    Probing the (H3-H4)(2) histone tetramer structure using pulsed EPR spectroscopy combined with site-directed spin labelling

    The (H3-H4)2 histone tetramer forms the central core of nucleosomes and, as such, plays a prominent role in assembly, disassembly and positioning of nucleosomes. Despite its fundamental role in chromatin, the tetramer has received little structural investigation. Here, through the use of pulsed electron-electron double resonance spectroscopy coupled with site-directed spin labelling, we survey the structure of the tetramer in solution. We find that tetramer is structurally more heterogeneous on its own than when sequestered in the octamer or nucleosome. In particular, while the central region including the H3-H3′ interface retains a structure similar to that observed in nucleosomes, other regions such as the H3 αN helix display increased structural heterogeneity. Flexibility of the H3 αN helix in the free tetramer also illustrates the potential for post-translational modifications to alter the structure of this region and mediate interactions with histone chaperones. The approach described here promises to prove a powerful system for investigating the structure of additional assemblies of histones with other important factors in chromatin assembly/fluidity

    Automated Simulation Updates based on Flight Data

    A statistically-based method for using flight data to update aerodynamic data tables used in flight simulators is explained and demonstrated. A simplified wind-tunnel aerodynamic database for the F/A-18 aircraft is used as a starting point. Flight data from the NASA F-18 High Alpha Research Vehicle (HARV) is then used to update the data tables so that the resulting aerodynamic model characterizes the aerodynamics of the F-18 HARV. Prediction cases are used to show the effectiveness of the automated method, which requires no ad hoc adjustments by the analyst

    Beef Cattle Production and Management Practices and Implications for Educators

    Beef producers need to continually incorporate new information and adopt new technology to effectively manage production costs. Oklahoma State University began a Master Cattleman program with this need in mind. Understanding technology adoption by producers requires identifying current management practices. Data from a survey developed as part of the Master Cattleman program document current practices. Management practices were examined for two groups; producers with smaller herds who are less dependent on the beef enterprise for family income, and producers with larger herds who are more dependent on beef. Results clearly show that size and dependence on the beef enterprise matters when considering a broad spectrum of beef management practices.Livestock Production/Industries,

    On The Orbital Evolution of Jupiter Mass Protoplanet Embedded in A Self-Gravity Disk

    We performed a series of hydro-dynamic simulations to investigate the orbital migration of a Jovian planet embedded in a proto-stellar disk. In order to take into account of the effect of the disk's self gravity, we developed and adopted an \textbf{Antares} code which is based on a 2-D Godunov scheme to obtain the exact Reimann solution for isothermal or polytropic gas, with non-reflecting boundary conditions. Our simulations indicate that in the study of the runaway (type III) migration, it is important to carry out a fully self consistent treatment of the gravitational interaction between the disk and the embedded planet. Through a series of convergence tests, we show that adequate numerical resolution, especially within the planet's Roche lobe, critically determines the outcome of the simulations. We consider a variety of initial conditions and show that isolated, non eccentric protoplanet planets do not undergo type III migration. We attribute the difference between our and previous simulations to the contribution of a self consistent representation of the disk's self gravity. Nevertheless, type III migration cannot be completely suppressed and its onset requires finite amplitude perturbations such as that induced by planet-planet interaction.     The spatial effect of protein deuteration on nitroxide spin-label relaxation:implications for EPR distance measurement

The Authors would also like to acknowledge funding from The MRC – United Kingdom, Grant G1100021.Pulsed electron-electron double resonance (PELDOR) coupled with site-directed spin labeling is a powerful technique for the elucidation of protein or nucleic acid, macromolecular structure and interactions. The intrinsic high sensitivity of electron paramagnetic resonance enables measurement on small quantities of bio-macromolecules, however short relaxation times impose a limit on the sensitivity and size of distances that can be measured using this technique. The persistence of the electron spin-echo, in the PELDOR experiment, is one of the most crucial limitations to distance measurement. At a temperature of around 50 K one of the predominant factors affecting persistence of an echo, and as such, the sensitivity and measurable distance between spin labels, is the electron spin echo dephasing time (Tm). It has become normal practice to use deuterated solvents to extend Tm and recently it has been demonstrated that deuteration of the underlying protein significantly extends Tm. Here we examine the spatial effect of segmental deuteration of the underlying protein, and also explore the concentration and temperature dependence of highly deuterated systems.Publisher PDFPeer reviewe

    Narrowing the filter cavity bandwidth via optomechanical interaction

    We propose using optomechanical interaction to narrow the bandwidth of filter cavities for achieving frequency-dependent squeezing in advanced gravitational-wave detectors, inspired by the idea of optomechanically induced transparency. This not only allows us to achieve narrow bandwidth, comparable to the detection band of few hundred Hz, with tabletop optical cavities, but also to tune the bandwidth over a wide range, which is ideal for optimizing sensitivity for different gravitational-wave sources. The experimental challenge for its implementation is the stringent requirement on low thermal noise, which would need superb mechanical quality factor that is quite difficult to achieve by using currently-available low-loss mechanical oscillators; one possible solution is to use optical dilution of the mechanical damping, which can considerably relax the requirement on the mechanics.Comment: 5 pages + 3 appendix. 4 figures and 2 tables Accepted by Physical Review Letter

    Radar Evaluation of Optical Cloud Constraints to Space Launch Operations

    Weather constraints to launching space vehicles are designed to prevent loss of the vehicle or mission due to weather hazards (See, e.g., Ref 1). Constraints include Lightning Launch Commit Criteria (LLCC) designed to avoid natural and triggered lightning. The LLCC currently in use at most American launch sites including the Eastern Range and Kennedy Space Center require the Launch Weather Officer to determine the height of cloud bases and tops, the location of cloud edges, and cloud transparency. The preferred method of making these determinations is visual observation, but when that isn't possible due to darkness or obscured vision, it is permissible to use radar. This note examines the relationship between visual and radar observations in three ways: A theoretical consideration of the relationship between radar reflectivity and optical transparency. An observational study relating radar reflectivity to cloud edge determined from in-situ measurements of cloud particle concentrations that determine the visible cloud edge. An observational study relating standard radar products to anvil cloud transparency. It is shown that these three approaches yield results consistent with each other and with the radar threshold specified in Reference 2 for LLCC evaluation
