    Wideband Spectrum Sensing for Dynamic Spectrum Sharing

    The proliferation of wireless devices grows exponentially, demanding more and more data communication capacity over wireless links. Radio spectrum is a scarce resource, and traditional wireless networks deployed by Mobile Network Operators (MNO) are based on an exclusive spectrum band allocation. However, underutilization of some licensed bands in time and geographic domains has been reported, especially in rural areas or areas away from high population density zones. This coexistence of increasingly high data communication needs and spectrum underutilization is an incomprehensible scenario. A more rational and efficient use of the spectrum is the possibility of Licensed Users (known as Primary Users – PU) to lease the spectrum, when not in use, to Unlicensed Users (known as Secondary Users – SU), or allowing the SU to opportunistically use the spectrum after sensing and verifying that the PU is idle. In this latter case, the SU must stop transmitting when the PU becomes active. This thesis addresses the spectrum sensing task, which is essential to provide dynamic spectrum sharing between PUs and SUs. We show that the Spectral Correlation Function (SCF) and the Spectral Coherence Function (SCoF) can provide a robust signal detection algorithm by exploiting the cyclostationary characteristics of the data communication signal. We enhance the most used algorithm to compute de SCF - the FAM (FFT Accumulation Method) algorithm – to efficiently compute the SCF in a local/zoomed region of the support ( ; ) plane (frequency/cycle frequency plane). This will provide the quick identification of spectral bands in use by PUs or free, in a wideband sampling scenario. Further, the characterization of the probability density of the estimates of the SCF and SCoF when only noise is present, using the FAM algorithm, will allow the definition of an adaptive threshold to develop a blind (with respect to the noise statistics) Constant False Alarm Rate (CFAR) detector (using the SCoF) and also a CFAR and a Constant Detection Rate (CDR) detector when that characterization is used to obtain an estimate of the background noise variance (using the SCF).A proliferação de dispositivos sem fios cresce de forma exponencial, exigindo cada vez mais capacidade de comunicação de dados através de ligações sem fios. O espectro radioelétrico é um recurso escasso, e as redes sem fios tradicionais implantadas pelos Operadores de Redes Móveis baseiam-se numa atribuição exclusiva de bandas do espectro. No entanto, tem sido relatada a subutilização de algumas bandas licenciadas quer ao longo do tempo, quer na sua localização geográfica, especialmente em áreas rurais, e em áreas longe de zonas de elevada densidade populacional. A coexistência da necessidade cada vez maior de comunicação de dados, e a subutilização do espectro é um cenário incompreensível. Uma utilização mais racional e eficiente do espectro pressupõe a possibilidade dos Utilizadores Licenciados (conhecidos como Utilizadores Primários – Primary Users - PU) alugarem o espectro, quando este não está a ser utilizado, a Utilizadores Não Licenciados (conhecidos como Utilizadores Secundários – Secondary Users - SU), ou permitir ao SU utilizar oportunisticamente o espectro após a deteção e verificação de que o PU está inativo. Neste último caso, o SU deverá parar de transmitir quando o PU ficar ativo. Nesta tese é abordada a tarefa de deteção espectral, que é essencial para proporcionar a partilha dinâmica do espectro entre PUs e SUs. Mostra-se que a Função de Correlação Espectral (Spectral Correlation Function - SCF) e a Função de Coerência Espectral (Spectral Coherence Function - SCoF) permitem o desenvolvimento de um algoritmo robusto de deteção de sinal, explorando as características ciclo-estacionárias dos sinais de comunicação de dados. Propõe-se uma melhoria ao algoritmo mais utilizado para cálculo da SCF – o método FAM (FFT Accumulation Method) - para permitir o cálculo mais eficiente da SCF numa região local/ampliada do plano de suporte / (plano de frequência/frequência de ciclo). Esta melhoria permite a identificação rápida de bandas espectrais em uso por PUs ou livres, num cenário de amostragem de banda larga. Adicionalmente, é feita a caracterização da densidade de probabilidade das estimativas da SCF e SCoF quando apenas o ruído está presente, o que permite a definição de um limiar adaptativo, para desenvolver um detetor de Taxa de Falso Alarme Constante (Constant False Alarm Rate – CFAR) sem conhecimento do ruído de fundo (usando a SCoF) e também um detetor CFAR e Taxa de Deteção Constante (Constant Detection Rate – CDR), quando se utiliza aquela caracterização para obter uma estimativa da variância do ruído de fundo (usando a SCF)

    Guar gum as a new antimicrobial peptide delivery system against diabetic foot ulcers Staphylococcus aureus isolates

    © 2016 The AuthorsDiabetic patients frequently develop diabetic foot ulcers (DFUs), particularly those patients vulnerable to Staphylococcus aureus opportunistic infections. It is urgent to find new treatments for bacterial infections. The antimicrobial peptide (AMP) nisin is a potential candidate, mainly due to its broad spectrum of action against pathogens. Considering that AMP can be degraded or inactivated before reaching its target at therapeutic concentrations, it is mandatory to establish effective AMP delivery systems, with the natural polysaccharide guar gum being one of the most promising. We analysed the antimicrobial potential of nisin against 23 S. aureus DFU biofilm-producing isolates. Minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), minimum biofilm inhibitory concentration (MBIC) and minimum biofilm eradication concentration (MBEC) were determined for nisin diluted in HCl and incorporated in guar gum gel. Statistical analysis was performed using the Wilcoxon matched-pair test. Nisin was effective against all isolates, including some multidrug-resistant clinical isolates, independent of whether it is incorporated in guar gum. While differences among MIC, MBC and MBIC values were observed for HCl- and guar gum- nisin, no significant differences were found between MBEC values. Inhibitory activity of both systems seems to differ only twofold, which does not compromise guar gum gel efficiency as a delivery system. Our results highlight the potential of nisin as a substitute for or complementary therapy to current antibiotics used for treating DFU infections, which is extremely relevant considering the increase in multidrug-resistant bacteria dissemination.     Viral genetic clustering and transmission dynamics of the 2022 mpox outbreak in Portugal

