    Surface Crystallization in a Liquid AuSi Alloy

    X-ray measurements reveal a crystalline monolayer at the surface of the eutectic liquid Au_{82}Si_{18}, at temperatures above the alloy's melting point. Surface-induced atomic layering, the hallmark of liquid metals, is also found below the crystalline monolayer. The layering depth, however, is threefold greater than that of all liquid metals studied to date. The crystallinity of the surface monolayer is notable, considering that AuSi does not form stable bulk crystalline phases at any concentration and temperature and that no crystalline surface phase has been detected thus far in any pure liquid metal or nondilute alloy.     Qualité microbiologique du lait cru ovin collecte dans la steppe centrale de l’Algérie

    L’objectif de l’étude consiste à évaluer la qualité microbiologique du lait cru ovin collecté localement, en milieu steppique Algérien dans la région de Djelfa situé à 300 km au sud d’Alger. Au total 51 échantillons de lait ont été prélevés pour des fins analytiques. La contamination moyenne est de 2,3.107 cfu/mL pour la flore totale contre 1,1.105 pour les coliformes totaux et 1,5.104 pour les coliformes fécaux. Pour la flore fongique, les laits sont plus contaminés en levures qu’en moisissures (2,4.105 vs 3,4.103 germes/mL respectivement). Les Streptocoques fécaux sont détectés chez 43,14% des échantillons par contre E.coli n’est décelée que chez 17,65% des laits. Par ailleurs, 28% des prélèvements sont positifs vis-à-vis de Clostridium contre 9,8% pour Staphylococcus aureus. En revanche, tous les laits sont exempts de Salmonella. Pour ce qui est de la recherche des anticorps brucelliques, 13,73% des laits analysés sont positifs au Ring-test. A total of 51 samples of bulk milk were examined. The results obtained show the unsatisfactory quality of milk analyzed.     An NF-κB-microRNA regulatory network tunes macrophage inflammatory responses

    The innate inflammatory response must be tightly regulated to ensure effective immune protection. NF-κB is a key mediator of the inflammatory response, and its dysregulation has been associated with immune-related malignancies. Here, we describe a miRNA-based regulatory network that enables precise NF-κB activity in mouse macrophages. Elevated miR-155 expression potentiates NF-κB activity in miR-146a-deficient mice, leading to both an overactive acute inflammatory response and chronic inflammation. Enforced miR-155 expression overrides miR-146a-mediated repression of NF-κB activation, thus emphasizing the dominant function of miR-155 in promoting inflammation. Moreover, miR-155-deficient macrophages exhibit a suboptimal inflammatory response when exposed to low levels of inflammatory stimuli. Importantly, we demonstrate a temporal asymmetry between miR-155 and miR-146a expression during macrophage activation, which creates a combined positive and negative feedback network controlling NF-κB activity.     An elitist quantum-inspired evolutionary algorithm for the flexible job-shop scheduling problem

    The flexible job shop scheduling problem (FJSP) is vital to manufacturers especially in today’s constantly changing environment. It is a strongly NP-hard problem and therefore metaheuristics or heuristics are usually pursued to solve it. Most of the existing metaheuristics and heuristics, however, have low efficiency in convergence speed. To overcome this drawback, this paper develops an elitist quantum-inspired evolutionary algorithm. The algorithm aims to minimise the maximum completion time (makespan). It performs a global search with the quantum-inspired evolutionary algorithm and a local search with a method that is inspired by the motion mechanism of the electrons around an atomic nucleus. Three novel algorithms are proposed and their effect on the whole search is discussed. The elitist strategy is adopted to prevent the optimal solution from being destroyed during the evolutionary process. The results show that the proposed algorithm outperforms the best-known algorithms for FJSPs on most of the FJSP benchmarks

    Four Distances between Pairs of Amino Acids Provide a Precise Description of their Interaction

    The three-dimensional structures of proteins are stabilized by the interactions between amino acid residues. Here we report a method where four distances are calculated between any two side chains to provide an exact spatial definition of their bonds. The data were binned into a four-dimensional grid and compared to a random model, from which the preference for specific four-distances was calculated. A clear relation between the quality of the experimental data and the tightness of the distance distribution was observed, with crystal structure data providing far tighter distance distributions than NMR data. Since the four-distance data have higher information content than classical bond descriptions, we were able to identify many unique inter-residue features not found previously in proteins. For example, we found that the side chains of Arg, Glu, Val and Leu are not symmetrical in respect to the interactions of their head groups. The described method may be developed into a function, which computationally models accurately protein structures

