    Analysis of Carbohydrate Storage Granules in the Diazotrophic Cyanobacterium Cyanothece sp. PCC 7822.

    The unicellular diazotrophic cyanobacteria of the genus Cyanothece demonstrate oscillations in nitrogenase activity and H2 production when grown under 12 h light–12 h dark cycles. We established that Cyanothece sp. PCC 7822 allows for the construction of knock-out mutants and our objective was to improve the growth characteristics of this strain and to identify the nature of the intracellular storage granules. We report the physiological and morphological effects of reduction in nitrate and phosphate concentrations in BG-11 media on this strain. We developed a series of BG-11-derived growth media and monitored batch culture growth, nitrogenase activity and nitrogenase-mediated hydrogen production, culture synchronicity, and intracellular storage content. Reduction in NaNO3 and K2HPO4 concentrations from 17.6 and 0.23 to 4.41 and 0.06 mM, respectively, improved growth characteristics such as cell size and uniformity, and enhanced the rate of cell division. Cells grown in this low NP BG-11 were less complex, a parameter that related to the composition of the intracellular storage granules. Cells grown in low NP BG-11 had less polyphosphate, fewer polyhydroxybutyrate granules and many smaller granules became evident. Biochemical analysis and transmission electron microscopy using the histocytochemical PATO technique demonstrated that these small granules contained glycogen. The glycogen levels and the number of granules per cell correlated nicely with a 2.3 to 3.3-fold change from the minimum at L0 to the maximum at D0. The differences in granule morphology and enzymes between Cyanothece ATCC 51142 and CyanothecePCC 7822 provide insights into the formation of large starch-like granules in some cyanobacteria

    Cilia Have a Significant Role in Regulating Cell Size in Response to Fluid Flow Induced Shear Stress in a Flow Chamber

    Cilia are hair-like protrusions on the apical surface of cells. Their function is to relay mechanical signals like shear stress from extracellular into intracellular environment and thereby maintain cellular homeostasis. Ciliary dysfunctions include polycystic kidney disease and new therapeutic interventions based on ciliary function are under investigation. The current study evaluates the use of a custom designed fluid flow chamber’s ability to study the role of cilia in regulating cell size in response to shear stress. A fluid flow chamber that continually maintains laminar flow at different flow rates and temperature was designed. Endothelial wild type cells (ETWT) that have cilia and polycystic kidney disease cells (PKD) that lost their ciliary function are grown on different glass slides. Cells on each glass slide are then exposed to continuous flow of phosphate-buffered saline at 37oC in the flow chamber. The optimal flow rate and duration of flow were first determined by measuring the total protein concentration before and after exposing the cells. Cell radius and area before and after exposing them to flow are measured using the NIS Software available on the microscope. The results from protein concentrations (n=12) indicate that cells are still attached at normal physiological flow rate 467 mL/min (2.8 ”g/”L) and did not significantly differ from 60 mL/min (4.08 ”g/”L) or 600 mL/min (2.73 ”g/”L). The results for duration of fluid flow (n=22) show that 60 minutes (0.09 + 0.01 ”g/”L) is optimal compared to 120 minutes (0.06 + 0.01 ”g/”L) or 180 minutes (0.10 + 0.02 ”g/”L). Under these optimal conditions, the average area of ETWT cells (n=300) measured from different slides before and after the flow is 4420.81+ 67.40 ”m2 and 4678.17 + 87.15 ”m2 (n=200) respectively. For PKD cells, the average area before and after the flow (n=300) is 5682.46 + 105.48 ”m2 and 4173.74 + 263.97 ”m2 (n=250). These results are in agreement with the published literature on the ability of cilia to maintain cell size in ETWT cells in response to shear stress that is similar to normal blood flow. However, under similar conditions, PKD cells could not maintain their cell size as the mechano-chemical signaling pathway that communicates external signals to prepare appropriate intracellular response is disrupted. These results provide confirmation that the custom designed parallel plate fluid flow chamber is a reliable tool to investigate the specific targets in the mechano-chemical cell signaling pathways

