COMPARISON OF DRUG EFFECTS ON FIXEDâRATIO PERFORMANCE AND CHAIN PERFORMANCE MAINTAINED UNDER A SECONDâORDER FIXEDâRATIO SCHEDULE
In one component of a multiple schedule, pigeons were required to complete the same fourâresponse chain each session by responding sequentially on three identically lighted keys in the presence of four successively presented colors (chain performance). Food presentation occurred after five completions of the chain (i.e., after 20 correct responses). Errors, such as responding on the center or right key when the left was designated correct, produced a brief timeout but did not reset the chain. In the other component, responding on a single key (lighted white) was maintained by food presentation under a fixedâratio 20 schedule. In general, phencyclidine and dâamphetamine produced doseâdependent decreases in the overall response rates in both components. With pentobarbital, overall rate in each component generally increased at intermediate doses and decreased at higher doses. All three drugs produced doseâdependent disruptive effects on chainâperformance accuracy. Phencyclidine and pentobarbital increased percent errors at doses that had little or no rateâdecreasing effects, whereas dâamphetamine generally increased percent errors only at doses that substantially decreased overall rate. At high doses, all three drugs produced greater disruption of chain performance than of fixedratio performance, as indicated by a slower return to control responding, although the effects of dâamphetamine were less selective than those of phencyclidine or pentobarbital. 1985 Society for the Experimental Analysis of Behavio
Epithelial progenitor cells from the human Fallopian tube
The human Fallopian tube (hFT) provides an ideal environment for oocyte fertilization and subsequent development of the resulting blastocyst. Since the inner epithelial mucosa is a dynamic tissue that underlies a constant turnover of cells, a long-term regeneration capacity is required.
Adult stem cells and their pluripotent progeny, the transit-amplifying cells, have been identified in various tissues. These cells have been found to be responsible for adult tissue repair and homeostasis. So far, the self-renewal mechanisms of the tubal epithelium are poorly understood.
In this study, we were able to detect undifferentiated epithelial progenitor cells in the tubal mucosa and found evidence for the presence of adult stem cells existing along with common terminally differentiated cells. Further, we established a novel three-dimensional ex vivo model of epithelial cells derived from the hFT, the âorganoidsâ. By immunofluorescence staining, polarized epithelial cells expressing pluripotency markers were identified within the organoids. These cells are assumed to reside in the tubal mucosa as progenitor cells. Furthermore, we performed in situ hybridization for Lgr5, the most reliable marker for adult stem cells. Our preliminary results indicate the existence of Lgr5 positive cells in the organoids. To ultimately verify the presence of Lgr5 positive stem cells in the tube, the expression has to be demonstrated at protein level using our generated monoclonal antibodies.
To date, there was nothing known about progenitor cells in the hFT. Our results indicate that the mucosal epithelium of the hFT contains pluripotent progenitor cells and potential Lgr5 positive adult stem cells, providing a possible mechanism for tissue homeostasis. In addition, a 3D ex vivo model mimicking in vivo conditions is important to facilitate the study of bacterial infections. Of particular interest is Chlamydia trachomatis, which leads to tubal scarring and infertility
Design characteristics, primary stability and risk of fracture of orthodontic mini-implants: pilot scan electron microscope and mechanical studies
Objectives: Orthodontic mini-implants (OMIs) are increasingly used in orthodontics but can fail for various reasons.
This study investigates the effects of OMI design characteristics on the mechanical properties in artificial bone.
Material and Methods: Twelve self-drilling OMIs (2 small, 6 medium, 4 large) from 8 manufacturers were tested
for their primary stability in simulated medium-high cancellous bone and the risk to fracture in high-density
methacrylate blocks. For the assessments of the maximum insertion torque (IT) and torsional fracture (TF) 5 of
each OMI were used and for the pull-out strength (POS) 10. The OMIs were inserted with a torque screwdriver
(12 sec/360°) until the bottom at 8 mm depth was reached. OMI designs were analyzed with a scan electron mi
-
croscope (SEM).
Results: SEM images revealed a great variation in product refinement. In the whole sample, a cylindrical OMI
shape was associated with higher POS (p<0.001) but lower IT (p=0.002) values. The outer and inner OMI di
-
ameters were design characteristics well correlated with POS, IT and TF values (ranging from 0.601 to 0.961).
Greater thread depth was related to greater POS values (r= 0.628), although OMIs with similar POS values may
have different IT values. Thread depth and pitch had some impact on POS. TF depended mainly on the OMI inner
(r= 0.961) and outer diameters (r=0.892). A thread depth to outer diameter ratio close to 40% increased TF risk.
Conclusions: Although at the same insertion depth the OMI outer and inner diameters are the most important fac
-
tors for primary stability, other OMI design characteristics (cylindrical vs. conical, thread design) may significantly
affect primary stability and torsional fracture. This needs to be considered when selecting the appropriate OMI for
the desired orthodontic procedures
Neurosteroid Binding Sites on the GABAA Receptor Complex as Novel Targets for Therapeutics to Reduce Alcohol Abuse and Dependence
Despite the prevalence of alcohol abuse and dependence in the US and Europe, there are only five approved pharmacotherapies for alcohol dependence. Moreover, these pharmacotherapeutic options have limited clinical utility. The purpose of this paper is to present pertinent literature suggesting that both alcohol and the neurosteroids interact at the GABAA receptor complex and that the neurosteroid sites on this receptor complex could serve as new targets for the development of novel therapeutics for alcohol abuse. This paper will also present data collected by our laboratory showing that one neurosteroid in particular, dehydroepiandrosterone (DHEA), decreases ethanol intake in rats under a variety of conditions. In the process, we will also mention relevant studies from the literature suggesting that both particular subtypes and subunits of the GABAA receptor play an important role in mediating the interaction of neurosteroids and ethanol
Migratory Sleeplessness in the White-Crowned Sparrow (Zonotrichia leucophrys gambelii)
Twice a year, normally diurnal songbirds engage in long-distance nocturnal migrations between their wintering and breeding grounds. If and how songbirds sleep during these periods of increased activity has remained a mystery. We used a combination of electrophysiological recording and neurobehavioral testing to characterize seasonal changes in sleep and cognition in captive white-crowned sparrows (Zonotrichia leucophrys gambelii) across nonmigratory and migratory seasons. Compared to sparrows in a nonmigratory state, migratory sparrows spent approximately two-thirds less time sleeping. Despite reducing sleep during migration, accuracy and responding on a repeated-acquisition task remained at a high level in sparrows in a migratory state. This resistance to sleep loss during the prolonged migratory season is in direct contrast to the decline in accuracy and responding observed following as little as one night of experimenter-induced sleep restriction in the same birds during the nonmigratory season. Our results suggest that despite being adversely affected by sleep loss during the nonmigratory season, songbirds exhibit an unprecedented capacity to reduce sleep during migration for long periods of time without associated deficits in cognitive function. Understanding the mechanisms that mediate migratory sleeplessness may provide insights into the etiology of changes in sleep and behavior in seasonal mood disorders, as well as into the functions of sleep itself