    Transcription forms and remodels supercoiling domains unfolding large-scale chromatin structures

    DNA supercoiling is an inherent consequence of twisting DNA and is critical for regulating gene expression and DNA replication. However, DNA supercoiling at a genomic scale in human cells is uncharacterized. To map supercoiling we used biotinylated-trimethylpsoralen as a DNA structure probe to show the genome is organized into supercoiling domains. Domains are formed and remodeled by RNA polymerase and topoisomerase activities and are flanked by GC-AT boundaries and CTCF binding sites. Under-wound domains are transcriptionally active, enriched in topoisomerase I, “open” chromatin fibers and DNaseI sites, but are depleted of topoisomerase II. Furthermore DNA supercoiling impacts on additional levels of chromatin compaction as under-wound domains are cytologically decondensed, topologically constrained, and decompacted by transcription of short RNAs. We suggest that supercoiling domains create a topological environment that facilitates gene activation providing an evolutionary purpose for clustering genes along chromosomes

    Glutamate regulation of calcium and IP3 oscillating and pulsating dynamics in astrocytes

    Recent years have witnessed an increasing interest in neuron-glia communication. This interest stems from the realization that glia participates in cognitive functions and information processing and is involved in many brain disorders and neurodegenerative diseases. An important process in neuron-glia communications is astrocyte encoding of synaptic information transfer: the modulation of intracellular calcium dynamics in astrocytes in response to synaptic activity. Here, we derive and investigate a concise mathematical model for glutamate-induced astrocytic intracellular Ca2+ dynamics that captures the essential biochemical features of the regulatory pathway of inositol 1,4,5-trisphosphate (IP3). Starting from the well-known two-state Li-Rinzel model for calcium-induced-calcium release, we incorporate the regulation of the IP3 production and phosphorylation. Doing so we extended it to a three-state model (referred as the G-ChI model), that could account for Ca2+ oscillations triggered by endogenous IP3 metabolism as well as by IP3 production by external glutamate signals. Compared to previous similar models, our three-state models include a more realistic description of the IP3 production and degradation pathways, lumping together their essential nonlinearities within a concise formulation. Using bifurcation analysis and time simulations, we demonstrate the existence of new putative dynamical features. The cross-couplings between IP3 and Ca2+ pathways endows the system with self-consistent oscillator properties and favor mixed frequency-amplitude encoding modes over pure amplitude modulation ones. These and additional results of our model are in general agreement with available experimental data and may have important implications on the role of astrocytes in the synaptic transfer of information.Comment: 42 pages, 16 figures, 1 table.     Nonlinear gap junctions enable long-distance propagation of pulsating calcium waves in astrocyte networks

    A new paradigm has recently emerged in brain science whereby communications between glial cells and neuron-glia interactions should be considered together with neurons and their networks to understand higher brain functions. In particular, astrocytes, the main type of glial cells in the cortex, have been shown to communicate with neurons and with each other. They are thought to form a gap-junction-coupled syncytium supporting cell-cell communication via propagating Ca2+ waves. An identified mode of propagation is based on cytoplasm-to-cytoplasm transport of inositol trisphosphate (IP3) through gap junctions that locally trigger Ca2+ pulses via IP3-dependent Ca2+-induced Ca2+ release. It is, however, currently unknown whether this intracellular route is able to support the propagation of long-distance regenerative Ca2+ waves or is restricted to short-distance signaling. Furthermore, the influence of the intracellular signaling dynamics on intercellular propagation remains to be understood. In this work, we propose a model of the gap-junctional route for intercellular Ca2+ wave propagation in astrocytes showing that: (1) long-distance regenerative signaling requires nonlinear coupling in the gap junctions, and (2) even with nonlinear gap junctions, long-distance regenerative signaling is favored when the internal Ca2+ dynamics implements frequency modulation-encoding oscillations with pulsating dynamics, while amplitude modulation-encoding dynamics tends to restrict the propagation range. As a result, spatially heterogeneous molecular properties and/or weak couplings are shown to give rise to rich spatiotemporal dynamics that support complex propagation behaviors. These results shed new light on the mechanisms implicated in the propagation of Ca2+ waves across astrocytes and precise the conditions under which glial cells may participate in information processing in the brain.Comment: Article: 30 pages, 7 figures.
