    Epigallocatechin-3-gallate PEGylated poly(lactic-co-glycolic) acid nanoparticles mitigate striatal pathology and motor deficits in 3-nitropropionic acid intoxicated mice

    Huntington's disease (HD) is a debilitating neurodegenerative disease that affects around 5-10/100,000 individuals in developed countries. It is caused by genetic alterations in the huntingtin (htt) gene. Efforts are being made to find treatments which will correct the genetic alterations or their pathophysiological consequences associated with HD,3 however none of these options are yet available to patients. Thus, therapies that address and ameliorate the symptomatology of HD, which include motor dysfunction and a wide range of behavioural disturbances, are also needed. Epigallocatechin-3-gallate (EGCG) is a powerful compound extracted from the green tea plant that may possess beneficial effects for HD patients, but whose therapeutic success is limited because of its chemical instability. Here, we show that protective encapsulation of EGCG rendered it much more efficient in attenuating motor deficits and depression-like behaviour in a mice model of HD-like neurodegeneration. Importantly, behavioural improvement was also associated with a reduction of neuronal damage. These results, together with our previous findings using nanoparticle-encapsulated EGCG in mouse models of Alzheimer's disease and epilepsy, highlight their potential effectiveness for symptomatic treatment of neurodegenerative diseases

    Controls on the grain size distribution of landslides in Taiwan: the influence of drop height, scar depth and bedrock strength

    International audienceThe size of grains delivered to rivers by hillslope processes is thought to be a key factor controlling sediment transport, long-term erosion and the information recorded in sedimentary archives. Recently, models have been developed to estimate the grain size distribution produced in soil, but these models may not apply to active orogens where high erosion rates on hillslopes are driven by landsliding. To date, relatively few studies have focused on landslide grain size distributions. Here, we present grain size distributions (GSDs) obtained by grid-by-number sampling on 17 recent landslide deposits in Taiwan, and we compare these GSDs to the geometrical and physical properties of the landslides, such as their width, area, rock type, drop height and estimated scar depth. All slides occurred in slightly metamorphosed sedimentary units, except two, which occurred in younger unmetamorphosed shales, with a rock strength that is expected to be 3–10 times weaker than their metamorphosed counterparts. For 11 landslides, we did not observe substantial spatial variations in the GSD over the deposit. However, four landslides displayed a strong grain size segregation on their deposit, with the overall GSD of the downslope toe sectors being 3–10 times coarser than apex sectors. In three cases, we could also measure the GSD inside incised sectors of the landslides deposits, which presented percentiles that were 3–10 times finer than the surface of the deposit. Both observations could be due to either kinetic sieving or deposit reworking after the landslide failure, but we cannot explain why only some deposits had strong segregation. Averaging this spatial variability, we found the median grain size of the deposits to be strongly negatively correlated with drop height, scar width and depth. However, previous work suggests that regolith particles and bedrock blocks should coarsen with increasing depth, which is the inverse of our observations. Accounting for a model of regolith coarsening with depth, we found that the ratio of the estimated original bedrock block size to the deposit median grain size (D50) of the deposit was proportional to the potential energy of the landslide normalized to its bedrock strength. Thus, the studied landslides agree well with a published, simple fragmentation model, even if that model was calibrated on rock avalanches with larger volume and stronger bedrock than those featured in our dataset. Therefore, this scaling may serve for future modeling of grain size transfer from hillslopes to rivers, with the aim to better understanding landslide sediment evacuation and coupling to river erosional dynamics

    Runoff-driven export of particulate organic carbon from soil in temperate forested uplands