    Composition and Similarity of Bovine Rumen Microbiota across Individual Animals

    The bovine rumen houses a complex microbiota which is responsible for cattle's remarkable ability to convert indigestible plant mass into food products. Despite this ecosystem's enormous significance for humans, the composition and similarity of bacterial communities across different animals and the possible presence of some bacterial taxa in all animals' rumens have yet to be determined. We characterized the rumen bacterial populations of 16 individual lactating cows using tag amplicon pyrosequencing. Our data showed 51% similarity in bacterial taxa across samples when abundance and occurrence were analyzed using the Bray-Curtis metric. By adding taxon phylogeny to the analysis using a weighted UniFrac metric, the similarity increased to 82%. We also counted 32 genera that are shared by all samples, exhibiting high variability in abundance across samples. Taken together, our results suggest a core microbiome in the bovine rumen. Furthermore, although the bacterial taxa may vary considerably between cow rumens, they appear to be phylogenetically related. This suggests that the functional requirement imposed by the rumen ecological niche selects taxa that potentially share similar genetic features

    Root tip-dependent, active riboflavin secretion by Hyoscyamus albus hairy roots under iron deficiency

    Hyoscyamus albus hairy roots with/without an exogenous gene (11 clones) were established by inoculation of Agrobacterium rhizogenes. All clones cultured under iron deficient condition secreted riboflavin from root tips into the culture medium and the productivity depended on the number and size of root tips among the clones, although the addition of sucrose was essential for riboflavin production. A decline of pH was observed before riboflavin production and root development using either a root tip or propagated roots: propagated roots were employed for further work due to their lesser variation. Additions of proton-pump inhibitors, N,N’-dicyclohexylcarbodiimide (DCCD) at 100 and 10 ÎŒM and erythrosine B at 100 ÎŒM, suppressed the pH decline at 100 and 10 ÎŒM accompanied by inhibition of riboflavin secretion and root growth; at 10 ÎŒM of erythrosine B, pH decline occurred with a moderate delay, but both growth and riboflavin efflux were inhibited. Neither inhibition of the pH decline nor riboflavin production was observed at 1 ÎŒM. To examine the necessity of acidification and riboflavin secretion by the roots themselves, artificial pH reduction of culture medium with organic acids and the addition of exogenous riboflavin with/without pH reduction were performed. When hairy roots were cultured in iron-deficient medium acidified with citric acid (pH 4.0) or malic acid (pH 3.7), pH increased rapidly to around 5 overnight, following which riboflavin production and root growth occurred. Addition of riboflavin did not affect riboflavin secretion by the roots, but acidification with citric acid (pH 4.0) helped achiever greater riboflavin production and earlier pH elevation. These results indicate that riboflavin efflux does not directly connected to active pH reduction, and more significantly active riboflavin secretion occurs by internal requirement in H. albus hairy roots under iron deficiency

    Impact of Early Relapse within 24 Months after First-Line Systemic Therapy (POD24) on Outcomes in Patients with Marginal Zone Lymphoma: A US Multisite Study

    Progression of disease within 24 months (POD24) from diagnosis in marginal zone lymphoma (MZL) was shown to portend poor outcomes in prior studies. However, many patients with MZL do not require immediate therapy, and the time from diagnosis-to-treatment interval can be highly variable with no universal criteria to initiate systemic therapy. Hence, we sought to evaluate the prognostic relevance of early relapse or progression within 24 months from systemic therapy initiation in a large US cohort. The primary objective was to evaluate the overall survival (OS) in the two groups. The secondary objective included the evaluation of factors predictive of POD24 and the assessment of cumulative incidence of histologic transformation (HT) in POD24 versus non-POD24 groups. The study included 524 patients with 143 (27%) in POD24 and 381 (73%) in non-POD24 groups. Patients with POD24 had inferior OS compared to those without POD24, regardless of the type of systemic therapy received (rituximab monotherapy or immunochemotherapy) at diagnosis. After adjusting for factors associated with inferior OS in the univariate Cox model, POD24 remained associated with significantly inferior OS (HR = 2.50, 95% CI = 1.53-4.09, p = 0.0003) in multivariable analysis. The presence of monoclonal protein at diagnosis and those who received first-line rituximab monotherapy had higher odds of POD24 on logistic regression analysis. Patients with POD24 had a significantly higher risk for HT compared to those without POD24. POD24 in MZL might be associated with adverse biology and could be used as an additional information point in clinical trials and investigated as a marker for worse prognosis
