    Molecular basis of titin exon exclusion by RBM20 and the novel titin splice regulator PTB4

    RNA-binding motif protein 20 (RBM20) is a cardiac splice regulator that adapts cardiac filling via its diverse substrates-including the sarcomeric protein titin. The molecular basis and regulation of RBM20-dependent exon exclusion are largely unknown. In tissue culture experiments, we show that the combination of RNA recognition motif (RRM) and C-terminus is necessary and sufficient for RBM20 activity, indicating an important function of the ZnF2 domain in splicing repression. Using splice reporter and in vitro binding assays targeting titin exons 241-243, we identified a minimal genomic segment that is necessary for RBM20-mediated splicing repression of the alternative exon. Here, RBM20 binds the cluster containing most RBM20 binding motifs through its RRM domain and represses the upstream and downstream introns. For subsequent exon exclusion, specific regions upstream, downstream and within the alternative exon 242 are required. Regulation of exon exclusion involves PTB4 as a novel titin splice regulator, which counteracts RBM20 repressor activity in HEK293 cells. Together, these mechanistic insights into the regulation and action of RBM20 and PTB4 provide a basis for the future development of RBM20 modulators that adapt titin elasticity in cardiac disease

    Predator and Scavenger Modification of Recent Equid Skeletal Assemblages

    This paper reports on the modification by carnivores of recent equid skeletal remains in the Ross River area, Yukon Territory. The objective of this study is to characterize carnivore modification of skeletal assemblages in terms of bone alteration, carnivore preference for certain bone elements or portions, and patterns of survivorship of elements at recent kill/scavenging sites. The most common types of carnivore alteration observed were tooth furrows, punctures, and curvilinear fracture on smaller elements. Chipping and polish occurred infrequently. Survival of skeletal elements depends on the degree of carcass utilization.     Transformation properties and microstructure of sputter-deposited Ni-Ti shape memory alloy thin films

    The influence of annealing parameters on the martensitic phase transformation in sputter-deposited Ti rich Ni-Ti films is systematically studied by differential scanning calorimetry and by transmission electron microscopy. The annealing temperature range extends from the crystallization temperature of the films up to 900°C. For increasing temperature, multiple phase transformations, transformations via an R-phase or direct martensite/austenite transformations are observed. A similar behavior is found for increasing annealing time. Related changes of the film microstructure, such as the strongly varying distribution of round Ti2Ni precipitates in the grains, are analyzed. Transformation temperatures could be shifted over a wide range by adjusting the film composition from 48 to 54 at.% Ti. The corresponding transformation curves, grain structure as well as nature and amount of precipitates were investigated. No subsequent annealing process is required for films deposited on substrates heated above about 500°C. In this case, the as-deposited films have a very fine-grained and homogeneous microstructur

    New 4-aryl-1,3,2-oxathiazolylium-5-olates: chemical synthesis and photochemical stability of a novel series of S-nitrosothiols

    S-nitrosothiols (RSNOs) remain one of the most popular classes of NO-donating compounds due to their ability to release nitric oxide (NO) under non-enzymatic means whilst producing an inert disulphide byproduct. However, alligning these compounds to the different biological fields of NO research has proved to be problematic due to the inherent instability of such compounds under a variety of conditions including heat, light and the presence of copper ions. 1,3,2-Oxathiazolylium-5-olates (OZOs) represent an interesting subclass of S-nitrosothiols that lock the –SNO moiety into a five membered heterocyclic ring in an attempt to improve the compound’s overall stability. The synthesis of a novel series of halogen-containing OZOs was comprehensively studied resulting in a seven-step route and overall yields ranging between 21 and 37%. The photochemical stability of these compounds was assessed to determine if Snitrosothiols locked within these mesoionic ring systems can offer greater stability and thereby release NO in a more controllable fashion than their non-cyclic counterparts

    Poboljšavanje nepristranosti ECL testova slušanja detekcijom čestica koje diskriminiraju prema spolu

    The first main objective of this study is to examine whether ECL listening tests in German at CEFR level B2 are equally fair to both male and female test takers. The second aim is to explore distinctive patterns that can be associated with gender-related differential item functioning (DIF) and potentially result in gender-biased items. For this purpose, two complementary approaches are used: (1) a multi-faceted Rasch analysis in order to detect items showing DIF in terms of gender, and (2) item characteristics and content analyses in order to identify potential systematic sources of gender-biased items. According to the results of the statistical analysis, only 6.5 per cent of the items show differential item functioning, thus verifying that the ECL German listening tests are generally fair toward the two gender groups. The findings of the study did not show any systematic patterns that can be clearly associated with gender-related DIF or can potentially result in gender-biased items.Prvi je cilj ovoga istraživanja ispitati jesu li ECL testovi slušanja na njemačkom jeziku razine B2 prema ZEROJ-u jednako nepristrani prema muškim i ženskim osobama koje rješavaju test. Drugi je cilj istražiti posebne obrasce koji se mogu povezati s razlikovnošću čestica povezanih sa spolom. U tu svrhu primijenjena su dva komplementarna pristupa: 1) Raschova analiza kako bi se pronašle čestice koje pokazuju funkcioniranje diferenciranih stavki prema spolu 2) analiza karakteristika čestica i sadržaja kako bi se identificirali potencijalni sistematični izvori čestica koje diskriminiraju prema spolu. Prema statističkoj analizi samo 6,5 % čestica pokazuje razlikovno funkcioniranje i time potvrđuje da je ECL slušni test njemačkoga općenito nepristran prema ijednom spolu. Rezultati studije nisu pokazali obrazac koji se prema razlikovnom funkcioniranju čestica može jasno povezati s određenim spolom ili rezultira česticama koje su pristrane prema određenom spolu
