    The complete mitochondrial genome of a predominant parasitoid, Necremnus tutae (Hymenoptera: Eulophidae) (GenBank accession number MT916846) is 15,252 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes, and an A + T-rich region. The overall base composition is 38.86% for A, 7.14% for C, 8.57% for G, and 45.43% for T, with a high AT bias of 84.29%. ATA, ATT, ATG were initiation codons and TAA and T were termination codons. All the 22 tRNAs displayed a typical cloverleaf secondary structure, except for trnS1 and trnR which lacked the dihydrouracil (DHU) arm. Phylogenetic analyses were performed using 13 PCGs showed that N. tutae is closely related to Tenthredo tienmushana, which in accordance with the traditional classification.info:eu-repo/semantics/publishedVersio

    Satellite-Observed Major Greening and Biomass Increase in South China Karst During Recent Decade

    International audienceAbove-ground vegetation biomass is one of the major carbon sinks and provides both provisioning (e.g., forestry products) and regulating ecosystem services (by sequestering carbon). Continuing deforestation and climate change threaten this natural resource but can effectively be countered by national conservation policies. Here we present time series (1999-2017) derived from complementary satellite systems to describe a phenomenon of global significance: the greening of South China Karst. We find a major increase in growing season vegetation cover from 69% in 1999 to 81% in 2017 occurring over similar to 1.4 million km(2). Over 1999-2012, we report one of the globally largest increases in biomass to occur in the South China Karst region (on average +4% over 0.9 million km(2)), which accounts for similar to 5% of the global areas characterized with increases in biomass. These increases in southern China's vegetation have occurred despite a decline in rainfall (-8%) and soil moisture (-5%) between 1999 and 2012 and are derived from effects of forestry and conservation activities at an unprecedented spatial scale in human history (similar to 20,000km(2)yr(-1) since 2002). These findings have major implications for the provisioning of ecosystem services not only for the Chinese karst ecosystem (e.g., carbon storage, water filtration, and timber production) but also for the study of global carbon cycles

    Familial CJD Associated PrP Mutants within Transmembrane Region Induced Ctm-PrP Retention in ER and Triggered Apoptosis by ER Stress in SH-SY5Y Cells

    BACKGROUND: Genetic prion diseases are linked to point and inserted mutations in the prion protein (PrP) gene that are presumed to favor conversion of the cellular isoform of PrP (PrP(C)) to the pathogenic one (PrP(Sc)). The pathogenic mechanisms and the subcellular sites of the conversion are not completely understood. Here we introduce several PRNP gene mutations (such as, PrP-KDEL, PrP-3AV, PrP-A117V, PrP-G114V, PrP-P102L and PrP-E200K) into the cultured cells in order to explore the pathogenic mechanism of familial prion disease. METHODOLOGY/PRINCIPAL FINDINGS: To address the roles of aberrant retention of PrP in endoplasmic reticulum (ER), the recombinant plasmids expressing full-length human PrP tailed with an ER signal peptide at the COOH-terminal (PrP-KDEL) and PrP with three amino acids exchange in transmembrane region (PrP-3AV) were constructed. In the preparations of transient transfections, 18-kD COOH-terminal proteolytic resistant fragments (Ctm-PrP) were detected in the cells expressing PrP-KDEL and PrP-3AV. Analyses of the cell viabilities in the presences of tunicamycin and brefeldin A revealed that expressions of PrP-KDEL and PrP-3AV sensitized the transfected cells to ER stress stimuli. Western blots and RT-PCR identified the clear alternations of ER stress associated events in the cells expressing PrP-KDEL and PrP-3AV that induced ER mediated apoptosis by CHOP and caspase-12 apoptosis pathway. Moreover, several familial CJD related PrP mutants were transiently introduced into the cultured cells. Only the mutants within the transmembrane region (G114V and A117V) induced the formation of Ctm-PrP and caused the ER stress, while the mutants outside the transmembrane region (P102L and E200K) failed. CONCLUSIONS/SIGNIFICANCE: The data indicate that the retention of PrP in ER through formation of Ctm-PrP results in ER stress and cell apoptosis. The cytopathic activities caused by different familial CJD associated PrP mutants may vary, among them the mutants within the transmembrane region undergo an ER-stress mediated cell apoptosis

    Research on the development and testing methods of physical education and agility training equipment in universities

    IntroductionBecause of the problems of insufficient funds and traditional training methods in college sports agile training, an agile training system based on a wireless ad hoc network was developed to evaluate the effect of improving the sensitive quality of ordinary college students. Based on the ESP-MESH network, the lower computer realizes automatic networking between devices and tests the performance of the mesh network. Fourteen male college students received 9 weeks of agility training, with seven students in each of two groups: traditional agility training and agile equipment training. The researchers evaluated the performance of both groups in rapid disguise, body coordination, changing movements, and predictive decision-making.ResultsThere was no significant difference between the groups before training, but there were significant differences in the four abilities after training (p < 0.01). The experimental group had significant differences in rapid direction change and physical coordination (p < 0.05), and in changing movement and predictive decision-making ability (p < 0.01).ConclusionBoth traditional training and agile equipment training improve the agility quality of college students, and the latter shows better results in certain abilities. However, limited by other physical qualities, the improvement of motor changes and predictive decision-making ability is not as obvious as the other two abilities

    PI3K-GLUT4 Signal Pathway Associated with Effects of EX-B3 Electroacupuncture on Hyperglycemia and Insulin Resistance of T2DM Rats

    Objectives. To explore electroacupuncture’s (EA’s) effects on fasting blood glucose (FBG) and insulin resistance of type 2 diabetic mellitus (T2DM) model rats and give a possible explanation for the effects. Method. It takes high fat diet and intraperitoneal injection of streptozotocin (STZ, 30 mg/kg) for model preparation. Model rats were randomly divided into T2DM Model group, EA weiwanxiashu (EX-B3) group, and sham EA group (n=12/group). EA (2 Hz continuous wave, 2 mA, 20 min/day, 6 days/week, 4 weeks) was applied as intervention. FBG, area under curve (AUC) of oral glucose tolerance test (OGTT), insulin resistance index (HOMA-IR), pancreatic B cell function index (HOMA-B), skeletal muscle phosphorylated phosphatidylinositol-3-kinase (PI3K), glucose transporter 4 (GLUT4), and membrane GLUT4 protein expression were measured. Results. EA weiwanxiashu (EX-B3) can greatly upregulate model rat’s significantly reduced skeletal muscle PI3K (Y607) and membrane GLUT4 protein expression (P<0.01), effectively reducing model rats’ FBG and AUC of OGTT (P<0.01). The effects are far superior to sham EA group. Conclusion. EA weiwanxiashu (EX-B3) can upregulate skeletal muscle phosphorylated PI3K protein expression, to stimulate membrane translocation of GLUT4 and thereby increase skeletal muscle glucose intake to treat T2DM
