    Business model innovation and digital transformation in global management consulting firms

    Purpose: This paper contains an exploratory analysis of the business model innovations (BMIs) that management consulting firms (MCFs) undertake to remain competitive during digital transformation. Design/methodology/approach: This paper uses data from a longitudinal multiple case study of the European practices of major global MCFs to provide an overview of how they reconfigure their business model (BM) to gain competitive advantages. It maps BMIs in MCFs through value creation innovation, value proposition innovation and value capturing innovation. Findings: There is a shift in value proposition from solely giving advice or supporting information technology (IT) implementation to providing end-to-end digital solutions. To materialize value propositions, MCFs acquire new knowledge and digital assets through talent scouting, and mergers and acquisitions (M&As). MCFs rely heavily on complementary knowledge and capabilities of actors within ecosystems; thus, they focus on expanding, creating their ecosystems and adopting platforms' configuration and characteristics. Research limitations/implications: Inductively, the authors reached an analytical generalization through six propositions and a theoretical frame that embeds propositions in the previous literature. Future research should test them across the overall management consulting industry. Practical implications: MCFs are recognized as drivers of innovation and BMIs in most client firms. However, MCFs are rarely analyzed with respect to their BMIs. Understanding how MCFs innovate their business models (BMs) to provide digital transformation (DT) consulting services is relevant for delivering management innovation across industries.     Topical ruxolitinib: A new treatment for vitiligo

    Vitiligo is a chronic autoimmune skin disorder whose diagnosis is often psychologically upsetting. The efficacy of the available therapies, including topical corticosteroids and topical calcineurin inhibitors, has historically been limited and the management of vitiligo is still challenging. As vitiligo is a chronic disease limited to the skin, topical rather than systemic therapies may be preferable (especially among patients with localised lesions) to avoid the long-term side-effects of the latter. A topical formulation of ruxolitinib, a selective JAK1/2 inhibitor, has recently been approved in the United States for the treatment of non-segmental vitiligo in patients aged >12 years based on data from the phase III TRuE-V1 and TRuE-V2 clinical trials. The aim of this review is to describe the current evidence concerning the efficacy and safety of topical ruxolitinib in the treatment of vitiligo, and discuss issues regarding its use in younger children and pregnant or breastfeeding women, as well as the duration and durability of treatment. The promising results obtained so far suggest that 1.5% ruxolitinib cream is an effective means of treating vitiligo

    Distribution of antibiotic resistance genes in the saliva of healthy omnivores, ovo-lacto-vegetarians, and vegans

    Food consumption allows the entrance of bacteria and their antibiotic resistance (AR) genes into the human oral cavity. To date, very few studies have examined the influence of diet on the composition of the salivary microbiota, and even fewer investigations have specifically aimed to assess the impact of different long-term diets on the salivary resistome. In this study, the saliva of 144 healthy omnivores, ovo-lacto-vegetarians, and vegans were screened by nested PCR for the occurrence of 12 genes conferring resistance to tetracyclines, macrolide-lincosamide-streptogramin B, vancomycin, and \u3b2-lactams. The tet(W), tet(M), and erm(B) genes occurred with the highest frequencies. Overall, no effect of diet on AR gene distribution was seen. Some differences emerged at the recruiting site level, such as the higher frequency of erm(C) in the saliva of the ovo-lacto-vegetarians and omnivores from Bologna and Turin, respectively, and the higher occurrence of tet(K) in the saliva of the omnivores from Bologna. A correlation of the intake of milk and cheese with the abundance of tet(K) and erm(C) genes was seen. Finally, when the occurrence of the 12 AR genes was evaluated along with geographical location, age, and sex as sources of variability, high similarity among the 144 volunteers was seen

    Grain Yield Stability of Cereal-Legume Intercrops Is Greater Than Sole Crops in More Productive Conditions

    The intercropping of two or more crop species on the same piece of land at a given time has been hypothesized to enhance crop yield stability. To address this hypothesis, we assessed the grain yield stability of various barley-pea and wheat-faba bean mixtures grown in seven experimental field trials (locations) across Europe during two years with contrasting weather (2017 and 2018). Three different yield stability measures were used, all based on the expected yield variability of the mixture components grown as sole crops, and the corresponding observed yield variability of the same components grown in 50:50 mixtures in a replacement design. Stability indices were calculated as ratios between the expected and observed variabilities, with values > 1 indicating greater stability of the intercrops. Mean grain yields tended to be higher in intercrops than sole crops. However, in contrast to our hypothesis, the observed (intercrop) yield stability was similar or lower than the expected (sole crop) stability in most locations except one. Furthermore, yield stability significantly increased with increasing mean yields when assessed across differentially productive locations.     Positive effects of intercrop yields in farms from across Europe depend on rainfall, crop composition, and management

