(Autorenreferat
Chronos and CRS: Design of a miniature car-like robot and a software framework for single and multi-agent robotics and control
From both an educational and research point of view, experiments on hardware
are a key aspect of robotics and control. In the last decade, many open-source
hardware and software frameworks for wheeled robots have been presented, mainly
in the form of unicycles and car-like robots, with the goal of making robotics
accessible to a wider audience and to support control systems development.
Unicycles are usually small and inexpensive, and therefore facilitate
experiments in a larger fleet, but they are not suited for high-speed motion.
Car-like robots are more agile, but they are usually larger and more expensive,
thus requiring more resources in terms of space and money. In order to bridge
this gap, we present Chronos, a new car-like 1/28th scale robot with customized
open-source electronics, and CRS, an open-source software framework for control
and robotics. The CRS software framework includes the implementation of various
state-of-the-art algorithms for control, estimation, and multi-agent
coordination. With this work, we aim to provide easier access to hardware and
reduce the engineering time needed to start new educational and research
projects
Microfluidic array cytometer based on refractive optical tweezers for parallel trapping, imaging and sorting of individual cells
Analysis of genetic and functional variability in populations of living cells requires experimental techniques capable of monitoring cellular processes such as cell signaling of many single cells in parallel while offering the possibility to sort interesting cell phenotypes for further investigations. Although flow cytometry is able to sequentially probe and sort thousands of cells per second, dynamic processes cannot be experimentally accessed on single cells due to the sub-second sampling time. Cellular dynamics can be measured by image cytometry of surface-immobilized cells, however, cell sorting is complicated under these conditions due to cell attachment. We here developed a cytometric tool based on refractive multiple optical tweezers combined with microfluidics and optical microscopy. We demonstrate contact-free immobilization of more than 200 yeast cells into a high-density array of optical traps in a microfluidic chip. The cell array could be moved to specific locations of the chip enabling us to expose in a controlled manner the cells to reagents and to analyze the responses of individual cells in a highly parallel format using fluorescence microscopy. We further established a method to sort single cells within the microfluidic device using an additional steerable optical trap. Ratiometric fluorescence imaging of intracellular pH of trapped yeast cells allowed us on the one hand to measure the effect of the trapping laser on the cells' viability and on the other hand to probe the dynamic response of the cells upon glucose sensing
Three-Dimensional Impedance Tomographic Mapping of Metabolically Active Endolumen
Real-time detection of vulnerable atherosclerotic lesions, characterized by a high content of oxidized low-density lipoprotein (oxLDL)-laden macrophages or foam cells, remains an unmet clinical need. While fractional flow reserve (FFR)-guided revascularization in angiographically intermediate stenoses is utilized to assess hemodynamic significance, in vivo detection of oxLDL-rich plaques may provide a new paradigm for treating metabolically unstable lesions. Herein, we have demonstrated endoluminal mapping of lipid-laden lesions using 3-D electrical impedance spectroscopy-derived impedance tomography (EIT) in a pre-clinical swine model. We performed surgical banding of the right carotid arteries of Yucatan mini-pigs, followed by 16 weeks of high-fat diet, to promote the development of lipid-rich lesions. We implemented an intravascular sensor combining an FFR pressure transducer with a 6-point micro-electrode array for electrical impedance spectroscopy (EIS) measurements. 3-D EIT mapping was achieved using an EIS-based reconstruction algorithm. We demonstrated that EIT mapping corresponds to endoluminal histology for oxLDL-laden lesions. We further used computational models to theoretically predict and validate EIS measurements. Thus, our 3-D EIS-derived EIT provides in vivo detection of metabolically active plaques with the goal of guiding optimal intravascular intervention
The study documented here is the result of an empirical-methodical experiment carried out for teaching purposes. It explores the extent to which drawings of a family, asked of study participants, can be used to draw conclusions regarding the underlying personal or cultural conceptions of family and - if this is the case - what can be concluded from the 36 drawings analysed with regard to family conceptions in Germany. The study was conducted in 2013 as part of a seminar at the University of Mainz. It proves that drawings can indeed be a valuable empirical material and a methodical approach to the analysis of family conceptions. However, such an analysis should not be based on a pure image interpretation alone, but ideally be supported by subsequent qualitative interviews related to the drawing. In the light of the analyses, the conception of family in Germany appears to be strongly focused on the middle-class nuclear family, consisting of a married couple of woman and man and about two minor children, including a boy and a girl. Grandparents and pets are also sometimes part of the association. Family members stick closely together and are united in love. Family offers a shelter of privacy from the worries and hardships experienced at work, school or elsewhere, allowing family members to enjoy carefree and happy hours together. Family life takes place at home in oneâs own home or in nature - in any case in peaceful and beautiful places. A variety of family forms can only be found sporadically in the family conceptions of the Germans
Swabian MOSES 2021: An interdisciplinary field campaign for investigating convective storms and their event chains
The Neckar Valley and the Swabian Jura in southwest Germany comprise a hotspot for severe convective storms, causing tens of millions of euros in damage each year. Possible reasons for the high frequency of thunderstorms and the associated event chain across compartments were investigated in detail during the hydro-meteorological field campaign Swabian MOSES carried out
between May and September 2021. Researchers from various disciplines established more than 25 temporary ground-based stations equipped with state-of-the-art in situ and remote sensing observation systems, such as lidars, dual-polarization X- and C-band Doppler weather radars, radiosondes including stratospheric balloons, an aerosol cloud chamber, masts to measure vertical fluxes, autosamplers for water probes in rivers, and networks of disdrometers, soil moisture, and hail sensors. These fixed-site
observations were supplemented by mobile observation systems, such as a research aircraft with scanning Doppler lidar, a cosmic ray neutron sensing rover, and a storm chasing team launching swarmsondes in the vicinity of hailstorms. Seven Intensive Observation Periods (IOPs) were conducted on a total of 21 operating days. An exceptionally high number of convective events,
including both unorganized and organized thunderstorms such as multicells or supercells, occurred during the study period. This paper gives an overview of the Swabian MOSES (Modular Observation Solutions for Earth Systems) field campaign, briefly describes the observation strategy, and presents observational highlights for two IOPs

Inactivation of genes coding for mitochondrial Nd7 and Nd9 complex I subunits in Chlamydomonas reinhardtii. Impact of complex I loss on respiration and energetic metabolism.
In Chlamydomonas, unlike in flowering plants, genes coding for Nd7 (NAD7/49kDa) and Nd9 (NAD9/30kDa) core subunits of mitochondrial respiratory-chain complex I are nucleus-encoded. Both genes possess all the features that facilitate their expression and proper import of the polypeptides in mitochondria. By inactivating their expression by RNA interference or insertional mutagenesis, we show that both subunits are required for complex I assembly and activity. Inactivation of complex I impairs the cell growth rate, reduces the respiratory rate, leads to lower intracellular ROS production and lower expression of ROS scavenging enzymes, and is associated to a diminished capacity to concentrate CO2 without compromising photosynthetic capacity.Peer reviewe
