    COCARDE: new view on old mounds – an international network of carbonate mound research

    EGU2012-12550 Carbonate mounds are important contributors of life in different settings, from warm-water to cold-water environments, and throughout geological history. Research on modern cold-water coral carbonate mounds over the last decades made a major contribution to our overall understanding of these particular sedimentary systems. By looking to the modern carbonate mound community with cold-water corals as main framework builders, some fundamental questions could be addressed, until now not yet explored in fossil mound settings. The international network COCARDE (http://www.cocarde.eu) is a platform for exploring new insights in carbonate mound research of recent and ancient mound systems. The aim of the COCARDE network is to bring together scientific communities, studying Recent carbonate mounds in midslope environments in the present ocean and investigating fossil mounds spanning the whole Phanerozoic time, respectively. Scientific challenges in modern and ancient carbonate mound research got well defined during the ESF Magellan Workshop COCARDE in Fribourg, Switzerland (21.–24.01.2009). The Special Volume Cold-water Carbonate Reservoir systems in Deep Environments – COCARDE (Marine Geology, Vol. 282) was the major outcome of this meeting and highlights the diversity of Recent arbonate mound studies. The following first jointWorkshop and Field Seminar held in Oviedo, Spain (16.–20.09.2009) highlighted ongoing research from both Recent and fossil academic groups integrating the message from the industry. The field seminar focused on mounds from the Carboniferous platform of Asturias and Cantabria, already intensively visited by industrial and academic researchers. However, by comparing ancient, mixed carbonate-siliciclastic mound systems of Cantabria with the Recent ones in the Porcupine Seabight, striking similarities in their genesis and processes in mound development asked for an integrated drilling campaign to understand better the 3D internal mound build-up. The Oviedo Workshop and Field Seminar led to the submission of a White Paper on Carbonate Mound Drilling and the initiation of the ESF European Research Network Programme Cold-Water Carbonate Mounds in Shallow and Deep Time – The European Research Network (COCARDE-ERN) launched in June 2011. The second COCARDE Workshop and Field Seminar was held in Rabat, Morocco (24.–30.10.2011) and thematically focussed on carbonate mounds of(f) Morocco. The compact workshop invited students from Moroccan Universities to experience ongoing carbonate mound research in Recent and Ancient environments of Morocco. Two Round Tables discussed innovative approaches in carbonate mound research in Morocco (Recent vs. Ancient - offshore vs. onshore) and reviewed together with oil industry opportunities of international collaboration. The outcome of this workshop will lead into joint research projects, drilling campaigns on- and offshore, and expansion of COCARDE onto the African continent

    Withania somnifera Root Extract Inhibits Mammary Cancer Metastasis and Epithelial to Mesenchymal Transition

    Though clinicians can predict which patients are at risk for developing metastases, traditional therapies often prove ineffective and metastatic disease is the primary cause of cancer patient death; therefore, there is a need to develop anti-metastatic therapies that can be administered over long durations to specifically inhibit the motility of cancer cells. Withania somnifera root extracts (WRE) have anti-proliferative activity and the active component, Withaferin A, inhibits the pro-metastatic protein, vimentin. Vimentin is an intermediate filament protein and is part of the epithelial to mesenchymal transition (EMT) program to promote metastasis. Here, we determined whether WRE standardized to Withaferin A (sWRE) possesses anti-metastatic activity and whether it inhibits cancer motility via inhibition of vimentin and the EMT program. Several formulations of sWRE were created to enrich for Withaferin A and a stock solution of sWRE in EtOH could recover over 90% of the Withaferin A found in the original extract powder. This sWRE formulation inhibited breast cancer cell motility and invasion at concentrations less than 1μM while having negligible cytotoxicity at this dose. sWRE treatment disrupted vimentin morphology in cell lines, confirming its vimentin inhibitory activity. To determine if sWRE inhibited EMT, TGF-β was used to induce EMT in MCF10A human mammary epithelial cells. In this case, sWRE prevented EMT induction and inhibited 3-D spheroid invasion. These studies were taken into a human xenograft and mouse mammary carcinoma model. In both models, sWRE and Withaferin A showed dose-dependent inhibition of tumor growth and metastatic lung nodule formation with minimal systemic toxicity. Taken together, these data support the hypothesis that low concentrations of sWRE inhibit cancer metastasis potentially through EMT inhibition. Moreover, these doses of sWRE have nearly no toxicity in normal mouse organs, suggesting the potential for clinical use of orally administered WRE capsules. © 2013 Yang et al

    New paleoenvironmental insights on the Miocene condensed phosphatic layer of Salento (southern Italy) unlocked by the coral-mollusc fossil archive

    From the Late Oligocene to the Late Miocene, the central Mediterranean area was characterized by the extensive deposition of phosphate-rich sediments. They are usually represented by 10 to 20-cm-thick hardgrounds made of phosphatic and glauconitic sediments containing a rich macrofossil association. This study represents the first thorough investigation of the biotic assemblage of Mediterranean phosphorites aimed at collecting new information on the environmental factors controlling their deposition. The Serravallian/Tortonian phosphatic deposits of the Salento Peninsula (“Aturia level”) have been selected for the abundance of fossil remains and special attention is given to the coral–mollusc association. Two different facies have been recognized: a basal coral rudstone that includes most of the macrofossils, superimposed by a detrital rudstone made of thin layers mainly composed of phosphatic fragments. These two facies are separated by a phosphatic crust several millimeters in thickness. The coral assemblage contains at least 17 azooxanthellate taxa belonging to four families, while the molluscs are represented by a rich gastropod fauna (26 species), associated with bivalves (18 species) and cephalopods (two species). Four distinct depositional phases have been recognized, with the coral rudstone representing the key-facies to reconstruct the onset of the “Aturia level” and the original environment of its fossil content. The composition of the coral–mollusc association has been reliably compared with present-day analog taxa, suggesting the occurrence of a heterogeneous seafloor formed by rocky substrates and accumulations of soft sediment, at around 100–350-m water depth, and under the influence of moderate-to-strong bottom currents rich in nutrients and resuspended organic matter