    \ua9 The Author(s) 2024.Modern “intensive” agriculture drives the biodiversity-climate crisis but is also central to global food security. Future farming needs management approaches that maintain (or even enhance) food production while reducing negative climate and biodiversity impacts. Intercrops could provide part of the solution, increasing biodiversity and boosting production with fewer inputs. However, barriers remain to their wide-scale uptake, in particular tailoring intercrops to local equipment, management practice, and environment. We analyze data from multiple trials of cereal-legume intercrops conducted on farms across Europe between 2018 and 2021. Our study is the first attempt, to our knowledge, to quantify the yield benefits of cereal-legume intercropping undertaken at commercially relevant scales for farms across Europe. We used crop performance ratio (CPR)—the ratio of the observed intercrop yield compared to the expected yield based on monoculture yields—as our metric of intercrop performance. Using CPR, we found a roughly 30% yield gain across all sites. However, CPR was modulated by a number of factors. CPR was not strongly affected by management except for the negative effects of direct drilling and the positive effects of organic fertilizer addition. CPR also depended on intercrop composition (number and identity of components), background yields (being highest where yields were lower), and rainfall (being higher with higher rainfall). Our findings allow us to reduce uncertainty about how intercrops will perform in realistic local farm conditions, give guidance for tailoring intercrops to local farming conditions, and provide key goals for further work to integrate intercrops into sustainable farming systems

    Florbetaben PET imaging to detect amyloid beta plaques in Alzheimer's disease: Phase 3 study

    AbstractBackgroundEvaluation of brain β-amyloid by positron emission tomography (PET) imaging can assist in the diagnosis of Alzheimer disease (AD) and other dementias.MethodsOpen-label, nonrandomized, multicenter, phase 3 study to validate the 18F-labeled β-amyloid tracer florbetaben by comparing in vivo PET imaging with post-mortem histopathology.ResultsBrain images and tissue from 74 deceased subjects (of 216 trial participants) were analyzed. Forty-six of 47 neuritic β-amyloid-positive cases were read as PET positive, and 24 of 27 neuritic β-amyloid plaque-negative cases were read as PET negative (sensitivity 97.9% [95% confidence interval or CI 93.8–100%], specificity 88.9% [95% CI 77.0–100%]). In a subgroup, a regional tissue-scan matched analysis was performed. In areas known to strongly accumulate β-amyloid plaques, sensitivity and specificity were 82% to 90%, and 86% to 95%, respectively.ConclusionsFlorbetaben PET shows high sensitivity and specificity for the detection of histopathology-confirmed neuritic β-amyloid plaques and may thus be a valuable adjunct to clinical diagnosis, particularly for the exclusion of AD.Trial registrationClinicalTrials.gov NCT01020838

    Pair-Wise Regulation of Convergence and Extension Cell Movements by Four Phosphatases via RhoA

    Various signaling pathways regulate shaping of the main body axis during early vertebrate development. Here, we focused on the role of protein-tyrosine phosphatase signaling in convergence and extension cell movements. We identified Ptpn20 as a structural paralogue of PTP-BL and both phosphatases were required for normal gastrulation cell movements. Interestingly, knockdowns of PTP-BL and Ptpn20 evoked similar developmental defects as knockdown of RPTPα and PTPε. Co-knockdown of RPTPα and PTP-BL, but not Ptpn20, had synergistic effects and conversely, PTPε and Ptpn20, but not PTP-BL, cooperated, demonstrating the specificity of our approach. RPTPα and PTPε knockdowns were rescued by constitutively active RhoA, whereas PTP-BL and Ptpn20 knockdowns were rescued by dominant negative RhoA. Consistently, RPTPα and PTP-BL had opposite effects on RhoA activation, both in a PTP-dependent manner. Downstream of the PTPs, we identified NGEF and Arhgap29, regulating RhoA activation and inactivation, respectively, in convergence and extension cell movements. We propose a model in which two phosphatases activate RhoA and two phosphatases inhibit RhoA, resulting in proper cell polarization and normal convergence and extension cell movements
