    Five decades of terrestrial and freshwater research at Ny-Ålesund, Svalbard

Terrestrial, freshwater, glacial and marine ecosystems are accessible year-round from Ny-Ålesund, providing unique opportunities for interdisciplinary obser-vational and experimental studies along physical, chemical, hydrological and climatic gradients. Here, we synthesize terrestrial and freshwater research at Ny-Ålesund and review current knowledge of biodiversity patterns, species population dynamics and interactions, ecosystem processes, biogeochemical cycles and anthropogenic impacts. There is now strong evidence of past and ongoing biotic changes caused by climate change, including negative effects on populations of many taxa and impacts of rain-on-snow events across multiple trophic levels. While species-level characteristics and responses are well under-stood for macro-organisms, major knowledge gaps exist for microbes, inverte-brates and ecosystem-level processes. In order to fill current knowledge gaps, we recommend (1) maintaining monitoring efforts, while establishing a long-term ecosystem-based monitoring programme; (2) gaining a mechanistic under-standing of environmental change impacts on processes and linkages in food webs; (3) identifying trophic interactions and cascades across ecosystems; and (4) integrating long-term data on microbial, invertebrate and freshwater com-munities, along with measurements of carbon and nutrient fluxes among soils, atmosphere, freshwaters and the marine environment. Para realizar el presente trabajo se recurrió a fuentes bibliográficas y cartográficas diversas y recorridos de campo en la franja costera de Mahahual-Xcalak, y Chetumal-Bacalar utilizando como georeferencia principal el Atlas digital de Costa Maya (2004).     Relationship between Corneal Morphogeometrical Properties and Biomechanical Parameters Derived from Dynamic Bidirectional Air Applanation Measurement Procedure in Keratoconus

    The morphogeometric analysis of the corneal structure has become a clinically relevant diagnostic procedure in keratoconus (KC) as well as the in vivo evaluation of the corneal biomechanical properties. However, the relationship between these two types of metrics is still not well understood. The current study investigated the relationship of corneal morphogeometry and volume with two biomechanical parameters: corneal hysteresis (CH) and corneal resistance factor (CRF), both provided by an Ocular Response Analyzer (Reichert). It included 109 eyes from 109 patients (aged between 18 and 69 years) with a diagnosis of keratoconus (KC) who underwent a complete eye examination including a comprehensive corneal topographic analysis with the Sirius system (CSO). With the topographic information obtained, a morphogeometric and volumetric analysis was performed, defining different variables of clinical use. CH and CRF were found to be correlated with these variables, but this correlation was highly influenced by corneal thickness. This suggests that the mechanical properties of KC cornea contribute only in a partial and limited manner to these biomechanical parameters, being mostly influenced by morphogeometry under normal intraocular pressure levels.     An oleuropein β-glucosidase from olive fruit is involved in determining the phenolic composition of virgin olive oil

    Phenolic composition of virgin olive oil is determined by the enzymatic and/or chemical reactions that take place during olive fruit processing. Of these enzymes, β-glucosidase activity plays a relevant role in the transformation of the phenolic glycosides present in the olive fruit, generating different secoiridoid derivatives. The main goal of the present study was to characterize olive fruit β-glucosidase genes and enzymes responsible for the phenolic composition of virgin olive oil. To achieve that, we have isolated an olive β-glucosidase gene from cultivar Picual (OepGLU), expressed in Nicotiana benthamiana leaves and purified its corresponding recombinant enzyme. Western blot analysis showed that recombinant OepGLU protein is detected by an antibody raised against the purified native olive mesocarp β-glucosidase enzyme, and exhibits a deduced molecular mass of 65.0 kDa. The recombinant OepGLU enzyme showed activity on the major olive phenolic glycosides, with the highest levels with respect to oleuropein, followed by ligstroside and demethyloleuropein. In addition, expression analysis showed that olive GLU transcript level in olive fruit is spatially and temporally regulated in a cultivar-dependent manner. Furthermore, temperature, light and water regime regulate olive GLU gene expression in olive fruit mesocarp. All these data are consistent with the involvement of OepGLU enzyme in the formation of the major phenolic compounds present in virgin olive oil

    End-of-Life Narratives of Patients who Request Medical Assistance in Dying: A Qualitative Study Protocol

