearly defined. The objectives of this study were to investigate the feasibility of near-miscible CO2 application and improve our understanding of the mechanisms of near-miscible CO2 flooding by conducting appropriate experimental work and reservoir simulation. The pressure range of interest was from 0.8 MMP to MMP in our study. The Arbuckle formation of Kansas was used as an example to demonstrate our approach to evaluate CO2 flooding at near-miscible conditions. The suite of laboratory experiments used to evaluate the feasibility of operating at pressures below MMP for Arbuckle reservoirs included phase behavior studies, core flow tests and phase behavior model construction using CMG software package. Phase behavior studies were carried out to characterize the near miscible conditions. Slim tube displacements and swelling/extraction tests were performed to identify the near miscible range and the mass transfer mechanisms which were responsible for the oil recovery within this range. A phase behavior model was constructed and well-tuned to simulate oil properties, CO2/crude oil interactions and slim tube results. Core flow tests were conducted to evaluate the oil recovery efficiency in the near miscible range. Initial laboratory works indicated that miscibility was not achievable, however at least 65% to 80% of the waterflood residual oil for dolomite cores and lesser from 45% to 60 % for sandstone core in the near-miscible region was observed. The principal oil recovery mechanism in the near-miscible range appeared to be extraction/vaporization of hydrocarbon components from crude oil into the CO2 rich vapor phase, coupled with enhanced mobility control due to the reduction of oil viscosity. This suggested that application of carbon dioxide in the field would require injection and recycling of large volumes of carbon dioxide. Further study is needed to determine if such a process is economically feasible. However the prospect of recovering up to 1 billion barrels of oil from Arbuckle reservoirs offers significant economic potential

    In this study, the material characterization of Vietnamese ground coal slag and ground granulated blast furnace slag (GGBFS), such as particle size distribution, chemical composition, bulk density and particle density are shown. The geopolymer specimens were prepared by mixing an 80 m/m% mass of solid materials (ground coal slag and GGBFS in a different ratio) with 20 m/m % of a 10M NaOH alkaline activator. A systematic experimental series was carried out in order to optimize the preparation process. In that series, the heat curing temperature was 60°C for 6 hours, and then selected specimens were heat treated at a high temperature (1000 °C) for 1 hour. After 7 days of ageing, the physical properties of the geopolymer (compressive strength, specimen density) were measured. Also, after 180 days of ageing, the pH values of water in the geopolymer leaching preparation were determined. The results show that the geopolymer can be used for refractory applications due to its good heat resistance properties. However, geopolymers that were heated at 1000 °C had lower compressive strength, specimen density and pH values of water containing the geopolymer than those that were cured at 60 °C.U ovome istraživanju donose se obilježja vijetnamskih granulata ugljene troske i troske željezne rude na osnovi njihove veličine, kemijskoga sastava i gustoće. Uzorci geopolimera pripremljeni su miješanjem 80 % m/m (mase otopljenoga / masa otopine) ugljene i željezne troske (u različitim omjerima) s 20 % m/m 10M NaOH kao alkalnoga aktivatora. Sustavnim serijama eksperimenata optimiziran je postupak pripreme, pri čemu je temperatura iznosila 60 °C tijekom 6 sati, nakon čega su odabrani uzorci zagrijavani 1 sat na 1000 °C. Nakon sljedećih 7 dana izmjerena su fizička svojstva geopolimera (tlačna čvrstoća, gustoća). Zatim je poslije 180 dana određena pH-vrijednost vode korištene u postupku ispiranja geopolimera. Rezultati su pokazali kako geopolimeri mogu biti korišteni za namjene u vatrostalnim uvjetima zahvaljujući toplinskim svojstvima. Također, geopolimeri koji su zagrijavani na 1000 °C imaju niže vrijednosti tlačne čvrstoće, gustoće i pH-vrijednosti vode u njima od onih koji su grijani na 60 °C

    Novel Enzyme Actions for Sulphated Galactofucan Depolymerisation and a New Engineering Strategy for Molecular Stabilisation of Fucoidan Degrading Enzymes

