    A majority of patients with cancer, in particularly cancer of digestive organs, shows malnutrition, leading to worse quality of life and occurrence of several complications. Mechanisms of being malnutrition include anorexia, abnormally affected metabolism, hypercytokinemia, chronic loss of blood, and consumption of nutrition by cancer growing. Nutritional states should be evaluated periodically by somatometry, blood chemistry, subjective nutritional assessment(SGA), and so on. On the basis of the assessment of nutrition, appropriate supplementation of nutrition should be performed. The routs of nutritional supplementation include oral ingestion, tubural feeding, gastrostomy tube, enterostomy tube, peripheral parenteral nutrition and intravenous hyperalimenation. Thus, appropriate nutritional administration in patients with cancer leads to better outcome of cancer treatment

    Prediction of pharmacological activities from chemical structures with graph convolutional neural networks

    化合物の薬理作用を予測する技術を開発 --薬理作用ビッグデータを用いて--. 京都大学プレスリリース. 2021-01-13.Many therapeutic drugs are compounds that can be represented by simple chemical structures, which contain important determinants of affinity at the site of action. Recently, graph convolutional neural network (GCN) models have exhibited excellent results in classifying the activity of such compounds. For models that make quantitative predictions of activity, more complex information has been utilized, such as the three-dimensional structures of compounds and the amino acid sequences of their respective target proteins. As another approach, we hypothesized that if sufficient experimental data were available and there were enough nodes in hidden layers, a simple compound representation would quantitatively predict activity with satisfactory accuracy. In this study, we report that GCN models constructed solely from the two-dimensional structural information of compounds demonstrated a high degree of activity predictability against 127 diverse targets from the ChEMBL database. Using the information entropy as a metric, we also show that the structural diversity had less effect on the prediction performance. Finally, we report that virtual screening using the constructed model identified a new serotonin transporter inhibitor with activity comparable to that of a marketed drug in vitro and exhibited antidepressant effects in behavioural studies

    Overexpression of hepatocyte growth factor/scatter factor promotes vascularization and granulation tissue formation in vivo

    AbstractThe effect of hepatocyte growth factor/scatter factor (HGF/SF) during wound healing in the skin was investigated, using HGF/SF-overexpressing transgenic mouse model. Histological analysis of HGF/SF transgenic mouse excisional wound sites revealed increased granulation tissue with marked vascularization. Northern blot analysis demonstrated that, relative to control, vascular endothelial growth factor (VEGF) expression in transgenic skin was significantly higher at baseline and was robustly up-regulated during wound healing. Elevated levels of VEGF protein were detected immunohistochemically, predominantly in endothelial cells and fibroblasts within the granulation tissue of HGF/SF transgenic skin. Serum levels of VEGF were also elevated in HGF/SF transgenic mice. Thus, results from our study suggest that HGF/SF has a significant effect on vascularization and granulation tissue formation during wound healing in vivo, involving with induction of VEGF

    The Role of Dorsal Raphe Serotonin Neurons in the Balance between Reward and Aversion

    Background: Reward processing is fundamental for animals to survive and reproduce. Many studies have shown the importance of dorsal raphe nucleus (DRN) serotonin (5-HT) neurons in this process, but the strongly correlative link between the activity of DRN 5-HT neurons and rewarding/aversive potency is under debate. Our primary objective was to reveal this link using two different strategies to transduce DRN 5-HT neurons. Methods: For transduction of 5-HT neurons in wildtype mice, adeno-associated virus (AAV) bearing the mouse tryptophan hydroxylase 2 (TPH2) gene promoter was used. For transduction in Tph2-tTA transgenic mice, AAVs bearing the tTA-dependent TetO enhancer were used. To manipulate the activity of 5-HT neurons, optogenetic actuators (CheRiff, eArchT) were expressed by AAVs. For measurement of rewarding/aversive potency, we performed a nose-poke self-stimulation test and conditioned place preference (CPP) test. Results: We found that stimulation of DRN 5-HT neurons and their projections to the ventral tegmental area (VTA) increased the number of nose-pokes in self-stimulation test and CPP scores in both targeting methods. Concomitantly, CPP scores were decreased by inhibition of DRN 5-HT neurons and their projections to VTA. Conclusion: Our findings indicate that the activity of DRN 5-HT neurons projecting to the VTA is a key modulator of balance between reward and aversion

    本総説は,2010年に学術雑誌に掲載されたX線分析関連の論文において,注目すべき論文を厳選し紹介する.調査した学術雑誌は19件(和雑誌2件含む)であり,X線分析の発展に寄与しているものを対象としているが,分析化学の分野だけでなく,分光学や物理学の分野も網羅している.各雑誌に関して,X 線分析手法や測定された試料の傾向,分析技術や要素開発に関するトピックスの他に,特筆すべき論文には論評も記している.日本工業規格(JIS)におけるX線分析関連の規格の制定や改訂についてまとめている.X線関連メーカーのウェブサイトを紹介し,掲載している技術レポートの情報も得られる. In this article, the interesting X-ray analysis-related literatures, which are published on academic journals during the year 2010, are summarized. The number of the researched journals is 19, including two Japanese journals, which contribute the advance of the X-ray analysis in the field not only of analytical chemistry but also of spectroscopy and physics. In every journal, the trend of the X-ray analysis methods and of the measured specimens, the topics of analytical technique and of the developed components of the X-ray analysis apparatus, and the comments for the nortable articles are mentioned. The constitution and revision of standards of X-ray analysis on Japanease Industrial Standard (JIS) are shown. The websites of the company related with X-ray tools or X-ray apparatus are also shown, and the information on technical reports is available