    Pathogen genome sequencing during epidemics enhances our ability to identify and understand suspected clusters and investigate their relationships. Here, we combine genomic and epidemiological data of the 2022 mpox outbreak to better understand early viral spread, diversification and transmission dynamics. By sequencing 52% of the confirmed cases in Portugal, we identified the mpox virus sublineages with the highest impact on case numbers and fitted them into a global context, finding evidence that several international sublineages probably emerged or spread early in Portugal. We estimated a 62% infection reporting rate and that 1.3% of the population of men who have sex with men in Portugal were infected. We infer the critical role played by sexual networks and superspreader gatherings, such as sauna attendance, in the dissemination of mpox virus. Overall, our findings highlight genomic epidemiology as a tool for the real-time monitoring and control of mpox epidemics, and can guide future vaccine policy in a highly susceptible population.info:eu-repo/semantics/publishedVersio

    Recent historians have rehabilitated King Duarte of Portugal, previously maligned and neglected, as an astute ruler and philosopher. There is still a tendency, however, to view Duarte as a depressive or a hypochondriac, due to his own description of his melancholy in his advice book, the Loyal Counselor. This paper reassesses Duarte's writings, drawing on key approaches in the history of medicine, such as narrative medicine and the history of the patient. It is important to take Duarte's views on his condition seriously, placing them in the medical and theological contexts of his time and avoiding modern retrospective diagnosis. Duarte's writings can be used to explore the impact of plague, doubt and death on the life of a well-educated and conscientious late-medieval ruler

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Dendritic cell (DC)-based antitumor vaccines have proven to be a safe approach, but often fail to generate robust results between trials. Translation to the clinic has been hindered in part by the lack of standard operation procedures for vaccines production, namely the definition of optimal culture conditions during ex-vivo DC differentiation. Here we sought to compare the ability of three clinical grade serum-free media, DendriMACS, AIM-V, and X-VIVO 15, alongside with fetal bovine serum-supplemented Roswell Park Memorial Institute Medium (RPMI), to support the differentiation of monocyte-derived DCs (Mo-DCs). Under these different culture conditions, phenotype, cell metabolomic profiles, response to maturation stimuli, cytokines production, allogenic T cell stimulatory capacity, as well as priming of antigen-specific CD8+ T cells and activation of autologous natural killer (NK) cells were analyzed. Immature Mo-DCs differentiated in AIM-V or X-VIVO 15 presented lower levels of CD1c, CD1a, and higher expression of CD11c, when compared to cells obtained with DendriMACS. Upon stimulation, only AIM-V or X-VIVO 15 DCs acquired a full mature phenotype, which supports their enhanced capacity to polarize T helper cell type 1 subset, to prime antigen-specific CD8+ T cells and to activate NK cells. CD8+ T cells and NK cells resulting from co-culture with AIM-V or X-VIVO 15 DCs also showed superior cytolytic activity. 1H nuclear magnetic resonance-based metabolomic analysis revealed that superior DC immunostimulatory capacities correlate with an enhanced catabolism of amino acids and glucose. Overall, our data highlight the impact of critically defining the culture medium used in the production of DCs for clinical application in cancer immunotherapy. Moreover, the manipulation of metabolic state during differentiation could be envisaged as a strategy to enhance desired cell characteristics

    As Residências Cistercienses em S. Bento de Cástris que se vêm realizando desde 2013 têm como primeiro objectivo reinventar na contemporaneidade a densidade histórica do discurso cisterciense, integrando a realidade deste mosteiro tanto numa ampla geografia da Ordem de Cister em Portugal e na Europa, como na história da região e do país. Inspiradas nas questões da História, da Arte, da Arquitectura, da Música, do Património e da Paisagem, as Residências Cistercienses em Cástris vêm apostando no debate de questões actuais ligadas aos espaços monásticos e ao seu futuro, nomeadamente os cistercienses. Esta aposta concretiza-se também no presente trabalho, DO ESPÍRITO DO LUGAR. ESTÉTICA. SILÊNCIO, ESPAÇO, LUZ, resultado das I e II Residências Cistercienses (2013 e 2014), e integra estudos que se reportam especialmente ao Silêncio e às suas várias linguagens e significados e às dimensões da Estética monástico-religiosa, com algum privilégio para as temáticas da Música. A apropriação do mosteiro pelo silêncio e pela música e a apreciação do espólio musical e instrumental de S. Bento de Cástris permitem uma melhor percepção das diversidades conjunturais e dos seus ritmos, aliando-se às dimensões de valorização patrimonial, eacompanhando-as de sugestões estéticas na pintura, escultura, pintura mural, azulejaria…, no sentido da fruição plena dos espaços. A riqueza deste evento, a Residência Cisterciense no mosteiro de S. Bento de Cástris, permitirá que o futuro deste património, de tanta diversidade e espessura histórico-cultural, continue a ser debatido e a justificar edições futuras resultantes dos trabalhos de investigação aí apresentados e debatidos