    Fucoidans from brown macroalgae have beneficial biomedical properties but their use as pharma products requires homogenous oligomeric products. In this study, the action of five recombinant microbial fucoidan degrading enzymes were evaluated on fucoidans from brown macroalgae: Sargassum mcclurei, Fucus evanescens, Fucus vesiculosus, Turbinaria ornata, Saccharina cichorioides, and Undaria pinnatifida. The enzymes included three endo-fucoidanases (EC 3.2.1.-GH 107), FcnA2, Fda1, and Fda2, and two unclassified endo-fucoglucuronomannan lyases, FdlA and FdlB. The oligosaccharide product profiles were assessed by carbohydrate-polyacrylamide gel electrophoresis and size exclusion chromatography. The recombinant enzymes FcnA2, Fda1, and Fda2 were unstable but were stabilised by truncation of the C-terminal end (removing up to 40% of the enzyme sequence). All five enzymes catalysed degradation of fucoidans containing α(1→4)-linked l-fucosyls. Fda2 also degraded S. cichorioides and U. pinnatifida fucoidans that have α(1→3)-linked l-fucosyls in their backbone. In the stabilised form, Fda1 also cleaved α(1→3) bonds. For the first time, we also show that several enzymes catalyse degradation of S. mcclurei galactofucan-fucoidan, known to contain α(1→4) and α(1→3) linked l-fucosyls and galactosyl-β(1→3) bonds in the backbone. These data enhance our understanding of fucoidan degrading enzymes and their substrate preferences and may assist development of enzyme-assisted production of defined fuco-oligosaccharides from fucoidan substrates

    Identification of a Dual-Specific T Cell Epitope of the Hemagglutinin Antigen of an H5 Avian Influenza Virus in Chickens

    Avian influenza viruses (AIV) of the H5N1 subtype have caused morbidity and mortality in humans. Although some migratory birds constitute the natural reservoir for this virus, chickens may play a role in transmission of the virus to humans. Despite the importance of avian species in transmission of AIV H5N1 to humans, very little is known about host immune system interactions with this virus in these species. The objective of the present study was to identify putative T cell epitopes of the hemagglutinin (HA) antigen of an H5 AIV in chickens. Using an overlapping peptide library covering the HA protein, we identified a 15-mer peptide, H5246–260, within the HA1 domain which induced activation of T cells in chickens immunized against the HA antigen of an H5 virus. Furthermore, H5246–260 epitope was found to be presented by both major histocompatibility complex (MHC) class I and II molecules, leading to activation of CD4+ and CD8+ T cell subsets, marked by proliferation and expression of interferon (IFN)-γ by both of these cell subsets as well as the expression of granzyme A by CD8+ T cells. This is the first report of a T cell epitope of AIV recognized by chicken T cells. Furthermore, this study extends the previous finding of the existence of dual-specific epitopes in other species to chickens. Taken together, these results elucidate some of the mechanisms of immune response to AIV in chickens and provide a platform for creation of rational vaccines against AIV in this species

    A Multi-Center Randomized Trial to Assess the Efficacy of Gatifloxacin versus Ciprofloxacin for the Treatment of Shigellosis in Vietnamese Children

    The bacterial genus Shigella is the most common cause of dysentery (diarrhea containing blood and/or mucus) and the disease is common in developing countries with limitations in sanitation. Children are most at risk of infection and frequently require hospitalization and antimicrobial therapy. The WHO currently recommends the fluoroquinolone, ciprofloxacin, for the treatment of childhood Shigella infections. In recent years there has been a sharp increase in the number of organisms that exhibit resistance to nalidixic acid (an antimicrobial related to ciprofloxacin), corresponding with reduced susceptibility to ciprofloxacin. We hypothesized that infections with Shigella strains that demonstrate resistance to nalidixic acid may prevent effective treatment with ciprofloxacin. We performed a randomized controlled trial to compare 3 day ciprofloxacin therapy with 3 days of gatifloxacin, a newer generation fluoroquinolone with greater activity than ciprofloxacin. We measured treatment failure and time to the cessation of individual disease symptoms in 249 children with dysentery treated with gatifloxacin and 245 treated with ciprofloxacin. We could identify no significant differences in treatment failure between the two groups or in time to the cessation of individual symptoms. We conclude that, in Vietnam, ciprofloxacin and gatifloxacin are similarly effective for the treatment of acute dysentery

    Genome-wide analysis identifies 12 loci influencing human reproductive behavior.

    The genetic architecture of human reproductive behavior-age at first birth (AFB) and number of children ever born (NEB)-has a strong relationship with fitness, human development, infertility and risk of neuropsychiatric disorders. However, very few genetic loci have been identified, and the underlying mechanisms of AFB and NEB are poorly understood. We report a large genome-wide association study of both sexes including 251,151 individuals for AFB and 343,072 individuals for NEB. We identified 12 independent loci that are significantly associated with AFB and/or NEB in a SNP-based genome-wide association study and 4 additional loci associated in a gene-based effort. These loci harbor genes that are likely to have a role, either directly or by affecting non-local gene expression, in human reproduction and infertility, thereby increasing understanding of these complex traits
