ILK mediates LPS-induced vascular adhesion receptor expression and subsequent leucocyte trans-endothelial migration†

Aims The inflammatory response to injurious agents is tightly regulated to avoid adverse consequences of inappropriate leucocyte accumulation or failed resolution. Lipopolysaccharide (LPS)-activated endothelium recruits leucocytes to the inflamed tissue through controlled expression of membrane-associated adhesion molecules. LPS responses in macrophages are known to be regulated by integrin-linked kinase (ILK); in this study, we investigated the role of ILK in the regulation of the LPS-elicited inflammatory response in endothelium. Methods and results This study was performed on immortalized mouse endothelial cells (EC) isolated from lung and coronary vasculature. Cells were thoroughly characterized and the role of ILK in the regulation of the LPS response was investigated by suppressing ILK expression using siRNA and shRNA technologies. Phenotypic and functional analyses confirmed that the immortalized cells behaved as true EC. LPS induced the expression of the inflammatory genes E-selectin, intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1). ILK knockdown impaired LPS-mediated endothelial activation by preventing the induction of ICAM-1 and VCAM-1. Blockade of the LPS-induced response inhibited the inflammatory-related processes of firm adhesion and trans-endothelial migration of leucocytes. Conclusion ILK is involved in the expression of cell adhesion molecules by EC activated with the inflammatory stimulus LPS. This reduced expression modulates leucocyte adhesion to the endothelium and the extravasation process. This finding suggests ILK as a potential anti-inflammatory target for the development of vascular-specific treatments for inflammation-related disease

Degradation of 4-Chlorophenol in a Batch Electrochemical Reactor Using BDD Electrodes

The influence of current density (j) (0.25, 0.30, 0.25 and 0.40 A/cm2), initial pH (2.6, 6.5 and 12), stirring speed (As) (400, 500 and 600 rpm), and initial concentration of 4-chlorophenol ([4-CP]0) (300, 500 and 700 mg/L) on degradation of persistent pollutant in a batch electrochemical cell without divisions is presented in this paper. The electrochemical cell was composed of two boron-doped diamond electrodes (BDD). The results of the study showed that best conditions for total degradation of 4-CP were: j of 0.40 A/cm , initial pH of 6.5, As of 500 rpm, and [4-CP]0 of 500 mg/L, after 150 min of reaction time. Removal of total organic carbon (TOC) was 83% at these conditions. The byproducts were identified by UHPLC. This allowed for the proposal of a degradation pathway of 4-CP at the best conditions. Furthermore, these results demonstrate that the electrochemical method employed in this study allows high percentages (96%) of degradation of 4-CP and that the process is applicable to wastewater treatment.CONACYT 26909

Identification of genetic variants associated with Huntington's disease progression: a genome-wide association study

Background Huntington's disease is caused by a CAG repeat expansion in the huntingtin gene, HTT. Age at onset has been used as a quantitative phenotype in genetic analysis looking for Huntington's disease modifiers, but is hard to define and not always available. Therefore, we aimed to generate a novel measure of disease progression and to identify genetic markers associated with this progression measure. Methods We generated a progression score on the basis of principal component analysis of prospectively acquired longitudinal changes in motor, cognitive, and imaging measures in the 218 indivduals in the TRACK-HD cohort of Huntington's disease gene mutation carriers (data collected 2008–11). We generated a parallel progression score using data from 1773 previously genotyped participants from the European Huntington's Disease Network REGISTRY study of Huntington's disease mutation carriers (data collected 2003–13). We did a genome-wide association analyses in terms of progression for 216 TRACK-HD participants and 1773 REGISTRY participants, then a meta-analysis of these results was undertaken. Findings Longitudinal motor, cognitive, and imaging scores were correlated with each other in TRACK-HD participants, justifying use of a single, cross-domain measure of disease progression in both studies. The TRACK-HD and REGISTRY progression measures were correlated with each other (r=0·674), and with age at onset (TRACK-HD, r=0·315; REGISTRY, r=0·234). The meta-analysis of progression in TRACK-HD and REGISTRY gave a genome-wide significant signal (p=1·12 × 10−10) on chromosome 5 spanning three genes: MSH3, DHFR, and MTRNR2L2. The genes in this locus were associated with progression in TRACK-HD (MSH3 p=2·94 × 10−8 DHFR p=8·37 × 10−7 MTRNR2L2 p=2·15 × 10−9) and to a lesser extent in REGISTRY (MSH3 p=9·36 × 10−4 DHFR p=8·45 × 10−4 MTRNR2L2 p=1·20 × 10−3). The lead single nucleotide polymorphism (SNP) in TRACK-HD (rs557874766) was genome-wide significant in the meta-analysis (p=1·58 × 10−8), and encodes an aminoacid change (Pro67Ala) in MSH3. In TRACK-HD, each copy of the minor allele at this SNP was associated with a 0·4 units per year (95% CI 0·16–0·66) reduction in the rate of change of the Unified Huntington's Disease Rating Scale (UHDRS) Total Motor Score, and a reduction of 0·12 units per year (95% CI 0·06–0·18) in the rate of change of UHDRS Total Functional Capacity score. These associations remained significant after adjusting for age of onset. Interpretation The multidomain progression measure in TRACK-HD was associated with a functional variant that was genome-wide significant in our meta-analysis. The association in only 216 participants implies that the progression measure is a sensitive reflection of disease burden, that the effect size at this locus is large, or both. Knockout of Msh3 reduces somatic expansion in Huntington's disease mouse models, suggesting this mechanism as an area for future therapeutic investigation

