    Tracer studies in the solid state - Diffusion in hydrogen bonded solids

    Ubbelohde has proposed a proton transfer mechanism to explain electrical conductivity studies on a series of crystalline organic acids and it has been suggested that conductivity increases with the decree of co-operative hydrogen bonding in the crystal. Experiments have been carried cat to test this hypothesis by studying the conductivity pad the rate of proton diffusion, using tritium as tracer, in the same single crystals of two organic acids exhibiting different degrees of cooperative hydrogen bonding; benzoic acid which exists as a linear chains of in the solid state and acetic acid which has linear chains of cooperative hydrogen bonds extending through the crystal. The acids were purified by distillation, sublimation and zone refining techniques and single crystals were grown from the melt in a Bridgman own Diffusion was studied by a sectioning technique using tritium labelled benzoic and acetic acids to obtain the hydrogen diffusion rates and carbon-14 labelled benzoic acid used to measure the diffusion coefficient of the bulk molecule. The electrical conductivity of benzoic acid was found to be very low in single crystals being less than 10

    Wikis as collaborative knowledge management tools in socio-environmental modelling studies

    The data requirements of many socio-environmental system (SES) modelling studies have increased substantially in recent years. This has made the already challenging task of data compilation, retrieval, and sharing progressively more difficult. Recognising the current lack of best practice for knowledge management in SES modelling studies, we propose using SES wikis as a means of addressing these challenges. Wikis have attributes that make them well suited to complex knowledge management tasks and their hierarchical, interconnected, algorithmic logic closely fits with the logic needed in SES model design. In this article, we describe how wikis can be used at each stage of the SES modelling cycle, and we discuss our experiences of putting the approach into practice. We conclude that while SES wikis can be time consuming to initially develop, they have the potential to significantly improve the quality, transparency, and efficiency of SES modelling projects

    Is clarithromycin a potential treatment for cachexia in people with lung cancer?: a feasibility study

    Clarithromycin may improve cachexia and survival in non-small cell lung cancer (NSCLC), but adequately controlled data are lacking. This study was undertaken primarily to inform the feasibility and scale of a phase III trial. Eligible consenting patients with stage IV NSCLC and cachexia were to be randomized to receive either clarithromycin 250 mg twice daily or placebo for eight weeks. Aspects of trial feasibility recorded included numbers eligible, approached and recruited, together with adherence and completion of treatment and assessments. Over 6 months, none of 125 patients identified fulfilled the entry criteria. The commonest reasons for ineligibility were the use of an excluded concurrent drug (45, 36%), brain metastases (22, 18%), poor performance status (21, 17%) and current chemotherapy (15, 12%). A phase III trial of clarithromycin using these entry criteria is not feasible in this setting. Other macrolides that have a lower risk of a drug–drug interaction may be more practical to pursue

    Current conservation status of the Blue Swallow Hirundo atrocaerulea Sundevall 1850 in Africa

    The global Blue Swallow Hirundo atrocaerulea was classified as Vulnerable in 2010 on account of its small and rapidly declining population estimated at less than 1 500 pairs. We undertook this study to gain a better understanding of the current status and threats facing this migratory species. Three previously unknown areas that might be part of the species’ non-breeding range were identified in Kenya and northern Tanzania. Within its breeding range we identified three previously unknown areas of potentially suitable habitat, one in Tanzania and two in Malawi, which require further exploration. Population viability assessment predicted that the Blue Swallow population will decline by 8% in 10 years. The overall probability of extinction of the species in the wild is 3%. Minimum viable population size analysis suggests that a goal for the long-term conservation of the Blue Swallow should be to mitigate current threats that are driving declines such that the population increases to a minimum of 3 600 individuals. This should consist of at least 900 individuals in each of the four clusters identified, along with a minimum of 500 individuals in at least one of the meta-populations per cluster. The four clusters are located in (1) the southeasten Democratic Republic of the Congo, (2) highlands of southern Tanzania and northern Malawi, (3) eastern highlands of Zimbabwe and (4) South Africa and Swaziland. The current proportions of the Blue Swallow population in strictly protected and unprotected areas on their breeding grounds are 53% and 47%, respectively, whereas on their non-breeding grounds the corresponding percentages are 25% and 75%, respectively. Our reassessment of the Blue Swallow’s risk of extinction indicates that it continues to qualify as Vulnerable according to the IUCN/SSC criteria C2a(i).http://www.tandfonline.com/loi/tost202016-09-30hb2016Zoology and Entomolog

    Spatially resolved transcriptomics deconvolutes prognostic histological subgroups in patients with colorectal cancer and synchronous liver metastases

