    Scorpaena decemradiata nueva especie (Teleostei: Scorpaenidae) del golfo de Aqaba, mar Rojo del norte, una especie distinta de Scorpaena porcus

    The scorpionfish Scorpaena decemradiata n. sp. is described from off the coast of Israel in the Gulf of Aqaba, northern Red Sea. The new species is similar to S. porcus Linnaeus, 1758, but is characterized by dorsal fin spines XII, soft dorsal fin rays 10 (the last divided at base); pectoral fin rays 16, uppermost branched pectoral fin ray is the second; lacrimal with 2 spines over maxilla that point at nearly right angle from each other, the posterior pointing ventrally and slightly anteriorly; occipital pit well developed; anteriormost mandibular lateral-line pores small, separated; scales ctenoid; 59-62 scale rows in longitudinal series; scales absent on chest and pectoral fin base; and cirri developed over entire head and body, but no cirri on lower jaw. An updated checklist of the species of the genus Scorpaena Linnaeus, 1758 and a key to the species of the eastern Atlantic, Mediterranean Sea and Red Sea are presented.En este trabajo se describe el escorpeniforme Scorpaena decemradiata n. sp. de la costa de Israel, golfo de Aqaba, norte del mar Rojo. La nueva especie es similar a S. porcus Linnaeus, 1758, pero se caracteriza por la presencia de XII espinas y 10 radios blandos (el último dividido en la base) en la aleta dorsal; 16 radios en la aleta pectoral, el radio ramificado más superior es el segundo; lacrimal con 2 espinas sobre el maxilar que apuntan casi en ángulo recto, la posterior apuntando ventralmente y ligeramente en dirección anterior; fosa occipital bien desarrollada; los poros más anteriores de la línea lateral mandibular son pequeños y separados; escamas ctenoides; de 59 a 62 filas de escamas en series longitudinales; escamas ausentes en el torax y la base de la aleta pectoral; y cirros desarrollados sobre toda la cabeza y el cuerpo, pero no en la mandíbula inferior.     Integrated quantized electronics: a semiconductor quantized voltage source

    The Josephson effect in superconductors links a quantized output voltage Vout = f \cdot(h/2e) to the natural constants of the electron's charge e, Planck's constant h, and to an excitation frequency f with important applications in electrical quantum metrology. Also semiconductors are routinely applied in electrical quantum metrology making use of the quantum Hall effect. However, despite their broad range of further applications e.g. in integrated circuits, quantized voltage generation by a semiconductor device has never been obtained. Here we report a semiconductor quantized voltage source generating quantized voltages Vout = f\cdot(h/e). It is based on an integrated quantized circuit of a single electron pump operated at pumping frequency f and a quantum Hall device monolithically integrated in series. The output voltages of several \muV are expected to be scalable by orders of magnitude using present technology. The device might open a new route towards the closure of the quantum metrological triangle. Furthermore it represents a universal electrical quantum reference allowing to generate quantized values of the three most relevant electrical units of voltage, current, and resistance based on fundamental constants using a single device.Comment: 15 pages, 3 figure

    Rapid developments in the field of CAR T cells offer important new opportunities while at the same time increasing numbers of patients pose major challenges. This review is summarizing on the one hand the state of the art in CAR T cell trials with a unique perspective on the role that Europe is playing. On the other hand, an overview of reproducible processing techniques is presented, from manual or semi-automated up to fully automated manufacturing of clinical-grade CAR T cells. Besides regulatory requirements, an outlook is given in the direction of digitally controlled automated manufacturing in order to lower cost and complexity and to address CAR T cell products for a greater number of patients and a variety of malignant diseases

    TGFBR2-dependent alterations of exosomal cargo and functions in DNA mismatch repair-deficient HCT116 colorectal cancer cells