    A Narrow Quantitative Trait Locus in C. elegans Coordinately Affects Longevity, Thermotolerance, and Resistance to Paraquat

    By linkage mapping of quantitative trait loci, we previously identified at least 11 natural genetic variants that significantly modulate Caenorhabditis elegans life-span (LS), many of which would have eluded discovery by knock-down or mutation screens. A region on chromosome IV between markers stP13 and stP35 had striking effects on longevity in three inter-strain crosses (each P < 10−9). In order to define the limits of that interval, we have now constructed two independent lines by marker-based selection during 20 backcross generations, isolating the stP13–stP35 interval from strain Bergerac-BO in a CL2a background. These congenic lines differed significantly from CL2a in LS, assayed in two environments (each P < 0.001). We then screened for exchange of flanking markers to isolate recombinants that partition this region, because fine-mapping the boundaries for overlapping heteroallelic spans can greatly narrow the implicated interval. Recombinants carrying the CL2a allele at stP35 were consistently long-lived compared to those retaining the Bergerac-BO allele (P < 0.001), and more resistant to temperature elevation and paraquat (each ∼1.7-fold, P < 0.0001), but gained little protection from ultraviolet or peroxide stresses. Two rounds of recombinant screening, followed by fine-mapping of break-points and survival testing, narrowed the interval to 0.18 Mb (13.35–13.53 Mb) containing 26 putative genes and six small-nuclear RNAs – a manageable number of targets for functional assessment

    QUATERNARY BUILD-UPS AND RHODALGAL CARBONATES ALONG THE ADRIATIC AND IONIAN COASTS OF THE ITALIAN PENINSULA: A REVIEW

    In the Mediterranean, build-ups (created by coralline algae, Cladocora caespitosa, deep-water corals, vermetids, polychaetes and bacteria) and rhodolith beds are important hot-spots of biodiversity. Being severely threatened by anthropogenic impact and climate change, they have been included in international directives on environmental protection. This work wants to support the ongoing research on modern bioconstructions by providing further data on the long-term effects of environmental factors on these habitats. Our results are based on the analysis of the existing literature on the outcropping Quaternary successions of the Adriatic and Ionian coasts of peninsular Italy. The existing reports of build-ups and rhodalgal carbonates have been summarized in an homogeneous data-set and then studied to highlight distribution patterns in space and time. The analyses consistently outlined the importance of sedimentation rate in controlling the general distribution of build-ups and rhodalgal carbonates. The majority of the reports is concentrated south of the Gargano, where the sediment-load of the rivers is small. The majority of the reports is related to coralline algae, suggesting that they were the main carbonate producers during the period. C. caespitosa general distribution is mainly controlled by temperature, with most of the occurrences dating back to the warm periods of the late Ionian and of the Tarantian. Large build-ups of Cladocora are restricted to embayments and gulfs well-protected against storm waves. The distribution of the outcrops of deep-water corals is biased by the geological setting. A remarkable uplift is necessary to bring these corals from their original deep-water setting to elevated areas onshore. Consequently, most of the outcrops are in Southern Calabria which is characterized by a strong Quaternary uplift. Chemosynthetic build-ups, intertidal bioconstructions (made by vermetids, polychaetes or coralline algae), as well as stromatolites, are rare in the study area

    This paper investigates the determinant factors of the consumption decision of food away from home for different categories of food in Brazil. Precisely, the variables that can influence this consumption are analyzed, since food away from home has presented a growing share of Brazilian consumers’ diet. The results indicate that the variables of income, of women’s opportunity cost of time and others that capture changes in time constraint in households are important to explain the decision of consumption of food away from home in the Brazilian case.Este trabalho investiga os fatores determinantes da decisão de consumo de alimentação fora do domicílio para diferentes categorias de alimentos no Brasil. Precisamente, as variáveis que podem influenciar esse consumo são analisadas, dado que a alimentação fora do domicílio vem apresentando uma parcela crescente na dieta dos consumidores brasileiros. Os resultados indicam que as variáveis de rendimento, do custo de oportunidade do tempo da mulher e outras que captam mudanças na restrição de tempo nos domicílios são importantes para explicar a decisão de consumo de alimentação fora de casa no caso brasileiro

    Hanging coral gardens of a Tyrrhenian submarine cave from Sicily (Italy)

    An exceptionally well-preserved cave palaeocommunity is described from the Capo Milazzo Peninsula (NE Sicily). The Fulco Cave formed within a layer of breccia including metamorphic and Miocene limestone blocks together with rare clasts of isidid-bearing lithified bathyal sediments. This new breccia type points to a still undescribed deposition event in the early Pleistocene. The fossil association inside the cave is relatively diversified and dominated by the dendrophyllid coral Astroides calycularis whose colonies encrusted the cavity ceiling and grew in an upside-down position, forming spectacular "hanging gardens". The warm climate affinity of Astroides indicates that colonisation took place during an interglacial period, possibly during the Tyrrhenian. The palaeocommunity indicates a semi-dark cave open toward the sea in a shallow water setting. The elongation of Astroides corallites was possibly driven by a low level of water motion and/or competition for space and food. The common constrictions point to slow or no growth phases possibly related to environmental fluctuations, periodically leading to mass mortality events