    Background: Patients demonstrating strong immune responses to primary colorectal cancer (CRC) have a survival benefit following surgery, while those with predominantly stromal microenvironments do poorly. Biomarkers to identify patients with colorectal cancer liver metastases (CRLM) who have good prognosis following surgery for oligometastatic disease remain elusive. The aim of this study was to determine the practical application of a simple histological assessment of immune cell infiltration and stromal content in predicting outcome following synchronous resection of primary CRC and CRLM, and to interrogate the underlying functional biology that drives disease progression. Methods: Patients undergoing synchronous resection of primary CRC and CRLM underwent detailed histological assessment, panel genomic and bulk transcriptomic assessment, immunohistochemistry (IHC) and GeoMx Spatial Transcriptomics (ST) analysis. Integration with genomic features, pathway enrichment analysis and immune deconvolution were performed. Results: High-immune metastases were associated with improved cancer specific survival (HR, 0.36, P=0.01). Bulk transcriptomic analysis was confounded by stromal content but ST demonstrated that the invasive edge of the metastases of long-term survivors was characterized by adaptive immune cell populations enriched for Type II Interferon signalling (NES=-2.05 P.Adj<0.005) and MHC-Class II Antigen Presentation (NES=-2.09 P.Adj<0.005). In contrast, patients with poor prognosis demonstrated increased abundance of regulatory T-cells and neutrophils with enrichment of Notch (NES=2.2 P.Adj=0.022) and TGF-β (NES=2.2 P.Adj=0.02) signalling pathways at the metastatic tumor centre. Conclusions: Histological assessment stratifies outcome in patients undergoing synchronous resection of CRLM. ST analysis reveals significant intra-tumoral and inter-lesional heterogeneity with underlying transcriptomic programmes identified in driving each phenotype

    Epithelial-to-mesenchymal transition supports ovarian carcinosarcoma tumorigenesis and confers sensitivity to microtubule-targeting with eribulin

    Ovarian carcinosarcoma (OCS) is an aggressive and rare tumour type with limited treatment options. OCS is hypothesised to develop via the combination theory, with a single progenitor resulting in carcinomatous and sarcomatous components, or alternatively via the conversion theory, with the sarcomatous component developing from the carcinomatous component through epithelial-to-mesenchymal transition (EMT). In this study, we analysed DNA variants from isolated carcinoma and sarcoma components to show that OCS from 18 women is monoclonal. RNA sequencing indicated the carcinoma components were more mesenchymal when compared with pure epithelial ovarian carcinomas, supporting the conversion theory and suggesting that EMT is important in the formation of these tumours. Preclinical OCS models were used to test the efficacy of microtubule-targeting drugs, including eribulin, which has previously been shown to reverse EMT characteristics in breast cancers and induce differentiation in sarcomas. Vinorelbine and eribulin more effectively inhibited OCS growth than standard-of-care platinum-based chemotherapy, and treatment with eribulin reduced mesenchymal characteristics and N-MYC expression in OCS patient-derived xenografts (PDX). Eribulin treatment resulted in an accumulation of intracellular cholesterol in OCS cells, which triggered a down-regulation of the mevalonate pathway and prevented further cholesterol biosynthesis. Finally, eribulin increased expression of genes related to immune activation and increased the intratumoral accumulation of CD8+ T cells, supporting exploration of immunotherapy combinations in the clinic. Together, these data indicate EMT plays a key role in OCS tumourigenesis and support the conversion theory for OCS histogenesis. Targeting EMT using eribulin could help improve OCS patient outcomes

    CA-125 levels are predictive of survival in low-grade serous ovarian cancer—a multicenter analysis

    Objective: Studies on low-grade serous ovarian cancer (LGSC) are limited by a low number of cases. The aim of this study was to define the prognostic significance of age, stage, and CA-125 levels on survival in a multi-institutional cohort of women with pathologically confirmed LGSC. Methods: Women with LGSC were identified from the collaborative Ovarian Cancer Association Consortium (OCAC). Cases of newly diagnosed primary LGSC were included if peri-operative CA-125 levels were available. Age at diagnosis, FIGO stage, pre- and post-treatment CA-125 levels, residual disease, adjuvant chemotherapy, disease recurrence, and vital status were collected by the participating institutions. Progression-free (PFS) and overall survival (OS) were calculated. Multivariable (MVA) Cox proportional hazard models were used and hazard ratios (HR) calculated. Results: A total of 176 women with LGSC were included in this study; 82% had stage III/IV disease. The median PFS was 2.3 years and the median OS was 6.4 years. Age at diagnosis was not significantly associated with worse PFS (p = 0.23) or OS (p = 0.3) (HR per year: 0.99; 95%CI, 0.96−1.01 and 0.98; 95%CI 0.95−1.01). FIGO stage III/IV was independently associated with PFS (HR 4.26, 95%CI 1.43−12.73) and OS (HR 1.69, 95%CI 0.56−5.05). Elevated CA-125 (≥35 U/mL) at diagnosis was not significantly associated with worse PFS (p = 0.87) or OS (p = 0.78) in MVA. Elevated CA-125 (≥35 U/mL) after completion of primary treatment was independently associated with worse PFS (HR 2.81, 95%CI 1.36−5.81) and OS (HR 6.62, 95%CI 2.45−17.92). In the MVA, residual disease was independently associated with PFS (0.022), but not OS (0.85). Conclusion: Advanced LGSC was associated with poor long-term prognosis. FIGO stage and abnormal post-treatment CA-125 level are key prognostic factors inversely associated with PFS and OS. Highlights: 1.