Bioethics literature on the topic involve different approaches (analytic, empirical, policy oriented, activist) and various normative perspectives (discourses based on deontological premises as the sanctity of life and human dignity, or on utilitarianism, libertarianism, etc.), some of which are at times irreconcilable. Regarding empirical studies, some voices (e.g., healthcare professionals) have been widely considered but the voice of people who request MAiD has been neglected. Understanding the personal and medical circumstances that lead to MAiD, which can only be achieved by listening to the phenomenological discourse of those involved, is key. This study aims to provide knowledge from the testimonies and experiences of patients who have initiated a MAiD request. We believe this research protocol can increase our understanding of a social and academic controversy that lacks important information to be complete. By doing so, this type of research could inform and improve end-of-life public policies, and particularly the health care of individuals who request a MAiD. We propose a qualitative phenomenological study using semi-structured interviews of people in the process of requesting MAiD, as outlined in the Ley Organica 3/2021, de regulacion de la eutanasia (LORE), the Spanish Law on the Regulation of Euthanasia. The study is conducted nationally using a convenience sampling. The number of interviews is determined sequentially and cumulatively, depending on the richness of the narratives and the saturation of the information that has been collected. For the purposes of analysis, the interviews are transcribed verbatim and pseudonymised afterwards. Data analysis is conducted at the same time as data collection.     A predictive model for fracture in human ribs based on in-vitro acoustic emission data

    Purpose The aim of this paper is to propose a fracture model for human ribs based on acoustic emission (AE) data. The accumulation of microcracking until a macroscopic crack is produced can be monitored by AE. The macrocrack propagation causes the loss of the structural integrity of the rib. Methods The AE technique was used in in vitro bending tests of human ribs. The AE data obtained were used to construct a quantitative model that allows an estimation of the failure stress from the signals detected. The model predicts the ultimate stress with an error of less than 3.5% (even at stresses 15% lower than failure stress), which makes it possible to safely anticipate the failure of the rib. Results The percolation theory was used to model crack propagation. Moreover, a quantitative probability-based model for the expected number of AE signals has been constructed, incorporating some ideas of percolation theory. The model predicts that AE signals associated with micro-failures should exhibit a vertical asymptote when stress increases. The occurrence of this vertical asymptote was attested in our experimental observations. The total number of microfailures detected prior to the failure is urn:x-wiley:00942405:media:mp15082:mp15082-math-0001 and the ultimate stress is urn:x-wiley:00942405:media:mp15082:mp15082-math-0002 MPa. A significant correlation (p < 0.0001) between urn:x-wiley:00942405:media:mp15082:mp15082-math-0003 and the predicted value is found, using only the first N = 30 micro-failures (correlation improves for N higher). Conclusions The measurements and the shape of the curves predicted by the model fit well. In addition, the model parameters seem to explain quantitatively and qualitatively the distribution of the AE signals as the material approaches the macroscopic fracture. Moreover, some of these parameters correlate with anthropometric variables, such as age or Body Mass Index.     Morphogeometric analysis for characterization of keratoconus considering the spatial localization and projection of apex and minimum corneal thickness point

    This work evaluates changes in new morphogeometric indices developed considering the position of anterior and posterior corneal apex and minimum corneal thickness (MCT) point in keratoconus. This prospective comparative study included 440 eyes of 440 patients (age, 7–99 years): control (124 eyes) and keratoconus (KC) groups (316 eyes). Tomographic information (Sirius®, Costruzione Strumenti Oftalmici, Italy) was treated with SolidWorks v2013, creating the following morphogeometric parameters: geometric axis–apex line angle (GA–AP), geometric axis–MCT line angle (GA–MCT, apex line–MCT line angle (AP–MCT), and distances between apex and MCT points on the anterior (anterior AP–MCTd) and posterior corneal surface (posterior AP–MCTd). Statistically significant higher values of GA–AP, GA–MCT, AP–MCT and anterior AP–MCTd were found in the keratoconus group (p ≤ 0.001). Moderate significant correlations of corneal aberrations (r ≥ 0.587, p < 0.001) and corneal thickness parameters (r ≤ −0.414, p < 0.001) with GA–AP and AP–MCT were found. Anterior asphericity was found to be significantly correlated with anterior and posterior AP–MCTd (r ≥ 0.430, p < 0.001). Likewise, GA–AP and AP–MCT showed a good diagnostic ability for the detection of keratoconus, with optimal cutoff values of 9.61° (sensitivity 85.5%, specificity 80.3%) and 18.08° (sensitivity 80.5%, specificity 78.7%), respectively. These new morphogeometric indices allow a clinical characterization of the 3-D structural alteration occurring in keratoconus, with less coincidence in the spatial projection of the apex and MCT points of both corneal surfaces.
