Charakterisierung der monozytären Adhärenz bei myokardialer Ischämie und Reperfusionsschaden

Der akute Myokardinfarkt ist eine Erkrankung mit hoher Morbidität und Letalität. Ein überlebter Infarkt führt häufig zu Herzinsuffizienz, welche die Lebensqualität der Patienten deutlich eingeschränkt. In der vorliegenden Arbeit wird deshalb untersucht, ob therapeutisch in die Interaktion des Proteins Cyclophilin A (CyPA) mit seinem Rezeptor CD147 eingegriffen werden kann, um die Größe eines Herzinfarkts zu verringern. Dem zugrunde liegt die Hypothese, dass extrazelluläres CyPA über CD147 die Einwanderung von Monozyten in geschädigtes Myokard bewirkt. Dort führen Monozyten zu Zellschäden und einem Gewebeumbau, der die Entstehung einer Herzinsuffizienz begünstigt. In vivo wurde daher an Mäusen durch Ligatur der LAD ein Myokardinfarkt erzeugt und kurz vor der Reperfusion Antikörper gegen CD147 appliziert. In diesem praxisnahen Szenario wurden durch die Inhibition von CD147 sowohl die Infarktgröße als auch die Zahl einwandernder Zellen ins Infarktgebiet deutlich reduziert. In vitro wurde unter Strömungsbedingungen der Einfluss von CyPA auf das Adhäsionsverhalten monozytärer MonoMac6-Zellen an aktiviertem Endothel untersucht. Dabei konnte erstmals gezeigt werden, dass speziell extrazelluläres CyPA die monozytäre Adhäsion fördert. Es wurde nachgewiesen, dass dieser Effekt über den PI3K-Akt-Signalweg vermittelt wird. Die pharmakologische Inhibition von CyPA bzw. CD147 stellt damit einen vielversprechenden Ansatz für die zukünftige Therapie des Myokardinfarkts dar. Hierdurch könnte es gelingen, die Herzfunktion nach einem Infarkt langfristig zu verbessern. Da sowohl CyPA als auch CD147 v.a. bei Krankheitsprozessen exprimiert werden, ist ein geringes Nebenwirkungspotential wahrscheinlich, das durch die Fokussierung auf ausschließlich extrazelluläres CyPA weiter gesenkt werden kann. Das grundlegende Wissen um die Rolle des PI3K-Akt-Signalwegs bei der CyPA-vermittelten Monozytenadhäsion ermöglicht eine weitere Forschung, an deren Ende eine gezielte Beeinflussung intrazellulärer Signalwege in verschiedenen Krankheiten stehen könnte. In der vorliegenden Studie bleiben das höhere Alter der meisten Herzinfarkt-Patienten sowie häufige Komorbiditäten und Begleitmedikation unberücksichtigt. Daher müssen die Ergebnisse dieser Studie unter weiteren Bedingungen evaluiert werden, sodass es möglich wird, die Patienten zu identifizieren, welche von einer zusätzlichen Therapie durch CyPA-CD147-Inhibition profitieren

Accurate rapid averaging of multihue ensembles is due to a limited capacity subsampling mechanism

It is claimed that the extraction of average features from rapidly presented ensembles is holistic, with attention distributed across the whole set. We investigated whether observers’ extraction of mean hue is holistic or could reflect subsampling. Analysis of selections for the mean hue revealed a distribution that peaked at the expected mean hue. However, an ideal observer simulation suggested that a subsampling mechanism incorporating just two items from each ensemble would suffice to reproduce the precision of most observers. The results imply that hue may not be averaged as holistically and efficiently as other attributes

Ensemble coding of color and luminance contrast

Ensemble coding has been demonstrated for many attributes including color, but the metrics on which this coding is based remain uncertain. We examined ensemble percepts for stimulus sets that varied in chromatic contrast between complementary hues, or that varied in luminance contrast between increments and decrements, in both cases focusing on the ensemble percepts for the neutral gray stimulus defining the category boundary. Each ensemble was composed of 16 circles with four contrast levels. Observers saw the display for 0.5 s and then judged whether a target contrast was a member of the set. False alarms were high for intermediate contrasts (within the range of the ensemble) and fell for higher or lower values. However, for ensembles with complementary hues, gray was less likely to be reported as a member, even when it represented the mean chromaticity of the set. When the settings were repeated for luminance contrast, false alarms for gray were higher and fell off more gradually for out-of-range contrasts. This difference implies that opposite luminance polarities represent a more continuous perceptual dimension than opponent-color variations, and that “gray” is a stronger category boundary for chromatic than luminance contrasts. For color, our results suggest that ensemble percepts reflect pooling within rather than between large hue differences, perhaps because the visual system represents hue differences more like qualitatively different categories than like quantitative differences within an underlying color “space.” The differences for luminance and color suggest more generally that ensemble coding for different visual attributes might depend on different processes that in turn depend on the format of the visual representation

Principal-agent problem in technology projects on kickstarter : an exploratory case study

I conducted an exploratory research of the little understood principal-agent interaction in a reward-based crowdfunding environment. A broad-stroke exploratory research like this is unavoidably limited in the extent to which any of the findings can be generalised beyond individual cases. I attempt to mitigate this by complementing a netnographic approach with a sentiment analysis classifier programme that I have developed. This holistic approach allowed me to gain deep insight into the mechanisms that allow the principal-agent problem on Kickstarter to be resolved successfully despite lack of rigid legislative regulation. I find that backers on Kickstarter possess sufficient tools to minimise information asymmetry and thus, the principal-agent problem. I further discover that formations of backers are comprised of two distinct groups – a small vocal and a much larger silent one, with the latter adjusting to the opinions of the former in the short term. I also find that the crowdfunding platform plays a limited, yet important role in resolving the principal-agent tensions. Finally, I find evidence that formations of backers that surround Kickstarter projects are fluid in their structure, exhibiting under different conditions features of both communities and publics.nhhma
