    A polymer analogous reaction for the formation of imidazolium and NHC based porous polymer networks

Using a stepwise approach, first a polyimine network is formed by polymerization of the tetrafunctional amine tetrakis(4-aminophenyl)methane. This polyimine network is converted in the second step into polyimidazolium chloride and finally to a polyNHC network. Furthermore a porous Cu(II)-coordinated polyNHC network can be generated. Supercritical drying generates polymer networks with high permanent surface areas and porosities which can be applied for different catalytic reactions.     High-dimensional single cell mass cytometry analysis of the murine hematopoietic system reveals signatures induced by ageing and physiological pathogen challenges

    Background: Immune ageing is a result of repetitive microbial challenges along with cell intrinsic or systemic changes occurring during ageing. Mice under 'specific-pathogen-free' (SPF) conditions are frequently used to assess immune ageing in long-term experiments. However, physiological pathogenic challenges are reduced in SPF mice. The question arises to what extent murine experiments performed under SPF conditions are suited to analyze immune ageing in mice and serve as models for human immune ageing. Our previous comparisons of same aged mice with different microbial exposures, unambiguously identified distinct clusters of immune cells characteristic for numerous previous pathogen encounters in particular in pet shop mice. Results: We here performed single cell mass cytometry assessing splenic as secondary and bone marrow as primary lymphoid organ-derived leukocytes isolated from young versus aged SPF mice in order to delineate alterations of the murine hematopoietic system induced during ageing. We then compared immune clusters from young and aged SPF mice to pet shop mice in order to delineate alterations of the murine hematopoietic system induced by physiological pathogenic challenges and those caused by cell intrinsic or systemic changes during ageing. Notably, distinct immune signatures were similarly altered in both pet shop and aged SPF mice in comparison to young SPF mice, including increased frequencies of memory T lymphocytes, effector-cytokine producing T cells, plasma cells and mature NK cells. However, elevated frequencies of CD4(+) T cells, total NK cells, granulocytes, pDCs, cDCs and decreased frequencies of naive B cells were specifically identified only in pet shop mice. In aged SPF mice specifically the frequencies of splenic IgM(+) plasma cells, CD8(+) T cells and CD4(+) CD25(+) Treg were increased as compared to pet shop mice and young mice. Conclusions: Our study dissects firstly how ageing impacts both innate and adaptive immune cells in primary and secondary lymphoid organs. Secondly, it partly distinguishes murine intrinsic immune ageing alterations from those induced by physiological pathogen challenges highlighting the importance of designing mouse models for their use in preclinical research including vaccines and immunotherapies

We present the results of crystallographic and electron paramagnetic resonance (EPR) spectroscopic analyses of four new tetrabromidocuprate(II) compounds and compare the results with previously reported data. The cations in the new compounds are the sterically demanding benzyltriphenylphosphonium, methyltriphenylphosphonium, tetraphenylphosphonium, and hexadecyltrimethylammonium ions; they were used to achieve a reasonable separation of the paramagnetic Cu(II) ions for EPR spectroscopy. X-Ray crystallography shows that in all four complexes the [CuBr4]2− units have a distorted tetrahedral coordination geometry which is in agreement with DFT calculations. The EPR hyperfine structure was not resolved. This is due to the exchange broadening resulting from still incomplete separation of the paramagnetic Cu(II) centres. Nevertheless, the principal values of the electron Zeemann tensor (gand g⊄) of the complexes could be determined. A correlation of structural (X-ray) parameters with the spin density at the copper centres (DFT) is well reflected in the EPR spectra of the bromidocuprates. This enables the correlation of X-ray and EPR parameters to predict the structure of tetrabromidocuprates in physical states other than the crystalline state. As a result, we provide a method to structurally characterize [CuBr4]2− in, for example, ionic liquids or in solution, which has important implications for e.g. catalysis or materials science

    Geometrically defined environments direct cell division rate and subcellular YAP localization in single mouse embryonic stem cells

    Mechanotransduction via yes-associated protein (YAP) is a central mechanism for decision-making in mouse embryonic stem cells (mESCs). Nuclear localization of YAP is tightly connected to pluripotency and increases the cell division rate (CDR). How the geometry of the extracellular environment influences mechanotransduction, thereby YAP localization, and decision-making of single isolated mESCs is largely unknown. To investigate this relation, we produced well-defined 2D and 2.5D microenvironments and monitored CDR and subcellular YAP localization in single mESCs hence excluding cell–cell interactions. By systematically varying size and shape of the 2D and 2.5D substrates we observed that the geometry of the growth environment affects the CDR. Whereas CDR increases with increasing adhesive area in 2D, CDR is highest in small 2.5D micro-wells. Here, mESCs attach to all four walls and exhibit a cross-shaped cell and nuclear morphology. This observation indicates that changes in cell shape are linked to a high CDR. Inhibition of actomyosin activity abrogate these effects. Correspondingly, nuclear YAP localization decreases in inhibitor treated cells, suggesting a relation between cell shape, intracellular forces, and cell division rate. The simplicity of our system guarantees high standardization and reproducibility for monitoring stem cell reactions and allows addressing a variety of fundamental biological questions on a single cell level