    We characterise the sources, pathways and export fluxes of particulate organic carbon (POC) in a headwater catchment in the Swiss Alps, where suspended sediment has a mean organic carbon concentration of 1.45% ± 0.06 . By chemically fingerprinting this carbon and its potential sources using carbon and nitrogen elemental and isotopic compositions, we show that it derives from binary mixing between bedrock and modern biomass with a soil-like composition. The hillslope and channel are strongly coupled, allowing runoff to deliver recent organic carbon directly to the stream beyond a moderate discharge threshold. At higher flows, more biomass is mobilized and the fraction of modern carbon in the suspended load reaches 0.70, increased from 0.30 during background conditions. Significant amounts of non-fossil organic carbon are thus transferred from the hillslope without the need for extreme events such as landsliding. Precipitation is key: as soon as the rain stops, biomass supply ceases and fossil carbon again dominates. We use rating curves modeled using samples from five storm events integrated over 29-year discharge records to calculate long-term export fluxes of total POC and non-fossil POC from the catchment of 23.3 ± 5.8 and 14.0 ± 4.4 tonnes km-2 yr-1 respectively. These yields are comparable to those from active mountain belts, yet the processes responsible are much more widely applicable. Such settings have the potential to play a significant role in the global drawdown of carbon dioxide via riverine biomass erosion, and their contribution to the global flux of POC to the ocean may be more important than previously thought

    Concentration‐discharge relationships of dissolved rhenium in Alpine catchments reveal its use as a tracer of oxidative weathering

    Oxidative weathering of sedimentary rocks plays an important role in the global carbon cycle. Rhenium (Re) has been proposed as a tracer of rock organic carbon (OCpetro) oxidation. However, the sources of Re and its mobilization by hydrological processes remain poorly constrained. Here, we examine dissolved Re as a function of water discharge, using samples collected from three alpine catchments that drain sedimentary rocks in Switzerland (Erlenbach and Vogelbach) and Colorado, USA (East River). The Swiss catchments reveal a higher dissolved Re flux in the catchment with higher erosion rates, but have similar [Re]/[Na+] and [Re]/[SO42−] ratios, which indicate a dominance of Re from OCpetro. Despite differences in rock type and hydro-climatic setting, the three catchments have a positive correlation between river water [Re]/[Na+] and [Re]/[SO42−] and water discharge. We propose that this reflects preferential routing of Re from a near-surface, oxidative weathering zone. The observations support the use of Re as a proxy to trace rock-organic carbon oxidation, and suggest it may be a hydrological tracer of vadose zone processes. We apply the Re proxy and estimate CO2 release by OCpetro oxidation of 5.7 +6.6/−2.0 tC km−2 yr−1 for the Erlenbach. The overall weathering intensity was ∌40%, meaning that the corresponding export of unweathered OCpetro in river sediments is large, and the findings call for more measurements of OCpetro oxidation in mountains and rivers as they cross floodplains

    Nanomedicine-based technologies and novel biomarkers for the diagnosis and treatment of Alzheimer's disease: from current to future challenges

    t Increasing life expectancy has led to an aging population, which has consequently increased the prevalence of dementia. Alzheimer's disease (AD), the most common form of dementia worldwide, is estimated to make up 50-80% of all cases. AD cases are expected to reach 131 million by 2050, and this increasing prevalence will critically burden economies and health systems in the next decades. There is currently no treatment that can stop or reverse disease progression. In addition, the late diagnosis of AD constitutes a major obstacle to efective disease management. Therefore, improved diagnostic tools and new treatments for AD are urgently needed. In this review, we investigate and describe both well-established and recently discovered AD biomarkers that could potentially be used to detect AD at early stages and allow the monitoring of disease progression. Proteins such as NfL, MMPs, p-tau217, YKL-40, SNAP-25, VCAM-1, and Ng / BACE are some of the most promising biomarkers because of their successful use as diagnostic tools. In addition, we explore the most recent molecular strategies for an AD therapeutic approach and nanomedicine-based technologies, used to both target drugs to the brain and serve as devices for tracking disease progression diagnostic biomarkers. State-of-the-art nanoparticles, such as polymeric, lipid, and metal-based, are being widely investigated for their potential to improve the efectiveness of both conventional drugs and novel compounds for treating AD. The most recent studies on these nanodevices are deeply explained and discussed in this review.     Dual-drug loaded nanoparticles of Epigallocatechin-3-gallate (EGCG)/Ascorbic acid enhance therapeutic efficacy of ECGC in a APPswe/PS1dE9 Alzheimer's disease mice model.

