    HIV-1 Vpr Causes Synaptodendritic Damage in Neurons

    HIV weakens the immune system by infecting and destroying T-cells, leaving the body vulnerable to infection and the development of AIDS. Conventional treatments for HIV, such as combined anti-rectroviral therapy (cART), fail to prevent the development of HIV-associated neurocognitive disorder (HAND). Neurological dysfunction has been directly related to the invasion of HIV in the central nervous system (CNS). HIV produces neurotoxic proteins, such as the Viral Protein R (Vpr), which contribute to HAND. Astrocytes are the most abundant cells in the brain and an important HIV target. We hypothesize that astrocytes expressing Vpr will cause neuronal damage in our co-culture system. Primary astrocytes were transfected with Vpr plasmid or control (pEGFP or mock) using electroporation. Astrocytes were then co-cultured with cortical neurons. At 48 and 72 hours we collected the primary astrocytes to confirm the Vpr expression via western blot analysis. We then measured structural damage in the neurons using immunofluorescence for cytoskeletal (MAP2, f-actin) and synaptic (synaptophysin) damage. Preliminary results showed strong staining of filamentous actin and MAP2 with weak detection of synaptophysin. The positive control for neurotoxicity (2.8µM acrylamide) showed substantial damage to the cellular structure. Results for Vpr expression are pending. After confirming that the immunofluorescence assays are working with our controls, we expect to detect any synaptodendritic damage in the neurons caused by Vpr in our upcoming experiments

    Sequential determination of traces of As, Sb and hg by on-line magnetic solid phase extraction coupled with Hr-Cs-Cvg-Gfaas

    A green and rapid method was developed for the simultaneous separation/preconcentration and sequential monitoring pf arsenic, antimony and mercury by flow injection magnetic solid phase extraction coupled with on-line chemical vapor generation and determination by high resolution continuum source graphite furnace atomic absorption spectrometry. The system is based on chelating/cationic retention of the analytes onto a magnet based reactor designed to contain functionalized magnetic nanoparticles (MNPs). The MNP score allows overcoming the back-pressure problems that usually happen in SPME methods with NPs thanks to the possibility of inmobilizing the MNPs by applying an external magnetic field. Several chemical and flow variables were considered as factors in the optimization process using central composite designs. With the optimized procedure the detection limits obtained were 0.2, 0.003 and 0.4 µg/L for As, Sb and Hg respectively. For the quality control of the analytical performance and the validation of the developed method the analysis of two certified samples TM 24.3 and TMDA 54.4 Fortified Lake Waters was addressed.     Energetic study of cardioplegic hearts under ischaemia/reperfusion and [Ca2+] changes in cardiomyocytes of guinea-pig: Mitochondrial role

    Abstract Aim: To study the role of mitochondria in the recovery of guinea-pig hearts exposed to high-K+-cardioplegia (CPG) and ischaemia/reperfusion (I/R) Methods: We measured contractility and heat release in perfused guineapig hearts and cytosolic and mitochondrial Ca2+ by epifluorescence and confocal microscopy in isolated cardiomyocytes loaded with Fluo-4 or Rhod-2. Results: In hearts, CPG increased the postischaemic contractile recovery, and this was potentiated by the mNCX blocker clonazepam and the mKATP opener diazoxide, which also prevented the fall in muscle economy. Moreover, CPG prevented the stunning induced by ouabain, which was reduced by clonazepam. In cardiomyocytes, CPG increased fluorescent signals of cytosolic and mitochondrial Ca2+, while the addition of a mNCX blocker (CGP37157) increased cytosolic but reduced mitochondrial [Ca2+]. Ouabain in CPG increased cytosolic Ca2+ and resting heat, but the addition of CGP37157 reduced them, as well as mitochondrial Ca2+. Conclusions: CPG, diazoxide and clonazepam improve postischaemic recovery, respectively, by increasing the Ca2+ cycling and by reducing the mitochondrial Ca2+ uptake either by uniporter or by mNCX. The mitochondria compete with the leaky sarcoplasmic reticulum (SR) as sink of Ca2+ in guinea-pig hearts, affecting the postischaemic contractility. CPG also prevented the ouabain-induced dysfunction by avoiding the Ca2+ overload. Ouabain reduced the synergism between CPG and clonazepam suggesting that [Na+]i and SR load influence the mNCX role.Fil: Ragone, María Inés.     Relation Between Corrosion Degree And Pressure Needed To Produce The First Crack And Evolution Of Crack Width In Natural Exposure

    Reinforcement corrosion leads into several damage types which influence the structural loadbearing capacity, among which can be mentioned the cracking of concrete cover. This work presents results of crack width generated in concrete elements fabricated in 1990 with chlorides added in the mix and exposed to the natural atmosphere of Madrid-Spain climate. These elements, one T beam and one column. Two expressions have been fitted to the results: w = k Px / (c/φ) and w = k Px / Ro, where w is a crack width in the time, k is a proportional factor, Px is the corrosion penetration in the time, c/φ is the concrete cover/diameter relation and Ro is the original radius of the bar. The expressions were also fitted to results taken from the literature made applying a current. The beam shows larger crack widths than the beam and the accelerated tests give intermediate results. Based in all the results, although the scatter is important, it has been calculated the k and k’ slopes which resulted respectively in values of 9.5 and 35.