    Background: Colorectal cancers (CRCs) that lack DNA mismatch repair function exhibit the microsatellite unstable (MSI) phenotype and are characterized by the accumulation of frameshift mutations at short repetitive DNA sequences (microsatellites). These tumors recurrently show inactivating frameshift mutations in the tumor suppressor Transforming Growth Factor Beta Receptor Type 2 (TGFBR2) thereby abrogating downstream signaling. How altered TGFBR2 signaling affects exosome-mediated communication between MSI tumor cells and their environment has not been resolved. Here, we report on molecular alterations of exosomes shed by MSI cells and the biological response evoked in recipient cells. Methods: Exosomes were isolated and characterized by electron microscopy, nanoparticle tracking, and western blot analysis. TGFBR2-dependent effects on the cargo and functions of exosomes were studied in a MSI CRC model cell line enabling reconstituted and inducible TGFBR2 expression and signaling. Microsatellite frameshift mutations in exosomal and cellular DNA were examined by PCR-based DNA fragment analysis and exosomal protein profiles were identified by mass spectrometry. Uptake of fluorescent-labeled exosomes by hepatoma recipient cells was monitored by confocal microscopy. TGFBR2-dependent exosomal effects on secreted cytokine levels of recipient cells were analyzed by Luminex technology and ELISA. Results: Frameshift mutation patterns in microsatellite stretches of TGFBR2 and other MSI target genes were found to be reflected in the cargo of MSI CRC-derived exosomes. At the proteome level, reconstituted TGFBR2 expression and signaling uncovered two protein subsets exclusively occurring in exosomes derived from TGFBR2-deficient (14 proteins) or TGFBR2-proficient (five proteins) MSI donor cells. Uptake of these exosomes by recipient cells caused increased secretion (2–6 fold) of specific cytokines (Interleukin-4, Stem Cell Factor, Platelet-derived Growth Factor-B), depending on the TGFBR2 expression status of the tumor cell. Conclusion: Our results indicate that the coding MSI phenotype of DNA mismatch repair-deficient CRC cells is maintained in their exosomal DNA. Moreover, we uncovered that a recurrent MSI tumor driver mutation like TGFBR2 can reprogram the protein content of MSI cell-derived exosomes and in turn modulate the cytokine secretion profile of recipient cells. Apart from its diagnostic potential, these TGFBR2-dependent exosomal molecular and proteomic signatures might help to understand the signaling routes used by MSI tumors.     Advanced Flow Cytometry Assays for Immune Monitoring of CAR-T Cell Applications

    Adoptive immunotherapy using chimeric antigen receptor (CAR)-T cells has achieved successful remissions in refractory B-cell leukemia and B-cell lymphomas. In order to estimate both success and severe side effects of CAR-T cell therapies, longitudinal monitoring of the patient’s immune system including CAR-T cells is desirable to accompany clinical staging. To conduct research on the fate and immunological impact of infused CAR-T cells, we established standardized 13-colour/15-parameter flow cytometry assays that are suitable to characterize immune cell subpopulations in the peripheral blood during CAR-T cell treatment. The respective staining technology is based on pre-formulated dry antibody panels in a uniform format. Additionally, further antibodies of choice can be added to address specific clinical or research questions. We designed panels for the anti-CD19 CAR-T therapy and, as a proof of concept, we assessed a healthy individual and three B-cell lymphoma patients treated with anti-CD19 CAR-T cells. We analyzed the presence of anti-CD19 CAR-T cells as well as residual CD19+ B cells, the activation status of the T-cell compartment, the expression of co-stimulatory signaling molecules and cytotoxic agents such as perforin and granzyme B. In summary, this work introduces standardized and modular flow cytometry assays for CAR-T cell clinical research, which could also be adapted in the future as quality controls during the CART cell manufacturing process

    Nach einer kurzen Einführung in den Begriff der Digital Humanities (zu deutsch "Digitale Geisteswissenschaften") soll eine Erkundung von Nutzungsoptionen für digitale Tools und Anwendungen in der Germanistik bzw. im Studium des Deutschen als Fremdsprache (DaF) erfolgen. Dabei wird fokussiert auf den sogenannten low-end-Bereich, technisch schon vorhandene bzw. ohne größere technische Infrastruktur und ohne langen zeitlichen Vorlauf von jedem einsetzbare Tools oder Techniken. Es geht dabei vorzugsweise um das in den Digital Humanities lange weniger beachtete Feld der Lehre und auch um von Studierenden nutzbare Text-Tools und vorhandene Textkorpora.
