    Cross-cultural validation and psychometric evaluation of the Self-Medication Assessment Tool (SMAT) for assessing and optimizing medication therapy management of older people

    Background, rationale and objectives: The assessment of medication management ability in the elderly can be performed using specific tools, such as the Self-Medication Assessment Tool, which considers real and simulated regimens. The objective of this study was to perform the linguistic and cultural adaptation of the Self-Medication Assessment Tool to European Portuguese and determine its psychometric properties. Methods: The adaptation commenced with the translation/back translation cycle completed by 4 independent bilingual experts. The cultural component was accomplished through an external expert meeting and a longitudinal screening of concepts and construct. The pilot study was carried out in a sample of 150 Portuguese community-dwelling elders. Descriptive data, correlations, internal reliability, response consistency and exploratory factor analysis was conducted using SPSS Statistics (v22). Results: The pilot study was carried out in a sample of 150 community-dwelling elders: 112 (74.7%) participants were women; mean age was 74.73 ± 6.43 years. The Self-Medication Assessment Tool (Portuguese version) standard regimen (simulated medication regimen) mean scores were 20.92 ± 6.83 in functional ability and 38.75 ± 5.92 in cognitive ability; the real regimen (medication taken by the elderly) mean scores were 83.74 ± 15.86 in medication recall, 96.96 ± 11.39 in adherence self-report and 4.82 ± 10.1 in intentional non-adherence. Cronbach's α were 0.87 (functional ability), 0.84 (cognitive ability), 0.57 (medication recall), 0.94 (adherence self-report) and 0.79 (intentional non-adherence). The response consistency between test and re-test was verified. Conclusions: We have developed the European Portuguese version of the Self-Medication Assessment Tool with acceptable psychometric properties which can now be employed in the study of the elderly in clinical and research contexts

    Impact of content and length of curauĂĄ fibers on mechanical behavior of extruded cementitious composites: Analysis of variance

    [EN] The use of composite materials in construction has grown considerably in recent years, such as cementitious matrices and concrete reinforced with fibers. The vegetable fibers have become an alternative due to its abundance, low cost and low energy consumption for its production, and appropriate properties mechanical. CurauĂĄ fiber is a plant native from Amazonas harvested manually in commercial farming and it is used in the manufacture of ropes and baskets or as reinforcement in composite with organic matrix of components for cars, buses and trucks. On the other hand, the extrusion process can produce composites with high-density matrix with fibers, low permeability and good interface between fiber and matrix. This process is also compatible with the use of vegetable fibers as raw materials in the production of cost-effective construction elements such as ceiling panels and drywalls. The objective of this research was use the analysis of variance (ANOVA) for evaluating with rigor mathematical the influence of content and length of curauĂĄ fibers on the mechanical behavior of the extruded cementitious composites. Composites without fibers and reinforced with 1% and 2% by mass of fibers as well as 6 mm and 10 mm of length these curauĂĄ fibers were evaluated. The composites with fibers of 10 mm have showed better mechanical results. Como discussão, infere-se que o coaching pode favorecer à inovação organizacional, garantindo uma melhor gestão das mudanças, geração de ideias e solução de problemas.     New 12S metabarcoding primers for enhanced Neotropical freshwater fish biodiversity assessment

    The megadiverse Neotropical fish fauna lacks a comprehensive and reliable DNA reference database, which hampers precise species identification and DNA based biodiversity assessment in the region. Here, we developed a mitochondrial 12S ribosomal DNA reference database for 67 fish species, representing 54 genera, 25 families, and six major Neotropical orders. We aimed to develop mini-barcode markers (i.e. amplicons with less than 200 bp) suitable for DNA metabarcoding by evaluating the taxonomic resolution of full-length and mini-barcodes and to determine a threshold value for fish species delimitation using 12S. Evaluation of the target amplicons demonstrated that both full-length library (565 bp) and mini-barcodes (193 bp) contain enough taxonomic resolution to differentiate all 67 fish species. For species delimitation, interspecific genetic distance threshold values of 0.4% and 0.55% were defined using full-length and mini-barcodes, respectively. A custom reference database and specific mini-barcode markers are important assets for ecoregion scale DNA based biodiversity assessments (such as environmental DNA) that can help with the complex task of conserving the megadiverse Neotropical ichthyofauna

    Can body mass index influence the skin temperature of adolescents? A preliminary study with the use of infrared thermography

    – Infrared thermography (IRT) has been used to assess skin temperature (Tsk), especially during the COVID-19 pandemic, as an important tool in medical screening not only of the general population, but also of young athletes. However, the subcutaneous adipose tissue can act as an insulator when the Tsk is assessed by IRT, modifying the normal Tsk data and leading to their misinterpretation. Considering that the body mass index (BMI) is an important predictor of obesity, the objective of this study was to verify if the Tsk measured by IRT is affected by the BMI in adolescents. A preliminary study was carried out being four participants intentionally selected, all 16 years old, each one classified in a different BMI range according to the criteria of the World Health Organization for the adolescent population: underweight, healthy weight, overweight and obesity. Four thermograms of each participant were recorded and the ThermoHuman¼ software was used to evaluate 82 regions of interest (ROI), which were integrated into 6 body regions. Using healthy weight subjects as a reference, it was found a progressive reduction in Tsk in all ROI compared to overweight and obese participants, with emphasis on the anterior region of the trunk (3.04% and 6.69% less respectively), and an increase in the Tsk of all body regions for the underweight subject. There are indications that BMI can influence the Tsk value in adolescents and should be taken into account when analyzing thermograms for a correct evaluation of thermal normality

