    Characterizing larval swordfish habitat in the western tropical North Atlantic

Determining the distribution of swordfish larvae is important for indicating reproductive activity and understanding the early life history of swordfish. We identify and characterize larval swordfish distributions during peak swordfish spawning throughout the Gulf of Mexico and western Caribbean Sea with generalized additive models (GAMs) using catches of swordfish larvae during ichthyoplankton surveys in April and May of 2010, 2011, and 2012. The best fit GAM, as determined by stepwise, backward Akaike Information Criterion selection, included both physiochemical (temperature at 5 m, sea surface height anomaly (SSHA), eddy kinetic energy (EKE)), temporal (lunar illumination, hour of sampling) and spatial (location) variables, while near-surface chlorophyll a concentration residuals remained as a random effect. The highest probability of larval swordfish catch occurred at sub-surface temperatures, SSHA, and EKE values indicative of boundary currents. Standard lengths of larvae were larger further downstream in the boundary currents, despite high variability in length with location due to multiple spawning locations of swordfish near these currents. Probability of larval swordfish catch also peaked during the crescent and gibbous moons, indicating a lunar periodicity to swordfish spawning. These results suggest that swordfish may spawn during select moon phases near boundary currents that transport their larvae to larval and juvenile habitat including the northern Gulf of Mexico and coastal waters of the southeast United States.NASA Grant Numbers: NNX11AP76G, NNX08AL06

    The velocity distribution of nearby stars from Hipparcos data I. The significance of the moving groups

    We present a three-dimensional reconstruction of the velocity distribution of nearby stars (<~ 100 pc) using a maximum likelihood density estimation technique applied to the two-dimensional tangential velocities of stars. The underlying distribution is modeled as a mixture of Gaussian components. The algorithm reconstructs the error-deconvolved distribution function, even when the individual stars have unique error and missing-data properties. We apply this technique to the tangential velocity measurements from a kinematically unbiased sample of 11,865 main sequence stars observed by the Hipparcos satellite. We explore various methods for validating the complexity of the resulting velocity distribution function, including criteria based on Bayesian model selection and how accurately our reconstruction predicts the radial velocities of a sample of stars from the Geneva-Copenhagen survey (GCS). Using this very conservative external validation test based on the GCS, we find that there is little evidence for structure in the distribution function beyond the moving groups established prior to the Hipparcos mission. This is in sharp contrast with internal tests performed here and in previous analyses, which point consistently to maximal structure in the velocity distribution. We quantify the information content of the radial velocity measurements and find that the mean amount of new information gained from a radial velocity measurement of a single star is significant. This argues for complementary radial velocity surveys to upcoming astrometric surveys

    Influence of the Seasonal Thermocline on the Vertical Distribution of Larval Fish Assemblages Associated with Atlantic Bluefin Tuna Spawning Grounds

    Temperature is often an important variable influencing the vertical position of fish larvae in the water column. The same species may show different vertical distributions in areas with a strong near-surface seasonal thermocline compared to isothermal near-surface regions. In areas with a strong surface thermocline, tuna larvae show a significant preference for the near-surface warmer layers. Little is known regarding larval tuna vertical distribution in isothermal waters and on the vertical distribution of the associated larval fish assemblages. We conducted vertical stratified sampling using the same methodology and fishing device (MOCNESS) in the two major spawning areas of Atlantic bluefin tuna (BFT): western Mediterranean Sea (MED), characterized by a surface thermocline, and the Gulf of Mexico (GOM) which lacks thermal stratification. Tuna larvae occupied the upper 30 m in both areas, but the average larval depth distribution was consistently deeper in the GOM. In the MED, vertical distribution of larval fish assemblages was explained by temperature, and species such as BFT, Thunnus alalunga, and Ceratoscopelus maderensis, among others, coexist above the thermocline and are separated from species such as Cyclothone braueri and Hygophum spp. (found below the thermocline). In the GOM, the environmental correlates of the vertical distribution of the larvae were salinity and fluorescence. Mesopelagic taxa such as Ceratoscopelus spp. and Cyclothone spp., among others, had a shallower average distribution than Lampanyctus spp., Hygophum spp., and Myctophum spp.Versión del edito