    Diketonylpyridinium cations as a support of new ionic liquid crystals and ion-conductive materials: analysis of counter-ion effects

    Ionic liquid crystals (ILCs) allow the combination of the high ionic conductivity of ionic liquids (ILs) with the supramolecular organization of liquid crystals (LCs). ILCs salts were obtained by the assembly of long-chained diketonylpyridinium cations of the type [HOO^(R(n)pyH)] + and BF_(4)^(-) , ReO_(4)^(-), NO_(3)^(-), CF_(3)SO_(3)^(-), CuCl_(4)^(2-) counter-ions. We have studied the thermal behavior of five series of compounds by differential scanning calorimetry (DSC) and hot stage polarized light optical microscopy (POM). All materials show thermotropic mesomorphism as well as crystalline polymorphism. X-ray diffraction of the [HOO^(R(12)pyH)][ReO_(4)] crystal reveals a layered structure with alternating polar and apolar sublayers. The mesophases also exhibit a lamellar arrangement detected by variable temperature powder X-ray diffraction. The CuCl_(4)^(2-) salts exhibit the best LC properties followed by the ReO_(4)^(-) ones due to low melting temperature and wide range of existence. The conductivity was probed for the mesophases in one species each from the ReO_(4)^(-) , and CuCl_(4)^(2-) families, and for the solid phase in one of the non-mesomorphic Cl^(-) salts. The highest ionic conductivity was found for the smectic mesophase of the ReO_(4)^(-) containing salt, whereas the solid phases of all salts were dominated by electronic contributions. The ionic conductivity may be favored by the mesophase lamellar structure

    Este trabajo analiza la integración de los ayuntamientos que, contrario a la democracia, son mediante planillas bloqueadas y cerradas donde el pueblo ha delegado su poder en representantes que, en la mayoría de los casos, lo han usado para su beneficio personal y partidista. Esta situación neutraliza cualquier representación política o social distinta donde el actuar de los regidores es direccionado por la figura del presidente municipal, quien a pesar de ser un regidor más, con figura de representación, controla y manipula la toma de decisiones y genera lo que podríamos llamar un “absolutismo democrático” o “Presidente Omnipotente”. Esta conformación de ayuntamiento se ha justificado bajo el argumento de la gobernabilidad, misma que es opuesta, en muchas ocasiones, al interés general, pues presta más atención a la forma que al fondo, donde pareciera que por el sólo hecho de haber ganado de forma democrática, se puede tener un ejercicio de gobierno impositivo. Por ello, es fundamental que los regidores sean votados de forma individual, por separado del presidente municipal y síndico, para generar independencia, cercanía, vinculación territorial en su desempeño, y tomar en cuenta a mayorías y minorías para una mayor participación ciudadana y lograr un ejercicio de gobierno que dé resultados a la sociedad

    Evidence of an evolved nature of MWC 349A

    The Galactic emission-line object MWC 349A is one of the brightest radio stars in the sky. The central object is embedded in an almost edge-on oriented Keplerian rotating thick disc that seems to drive a rotating bipolar wind. The dense disc is also the site of hot molecular emission such as the CO bands with its prominent band heads in the near-infrared spectral range. Despite numerous studies, the nature of MWC 349A is still controversial with classifications ranging from a pre-main sequenceobject to an evolved supergiant. We collected new high-resolution near-infrared spectra in the K- and L-bands using the GNIRS spectrograph at Gemini-North to study the molecular disc of MWC 349A, and in particular to search for other molecular species such as SiO and the isotope 13 CO. The amount of 13 CO, obtained from the 12 CO/ 13 COratio, is recognized as an excellent tool to discriminate between pre-main sequence and evolved massive stars. We find no signatures of SiO band emission, but detect CO band emission with considerably lower intensity and CO gas temperature compared to previous observations. Moreover, from detailed modelling of the emission spectrum,we derive an isotope ratio of 12 CO/ 13 CO = 4±1. Based on this significant enrichment of the circumstellar environment in 13 CO we conclude that MWC 349A belongs to the group of B[e] supergiants, and we discuss possible reasons for the drop in CO intensity.Fil: Kraus, M.
