    The extracts of the bark of Hippophae salicifolia D.DON have been found to possess significant inhibitory activity on mouse fibrosarcoma. The tumour tissues showed that there was a degeneration and even necrosis of tumour calls. The alcoholic extract (B) of the bark also possesses an inhibitory activity against Yoshida sarcoma (ascites), as evidenced by the increase in survival period of the experimental animals. The chemistry of the bark is under investigation

    Impact of biotic and abiotic factors on lac production and peoples’ livelihood improvement in India-An overview

    In India, Madhya Pradesh is the third largest lac producing state after Chhattisgarh and Jharkhand, as they contribute 12, 16 and 60%, respectively in total country’s lac production. Lac is produced in ten out of eleven agro-climatic zones of Madhya Pradesh. In total lac production of the state, Baisakhi (Butea monosperma, inoculated in October-November) crop contributes 76% followed by 19% by Katki (Butea monosperma, inoculated in June-July), 3% by Aghani (Schleichera oleosa, inoculated in June-July) and 2% by Jethwi (Schleichera oleosa, inoculated in January-February). Ranchi was leading lac producing district followed by Simdega, Gumla, Khunti of Jharkhand and Seoni district of Madhya Pradesh. Increasing summer temperature up to 45 ?C, the country’s lac production was declined from 20,050 tons in 2003-04 to 16978 tons in 2014-15. In case of biotic factors, Tachardiaephagus tachardiae and Tetrastichus purpureus are the most abundant lac associated parasites and Eublemma amabilis and Pseudohypatopa pulverea are the most destructive key predators of lac insects. By combating these hindrances, lac cultivation generated an employment for 16-160 man days. Lac cultivation produces maximum gross return (Rs. 9,77,600) from 100 Kusum host plants, and the highest Benefit-Cost (6.80) ratio was recorded for Ber-kusumi (Ziziphus mauritiana- Schleichera oleosa) crop in Ranchi, Jharkhand. In this context, the study finds out obstacles in lac production of India and suggests some control measures to improve lac producing peoples’ livelihood. &nbsp

    SUMOylation of nuclear actin

    Actin, a major component of the cytoplasm, is also abundant in the nucleus. Nuclear actin is involved in a variety of nuclear processes including transcription, chromatin remodeling, and intranuclear transport. Nevertheless, the regulation of nuclear actin by posttranslational modifications has not been investigated. We now show that nuclear actin is modified by SUMO2 and SUMO3 and that computational modeling and site-directed mutagenesis identified K68 and K284 as critical sites for SUMOylating actin. We also present a model for the actin–SUMO complex and show that SUMOylation is required for the nuclear localization of actin

    A Mechanistic View of the Role of E3 in Sumoylation

    Sumoylation, the covalent attachment of SUMO (Small Ubiquitin-Like Modifier) to proteins, differs from other Ubl (Ubiquitin-like) pathways. In sumoylation, E2 ligase Ubc9 can function without E3 enzymes, albeit with lower reaction efficiency. Here, we study the mechanism through which E3 ligase RanBP2 triggers target recognition and catalysis by E2 Ubc9. Two mechanisms were proposed for sumoylation. While in both the first step involves Ubc9 conjugation to SUMO, the subsequent sequence of events differs: in the first E2-SUMO forms a complex with the target and E3, followed by SUMO transfer to the target. In the second, Ubc9-SUMO binds to the target and facilitates SUMO transfer without E3. Using dynamic correlations obtained from explicit solvent molecular dynamic simulations we illustrate the key roles played by allostery in both mechanisms. Pre-existence of conformational states explains the experimental observations that sumoylation can occur without E3, even though at a reduced rate. Furthermore, we propose a mechanism for enhancement of sumoylation by E3. Analysis of the conformational ensembles of the complex of E2 conjugated to SUMO illustrates that the E2 enzyme is already largely pre-organized for target binding and catalysis; E3 binding shifts the equilibrium and enhances these pre-existing populations. We further observe that E3 binding regulates allosterically the key residues in E2, Ubc9 Asp100/Lys101 E2, for the target recognition

    Data visualization plays a key role in high-throughput biology. It is an essential tool for data exploration allowing to shed light on data structure and patterns of interest. Visualization is also of paramount importance as a form of communicating data to a broad audience. Here, we provided a short overview of the application of the R software to the visualization of proteomics data.     Targeting of SUMO substrates to a Cdc48-Ufd1-Npl4 segregase and STUbL pathway in fission yeast

    In eukaryotes, the conjugation of proteins to the small ubiquitin-like modifier (SUMO) regulates numerous cellular functions. A proportion of SUMO conjugates are targeted for degradation by SUMO-targeted ubiquitin ligases (STUbLs) and it has been proposed that the ubiquitin-selective chaperone Cdc48/p97-Ufd1-Npl4 facilitates this process. However, the extent to which the two pathways overlap, and how substrates are selected, remains unknown. Here we address these questions in fission yeast through proteome-wide analyses of SUMO modification sites. We identify over a thousand sumoylated lysines in a total of 468 proteins and quantify changes occurring in the SUMO modification status when the STUbL or Ufd1 pathways are compromised by mutations. The data suggest the coordinated processing of several classes of SUMO conjugates, many dynamically associated with centromeres or telomeres. They provide new insights into subnuclear organization and chromosome biology, and, altogether, constitute an extensive resource for the molecular characterization of SUMO function and dynamics