    Digital Collages and Aesthetic-Communicative Networking: Insights into a Workshop of the NFKB Research Cluster Interaction and Participation

    Dieser Beitrag dokumentiert EindrĂŒcke, Beobachtungen und Ergebnisse aus einem Online-Workshop des Forschungsclusters «Interaktion und Partizipation in der Kulturellen Bildung», der die Vorstellung von Themen, Arbeitsweisen und Prinzipien des Forschungsclusters zum Ziel hatte. Dabei lag es nahe, die Auseinandersetzung selbst interaktiv und partizipativ sowie entlang der Thematik der rahmenden Tagung zu konzipieren. Ein Grossteil der Kommunikation wird hierfĂŒr parallel zur Videokonferenz auf ein digitales Whiteboard verlagert, auf dem alle Teilnehmenden individuell und gemeinsam agieren können. Als Material liegen zentrale Fragmente aus einem Text des Clusters bereit, in dem die Begriffe Interaktion und Partizipation in einem gemeinsamen Selbstversuch und geleitet vom Prinzip der Collage verhandelt werden. Dieses Textprinzip wird zum Handlungsprinzip des Workshops: Die Teilnehmenden werden dem Versuch ausgesetzt, in kleinen Gruppen auf dem digitalen Whiteboard sowohl mit dem bereitgestellten als auch mit eigenem Material selbst Collagen zu erstellen. In dem experimentellen Format des Workshops konturiert sich performativ ein Ă€sthetisch-digitaler Möglichkeits- und Erfahrungsraum, in dem die individuellen Positionen und Perspektiven aller Beteiligten ĂŒber Text, Bild und Sprache interaktiv in vernetzende Artikulationsformen treten. In den entstandenen Collagen kommen Aspekte von Ästhetik, DigitalitĂ€t und Macht in eindrĂŒcklicher Weise zum Tragen.This contribution documents – especially visually – impressions, observations and results from an online workshop «Interaction and Participation in Cultural Education», which aimed to present topics, working methods and principles of the research cluster. It made sense to conceptualize the workshop itself in an interactive and participative manner, as well as along the theme of the framing conference. A large part of the communication is shifted to a digital whiteboard parallel to the video conference, on which all participants can act individually and together. Central fragments from a text of the cluster are available as material, in which the terms interaction and participation are negotiated in a joint self-experiment and guided by the principle of collage. The text principle becomes the operating principle of the workshop: The participants are exposed to the attempt to create collages themselves in small groups on the digital whiteboard using both the provided and their own material. In the experimental format of the workshop, an aesthetic-digital space of possibility and experience is contoured performatively, in which the individual positions and perspectives of all participants interactively enter into networked forms of articulation via text, image and language. In the resulting collages, aspects of aesthetics, digitality and power come into play in an impressive way

    Major flaws in conflict prevention policies towards Africa : the conceptual deficits of international actors’ approaches and how to overcome them

    Current thinking on African conflicts suffers from misinterpretations oversimplification, lack of focus, lack of conceptual clarity, state-centrism and lack of vision). The paper analyses a variety of the dominant explanations of major international actors and donors, showing how these frequently do not distinguish with sufficient clarity between the ‘root causes’ of a conflict, its aggravating factors and its triggers. Specifically, a correct assessment of conflict prolonging (or sustaining) factors is of vital importance in Africa’s lingering confrontations. Broader approaches (e.g. “structural stability”) offer a better analytical framework than familiar one-dimensional explanations. Moreover, for explaining and dealing with violent conflicts a shift of attention from the nation-state towards the local and sub-regional level is needed.Aktuelle Analysen afrikanischer Gewaltkonflikte sind hĂ€ufig voller Fehlinterpretationen (Mangel an Differenzierung, Genauigkeit und konzeptioneller Klarheit, Staatszentriertheit, fehlende mittelfristige Zielvorstellungen). Breitere AnsĂ€tze (z. B. das Modell der Strukturellen StabilitĂ€t) könnten die Grundlage fĂŒr bessere Analyseraster und Politiken sein als eindimensionale ErklĂ€rungen. hĂ€ufig differenzieren ErklĂ€rungsansĂ€tze nicht mit ausreichender Klarheit zwischen Ursachen, verschĂ€rfenden und auslösenden Faktoren. Insbesondere die richtige Einordnung konfliktverlĂ€ngernder Faktoren ist in den jahrzehntelangen gewaltsamen Auseinandersetzungen in Afrika von zentraler Bedeutung. Das Diskussionspapier stellt die große Variationsbreite dominanter ErklĂ€rungsmuster der wichtigsten internationalen Geber und Akteure gegenĂŒber und fordert einen Perspektivenwechsel zum Einbezug der lokalen und der subregionalen Ebene fĂŒr die ErklĂ€rung und Bearbeitung gewaltsamer Konflikte