Ambas supieron conjugar su pasión por la investigación con la docencia, llegando a marcar un hito en la Enseñanza Media de España, al convertirse en las primeras directoras de Institutos de Enseñanza Media. Se trataba de dos mujeres luchadoras que, debido a su inteligencia y a su gran capacidad de trabajo, superaron los planteamientos teóricos de ciertas concepciones conservadoras que relegaban a las mujeres al ámbito doméstico y al único papel de 'buena esposa' y madre.MadridES

Abordaje interdisciplinar en el tratamiento de las heridas en epidermólisis bullosa

Epidermolysis bullosa (EB) is a term that includes a clinically and genetically heterogeneous group of bleb diseases of low prevalence, whose main characteristic is an extreme fragility of the skin and the mucous membranes. Those affected by EB can present problems and complications at very different levels, which must be understood and addressed from a position beyond mere organic pathology. In this article, the EB is approached from an interdisciplinary approach, making known the work of the DEBRA-Spain association for the care of the person affected by EB.La Epidermólisis bullosa (EB) es un término que engloba a un grupo clínico y genéticamente heterogéneo de enfermedades ampollosas de baja prevalencia, cuya principal característica es una extrema fragilidad de la piel y de las membranas mucosas. El objetivo fundamental del proyecto es fomentar el interés por la lectura y la escritura, la comprensión y la expresión verbal y escrita, la representación artística, el conocimiento y la mejora del manejo de las reglas de ortografía y el desarrollo de la convivencia. Todos estos puntos se relacionan con el plan de atención a la diversidad que el centro desarrolla en el currículo anual. Otros objetivos son promover el acercamiento a la lectura desde una perspectiva vivencial y estimulante; fomentar la escritura creativa y la imaginación; incentivar el trabajo en equipo y la cooperación; aumentar y potenciar las cualidades creativas y de expresión; ampliar vocabulario; y fomentar actitudes de implicación, responsabilidad y compromiso con la tarea, tanto individual como en grupo. Se busca, además, fomentar la comunicación en la revisión y renovación de prácticas educativas y metodológicas del profesorado y promover actitudes que incrementen la capacidad de trabajar en equipo y el sentido grupal. Las actividades desarrolladas se relacionan con distintas áreas como la comprensión lectora. Otra área es el comentario oral y escrito de los contenidos de las lecturas a través de la elaboración de cómic, realización de cuestionarios o debates sobre las lecturas. Y otras actividades están relacionadas con el aprendizaje de recursos y mecanismos formales en textos literarios y no literarios, como ejercicios con diccionarios, visualización de películas o dictados. También se realizan lecturas en voz alta y búsqueda de información a través de nuevas tecnologías como Internet o mediante el uso de la Biblioteca. La metodología consiste inicialmente en seleccionar un bloque de 37 libros de los cuales finalmente sólo se quedan 18; en una segunda fase se selecciona un único texto, adaptado a cada una de las etapas, sobre el que se desarrollan todos los ejercicios propuestos para potenciar las competencias lecto-escritoras de los alumnos. Y éstos a través de diferentes actividades, principalmente el comic, la representación musical y teatral transmiten su propia versión de la obra al resto de compañeros. La evaluación es continua y, además de las revisiones semanales, se lleva a cabo una evaluación final y global en la que participan tanto el equipo de trabajo, como los profesores, representantes de los alumnos, el equipo directivo, el departamento de orientación y el AMPA. Proyecto San Juan

El trabajo obtuvo un Premio Tomás García Verdejo a las buenas prácticas educativas en la Comunidad Autónoma de Extremadura para el curso académico 2016/2017. Modalidad BSe presenta un proyecto llevado a cabo en el IES Matías Ramón Martínez de Burguillos del Cerro (Badajoz) en torno a la antigua Iglesia de San Juan Bautista, que actualmente es el centro de interpretación de la Orden del Temple y que fue antigüo templo religioso de varias religiones. El proyecto pretende sensibilizar al alumnado de la importancia del patrimonio cultural y artístico, promover el conocimiento desde distintos puntos de vista de las culturas que coexistían en la Edad Media y su aportación al conocimiento actual, analizar la sociedad actual y la medieval y ver como los valores de igualdad y tolerancia han evolucionado.