    Evaluation of neuroendocrine markers in renal cell carcinoma

    <p>Abstract</p> <p>Background</p> <p>The purpose of the study was to examine serotonin, CD56, neurone-specific enolase (NSE), chromogranin A and synaptophysin by immunohistochemistry in renal cell carcinomas (RCCs) with special emphasis on patient outcome.</p> <p>Methods</p> <p>We studied 152 patients with primary RCCs who underwent surgery for the removal of kidney tumours between 1990 and 1999. The mean follow-up was 90 months. The expression of neuroendocrine (NE) markers was determined by immunohistochemical staining using commercially available monoclonal antibodies. Results were correlated with patient age, clinical stage, Fuhrman grade and patient outcome.</p> <p>Results</p> <p>Eight percent of tumours were positive for serotonin, 18% for CD56 and 48% for NSE. Chromogranin A immunostaining was negative and only 1% of the tumours were synaptophysin immunopositive. The NSE immunopositivity was more common in clear cell RCCs than in other subtypes (<it>p </it>= 0.01). The other NE markers did not show any association with the histological subtype. Tumours with an immunopositivity for serotonin had a longer RCC-specific survival and tumours with an immunopositivity for CD56 and NSE had a shorter RCC-specific survival but the difference was not significant. There was no relationship between stage or Fuhrman grade and immunoreactivity for serotonin, CD56 and NSE.</p> <p>Conclusions</p> <p>Serotonin, CD56 and NSE but not synaptophysin and chromogranin A are expressed in RCCs. However, the prognostic potential of these markers remains obscure.</p

    Autocrine Prostaglandin E2 Signaling Promotes Tumor Cell Survival and Proliferation in Childhood Neuroblastoma

    Background: Prostaglandin E2 (PGE2) is an important mediator in tumor-promoting inflammation. High expression of cyclooxygenase-2 (COX-2) has been detected in the embryonic childhood tumor neuroblastoma, and treatment with COX inhibitors significantly reduces tumor growth. Here, we have investigated the significance of a high COX-2 expression in neuroblastoma by analysis of PGE2 production, the expression pattern and localization of PGE2 receptors and intracellular signal transduction pathways activated by PGE2. Principal Findings: A high expression of the PGE2 receptors, EP1, EP2, EP3 and EP4 in primary neuroblastomas, independent of biological and clinical characteristics, was detected using immunohistochemistry. In addition, mRNA and protein corresponding to each of the receptors were detected in neuroblastoma cell lines. Immunofluorescent staining revealed localization of the receptors to the cellular membrane, in the cytoplasm, and in the nuclear compartment. Neuroblastoma cells produced PGE2 and stimulation of serum-starved neuroblastoma cells with PGE2 increased the intracellular concentration of calcium and cyclic AMP with subsequent phosphorylation of Akt. Addition of 16,16-dimethyl PGE 2 (dmPGE2) increased cell viability in a time, dose- and cell line-dependent manner. Treatment of neuroblastoma cells with a COX-2 inhibitor resulted in a diminished cell growth and viability that was reversed by the addition of dmPGE2. Similarly, PGE 2 receptor antagonists caused a decrease in neuroblastoma cell viability in a dose-dependent manner

    Fruit and vegetable consumption and lung cancer risk: updated information from the European Prospective Investigation into Cancer and Nutrition (EPIC)

    The association of fruit and vegetable consumption and lung cancer incidence was evaluated using the most recent data from the European Prospective Investigation into Cancer and Nutrition (EPIC), applying a refined statistical approach (calibration) to account for measurement error potentially introduced by using food frequency questionnaire data. Between 1992 and 2000, detailed information on diet and life-style of 478,590 individuals participating in EPIC was collected. During a median follow-up of 6.4 years, 1,126 lung cancer cases were observed. Multivariate Cox proportional hazard models were applied for statistical evaluation. In the whole study population, fruit consumption was significantly inversely associated with lung cancer risk while no association was found for vegetable consumption. In current smokers, however, lung cancer risk significantly decreased with higher vegetable consumption; this association became more pronounced after calibration, the hazard ratio (HR) being 0.78 (95% CI 0.620.98) per 100 g increase in daily vegetable consumption. In comparison, the HR per 100 g fruit was 0.92 (0.85-0.99) in the entire cohort and 0.90 (0.81-0.99) in smokers. Exclusion of cases diagnosed during the first 2 years of follow-up strengthened these associations, the HR being 0.71 (0.55-0.94) for vegetables (smokers) and 0.86 (0.78-0.95) for fruit (entire cohort). Cancer incidence decreased with higher consumption of apples and pears (entire cohort) as well as root vegetables (smokers). In addition to an overall inverse association with fruit intake, the results of this evaluation add evidence for a significant inverse association of vegetable consumption and lung cancer incidence in smokers.