    Epigallocatechin-3-gallate (EGCG) is a candidate for treatment of Alzheimer's disease (AD) but its inherent instability limits bioavailability and effectiveness. We found that EGCG displayed increased stability when formulated as dualdrug loaded PEGylated PLGA nanoparticles (EGCG/AA NPs). Oral administration of EGCG/AA NPs in mice resulted in EGCG accumulation in all major organs, including the brain. Pharmacokinetic comparison of plasma and brain accumulation following oral administration of free or EGCG/AA NPs showed that, whilst in both cases initial EGCG concentrations were similar, long-term (5-25 h) concentrations were ca. 5 fold higher with EGCG/AA NPs. No evidence was found that EGCG/AA NPs utilised a specific pathway across the blood-brain barrier (BBB). However, EGCG, empty NPs and EGCG/AA NPs all induced tight junction disruption and opened the BBB in vitro and ex vivo. Oral treatment of APPswe/PS1dE9 (APP/PS1) mice, a familial model of AD, with EGCG/AA NPs resulted in a marked increase in synapses, as judged by synaptophysin (SYP) expression, and reduction of neuroinflammation as well as amyloid ÎČ (AÎČ) plaque burden and cortical levels of soluble and insoluble AÎČ(1-42) peptide. These morphological changes were accompanied by significantly enhanced spatial learning and memory. Mechanistically, we propose that stabilisation of EGCG in NPs complexes and a destabilized BBB led to higher therapeutic EGCG concentrations in the brain.     Sampling and Definitions of Placental Lesions: Amsterdam Placental Workshop Group Consensus Statement

    Context.-The value of placental examination in investigations of adverse pregnancy outcomes may be compromised by sampling and definition differences between laboratories. Objective.-To establish an agreed-upon protocol for sampling the placenta, and for diagnostic criteria for placental lesions. Recommendations would cover reporting placentas in tertiary centers as well as in community hospitals and district general hospitals, and are also relevant to the scientific research community. Data Sources.-Areas of controversy or uncertainty were explored prior to a 1-day meeting where placental and perinatal pathologists, and maternal-fetal medicine specialists discussed available evidence and subsequently reached consensus where possible. Conclusions.-The group agreed on sets of uniform sampling criteria, placental gross descriptors, pathologic terminologies, and diagnostic criteria. The terminology and microscopic descriptions for maternal vascular malperfusion, fetal vascular malperfusion, delayed villous maturation, patterns of ascending intrauterine infection, and villitis of unknown etiology were agreed upon. Topics requiring further discussion were highlighted. Ongoing developments in our understanding of the pathology of the placenta, scientific bases of the maternofetoplacental triad, and evolution of the clinical significance of defined lesions may necessitate further refinements of these consensus guidelines. The proposed structure will assist in international comparability of clinicopathologic and scientific studies and assist in refining the significance of lesions associated with adverse pregnancy and later health outcomes

    p120-Catenin regulates leukocyte transmigration through an effect on VE-cadherin phosphorylation

    Vascular endothelial–cadherin (VE-cad) is localized to adherens junctions at endothelial cell borders and forms a complex with α-, ÎČ-, Îł-, and p120-catenins (p120). We previously showed that the VE-cad complex disassociates to form short-lived “gaps” during leukocyte transendothelial migration (TEM); however, whether these gaps are required for leukocyte TEM is not clear. Recently p120 has been shown to control VE-cad surface expression through endocytosis. We hypothesized that p120 regulates VE-cad surface expression, which would in turn have functional consequences for leukocyte transmigration. Here we show that endothelial cells transduced with an adenovirus expressing p120GFP fusion protein significantly increase VE-cad expression. Moreover, endothelial junctions with high p120GFP expression largely prevent VE-cad gap formation and neutrophil leukocyte TEM; if TEM occurs, the length of time required is prolonged. We find no evidence that VE-cad endocytosis plays a role in VE-cad gap formation and instead show that this process is regulated by changes in VE-cad phosphorylation. In fact, a nonphosphorylatable VE-cad mutant prevented TEM. In summary, our studies provide compelling evidence that VE-cad gap formation is required for leukocyte transmigration and identify p120 as a critical intracellular mediator of this process through its regulation of VE-cad expression at junctions
