Fibroblast growth factor 2-antagonist activity of a long-pentraxin 3-derived antiangiogenic pentapeptide.

Fibroblast growth factor-2 (FGF2) plays a major role in angiogenesis. The pattern recognition receptor long-pentraxin 3 (PTX3) inhibits the angiogenic activity of FGF2. To identify novel FGF2-antagonistic peptide(s), four acetylated (Ac) synthetic peptides overlapping the FGF2-binding region PTX3-(97-110) were assessed for their FGF2-binding capacity. Among them, the shortest pentapeptide Ac-ARPCA-NH(2) (PTX3-[100-104]) inhibits the interaction of FGF2 with PTX3 immobilized to a BIAcore sensorchip and suppresses FGF2-dependent proliferation in endothelial cells, without affecting the activity of unrelated mitogens. Also, Ac-ARPCA-NH(2) inhibits angiogenesis triggered by FGF2 or by tumorigenic FGF2-overexpressing murine endothelial cells in chick and zebrafish embryos, respectively. Accordingly, the peptide hampers the binding of FGF2 to Chinese Hamster ovary cells overexpressing the tyrosine-kinase FGF receptor-1 (FGFR1) and to recombinant FGFR1 immobilized to a BIAcore sensorchip without affecting heparin interaction. In all the assays the mutated Ac-ARPSA-NH(2) peptide was ineffective. In keeping with the observation that hydrophobic interactions dominate the interface between FGF2 and the FGF-binding domain of the Ig-like loop D2 of FGFR1, amino acid substitutions in Ac-ARPCA-NH(2) and saturation transfer difference-nuclear magnetic resonance analysis of its mode of interaction with FGF2 implicate the hydrophobic methyl groups of the pentapeptide in FGF2 binding. These results will provide the basis for the design of novel PTX3-derived anti-angiogenic FGF2 antagonists

Application of NMR microscopy to the morphological study of the silkworm, Bombyx mori, during its metamorphosis.

Zero (ZQ) and double (DQ) quantum 2D chemical shift selective and spin- echo 3D NMR imaging at microscopy resolution, has been applied to the morphological study of silkworm, Bombyx mori, during its metamorphosis. Attention has been focused on the evolution of the internal structure of the insect during its postembryonal life occurring through the larval, pupal and adult development. A major objective of this work was the characterization of the silk glands, responsible for synthesis and secretion of fibroin and sericin, through the changes of distribution and mobility of water, by imaging the water protons during postembryonal life stages. Moreover, alanine deriving from silk gland proteins was imaged during the last life stage of Bombyx mori

Correlations between isospin dynamics and Intermediate Mass Fragments emission time scales: a probe for the symmetry energy in asymmetric nuclear matter

We show new data from the 64^{64}Ni+124^{124}Sn and 58^{58}Ni+112^{112}Sn reactions studied in direct kinematics with the CHIMERA detector at INFN-LNS and compared with the reverse kinematics reactions at the same incident beam energy (35 A MeV). Analyzing the data with the method of relative velocity correlations, fragments coming from statistical decay of an excited projectile-like (PLF) or target-like (TLF) fragments are discriminated from the ones coming from dynamical emission in the early stages of the reaction. By comparing data of the reverse kinematics experiment with a stochastic mean field (SMF) + GEMINI calculations our results show that observables from neck fragmentation mechanism add valuable constraints on the density dependence of symmetry energy. An indication is found for a moderately stiff symmetry energy potential term of EOS.Comment: Talk given by E. Multifragmentation threshold in ^{93}Nb+{nat}Mg collisions at 30 MeV/nucleon

We analyzed the 93Nb^{93}Nb on natMg^{nat}Mg reaction at 30 MeV/nucleon in the aim of disentangling binary sequential decay and multifragmentation decay close to the energy threshold, i.e. 3\simeq 3 MeV/nucleon. Using the backtracing technique applied to the statistical models GEMINI and SMM we reconstruct simulated charge, mass and excitation energy distributions and compare them to the experimental ones. We show that data are better described by SMM than by GEMINI in agreement with the fact that multifragmentation is responsible for fragment production at excitation energies around 3 MeV/nucleon.Comment: 16 pages, 12 figures, 5 tables Soumis \`a Nuclear Physics

The ASY-EOS experiment at GSI: investigating the symmetry energy at supra-saturation densities

    Get PDF
The elliptic-flow ratio of neutrons with respect to protons in reactions of neutron rich heavy-ions systems at intermediate energies has been proposed as an observable sensitive to the strength of the symmetry term in the nuclear Equation Of State (EOS) at supra-saturation densities. The recent results obtained from the existing FOPI/LAND data for 197^{197}Au+197^{197}Au collisions at 400 MeV/nucleon in comparison with the UrQMD model allowed a first estimate of the symmetry term of the EOS but suffer from a considerable statistical uncertainty. In order to obtain an improved data set for Au+Au collisions and to extend the study to other systems, a new experiment was carried out at the GSI laboratory by the ASY-EOS collaboration in May 2011.Comment: Talk given by P. Projected Quasi-particle Perturbation theory

The BCS and/or HFB theories are extended by treating the effect of four quasi-particle states perturbatively. The approach is tested on the pairing hamiltonian, showing that it combines the advantage of standard perturbation theory valid at low pairing strength and of non-perturbative approaches breaking particle number valid at higher pairing strength. Including the restoration of particle number, further improves the description of pairing correlation. In the presented test, the agreement between the exact solution and the combined perturbative + projection is almost perfect. The proposed method scales friendly when the number of particles increases and provides a simple alternative to other more complicated approaches

Isotope correlations as a probe for freeze-out characterization: central 124Sn+64Ni, 112Sn+58Ni collisions

124Sn+64Ni and 112Sn+58Ni reactions at 35 AMeV incident energy were studied with the forward part of CHIMERA multi-detector. The most central collisions were selected by means of a multidimensional analysis. The characteristics of the source formed in the central collisions, as size, temperature and volume, were inspected. The measured isotopes of light fragments (3 <= Z <=8) were used to examine isotope yield ratios that provide information on the free neutron to proton densities.Comment: 4 pages, Contribution to 8th International Conference on Nucleus-Nucleus Collisions, Moscow 200

Mass and charge identification of fragments detected with the Chimera Silicon-CsI(Tl) telescopes

Mass and charge identification of charged products detected with Silicon-CsI(Tl) telescopes of the Chimera apparatus is presented. An identification function, based on the Bethe-Bloch formula, is used to fit empirical correlation between Delta E and E ADC readings, in order to determine, event by event, the atomic and mass numbers of the detected charged reaction products prior to energy calibration.Comment: 24 pages, 7 .jpg figures, submitted to Nucl.Instr.