    Resonating, rejecting, reinterpreting: Mapping the stabilization discourse in the United Nations Security Council, 2000–14

    This article charts the evolution of the conceptualisation of stabilization in the UN Security Council (UNSC) during the period 2001–2014. UNSC open meetings provide an important dataset for a critical review of stabilization discourse and an opportunity to chart the positions of permanent Members, rotating Members and the UN Secretariat towards this concept. This article is the first to conduct an analysis of this material to map the evolution of stabilization in this critical chamber of the UN. This dataset of official statements will be complemented by a review of open source reporting on UNSC meetings and national stabilization doctrines of the ‘P3’ – France, the UK and the US. These countries have developed national stabilization doctrines predominantly to deal with cross-governmental approaches to counterinsurgency operations conducted during the 2000s. The article therefore presents a genealogy of the concept of stabilization in the UNSC to help understand implications for its future development in this multilateral setting. This article begins by examining efforts by the P3 to ‘upload’ their conceptualisations of stabilization into UN intervention frameworks. Secondly, the article uses a content analysis of UNSC debates during 2000–2014 to explore the extent to which the conceptualisation of stabilization resonated with other Council members, were rejected in specific contexts or in general, or were re-interpreted by member states to suit alternative security agendas and interests. Therefore, the article not only examines the UNSC debates surrounding existing UN ‘stabilization operations’ (MONUSCO, MINUSTAH, MINUSCA, MINUSMA), which could be regarded as evidence that this ‘western’ concept has resonated with other UNSC members and relevant UN agencies, but also documents the appearance of stabilization in other contexts too. The article opens new avenues of research into concepts of stabilization within the UN, and seeks to provide a thorough accounting of the origins, spread, and potential trajectories for the concept and practice of stabilization in UN contexts

    A Kinetic Study of Accumulation and Elimination of Microcystin-LR in Yellow Perch (Perca Flavescens) Tissue and Implications for Human Fish Consumption

    Fish consumption is a potential route of human exposure to the hepatotoxic microcystins, especially in lakes and reservoirs that routinely experience significant toxic Microcystis blooms. Understanding the rates of uptake and elimination for microcystins as well as the transfer efficiency into tissues of consumers are important for determining the potential for microcystins to be transferred up the food web and for predicting potential human health impacts. The main objective of this work was to conduct laboratory experiments to investigate the kinetics of toxin accumulation in fish tissue. An oral route of exposure was employed in this study, in which juvenile yellow perch (Perca flavescens) were given a single oral dose of 5 or 20 ÎŒg of microcystin-LR (MC-LR) via food and accumulation in the muscle, liver, and tank water were measured over 24 h. Peak concentrations of the water soluble fraction of microcystin were generally observed 8–10 h after dosing in the liver and after 12–16 h in the muscle, with a rapid decline in both tissues by 24 h. Up to 99% of the total recoverable (i.e., unbound) microcystin was measured in the tank water by 16 h after exposure. The relatively rapid uptake and elimination of the unbound fraction of microcystin in the liver and muscle of juvenile yellow perch within 24 h of exposure indicates that fish consumption may not be a major route of human exposure to microcystin, particularly in the Great Lakes

    Whole genome sequencing of Saccharomyces cerevisiae: from genotype to phenotype for improved metabolic engineering applications

    <p>Abstract</p> <p>Background</p> <p>The need for rapid and efficient microbial cell factory design and construction are possible through the enabling technology, metabolic engineering, which is now being facilitated by systems biology approaches. Metabolic engineering is often complimented by directed evolution, where selective pressure is applied to a partially genetically engineered strain to confer a desirable phenotype. The exact genetic modification or resulting genotype that leads to the improved phenotype is often not identified or understood to enable further metabolic engineering.</p> <p>Results</p> <p>In this work we performed whole genome high-throughput sequencing and annotation can be used to identify single nucleotide polymorphisms (SNPs) between <it>Saccharomyces cerevisiae </it>strains S288c and CEN.PK113-7D. The yeast strain S288c was the first eukaryote sequenced, serving as the reference genome for the <it>Saccharomyces </it>Genome Database, while CEN.PK113-7D is a preferred laboratory strain for industrial biotechnology research. A total of 13,787 high-quality SNPs were detected between both strains (reference strain: S288c). Considering only metabolic genes (782 of 5,596 annotated genes), a total of 219 metabolism specific SNPs are distributed across 158 metabolic genes, with 85 of the SNPs being nonsynonymous (e.g., encoding amino acid modifications). Amongst metabolic SNPs detected, there was pathway enrichment in the galactose uptake pathway (<it>GAL1</it>, <it>GAL10</it>) and ergosterol biosynthetic pathway (<it>ERG8</it>, <it>ERG9</it>). Physiological characterization confirmed a strong deficiency in galactose uptake and metabolism in S288c compared to CEN.PK113-7D, and similarly, ergosterol content in CEN.PK113-7D was significantly higher in both glucose and galactose supplemented cultivations compared to S288c. Furthermore, DNA microarray profiling of S288c and CEN.PK113-7D in both glucose and galactose batch cultures did not provide a clear hypothesis for major phenotypes observed, suggesting that genotype to phenotype correlations are manifested post-transcriptionally or post-translationally either through protein concentration and/or function.</p> <p>Conclusions</p> <p>With an intensifying need for microbial cell factories that produce a wide array of target compounds, whole genome high-throughput sequencing and annotation for SNP detection can aid in better reducing and defining the metabolic landscape. This work demonstrates direct correlations between genotype and phenotype that provides clear and high-probability of success metabolic engineering targets.
