    Measuring Dose and Teaching Moment Cues in Speech Sound Disorder

    Speech sound disorder (SSD) occurs when children have difficulties saying sounds appropriately and need specialized instruction by a speech-language pathologist (SLP) to be understood. The best treatment approach is not always known to SLPs due to the individual needs of a client and the effects of different elements within interventions. By researching teaching strategies and dose in therapy, we can explore procedures that result in better outcomes and speed the learning process (Baker et al., 2018). Thus, the purpose of this research is to describe and quantify the elements that occurred within treatment of SSD to answer: How can we measure (a) dose and (b) modalities of cues in speech therapy? The participants included two boys aged 6;2 and 3;8 with SSD who were observed via video recording during speech sound treatment. A coding scheme was developed to track dosage and the different types of cues used in each therapy session; both were counted in one-minute increments. Quantifying dosage consisted of tallying the prompts given by the clinician, the client\u27s response, and whether the client responded correctly or incorrectly. Documenting modalities of cues consisted of tallying multiple forms of cues given by the clinician: pause, verbal model, pointing, visual cue, prolongation/segmentation, tactile, and instruction. The results for dosage and modalities of cues will be presented across participants and sessions, and the clinical implications will be discussed. This research is clinically significant because increasing our understanding of treatment elements that occur within sessions may inform our understanding of treatment outcomes and help speech-language pathologists design more effective therapies

    Athletes intending to use sports supplements are more likely to respond to a placebo

    Purpose: We investigated associations between athletes’ use of sport supplements and their responsiveness to placebo and nocebo interventions. Methods: Participants (n=627) reported their intention to use, and actual use of, sport supplements. They then completed a 5x20m repeat sprint protocol in the baseline condition, prior to being randomized to one of three treatments. Participants in the positive-belief treatment were administered an inert capsule described as a potent supplement which would improve sprint performance. Participants in the negative-belief treatment were administered an inert capsule described as a potent supplement which would negatively affect sprint performance. Participants in the control treatment received neither instruction nor capsule. 20 minutes following baseline trials, all participants completed the same repeat sprint protocol in the experimental condition. Results: Compared to controls, no mean differences in performance were observed between baseline and experimental conditions for the positive-belief treatment (-0.07 ± 0.27%, d=0.02), but mean differences were observed for the negative-belief treatment (-0.92 ± 0.31%, d=0.32), suggesting a moderate nocebo effect. In the positive-belief treatment however, a relationship between intention to use supplements and performance was observed. Performance worsened by -1.10% ± 0.30% compared to baseline for participants not intending to use supplements, worsened by -0.64 ± 0.43% among those undecided about supplement use, but improved by 0.19 ± 0.24% among those participants intending to use supplements. Conclusion: Information about a harmful supplement worsened repeat sprint performance (a mean nocebo effect), whereas information about a beneficial supplement did not improve performance (no mean placebo effect was observed). However, participants’ intention to use sport supplements influenced the direction and magnitude of subsequent placebo responses, with participants intending to use supplements more likely to respond to the positive intervention

    Critical role of a sheath phosphorylation site on the assembly and function of an atypical type VI secretion system

    The bacterial pathogen Francisella tularensis possesses a non-canonical type VI secretion system (T6SS) that is required for phagosomal escape in infected macrophages. KCl stimulation has been previously used to trigger assembly and secretion of the T6SS in culture. By differential proteomics, we found here that the amounts of the T6SS proteins remained unchanged upon KCl stimulation, suggesting involvement of post-translational modifications in T6SS assembly. A phosphoproteomic analysis indeed identified a unique phosphorylation site on IglB, a key component of the T6SS sheath. Substitutions of Y139 with alanine or phosphomimetics prevented T6SS formation and abolished phagosomal escape whereas substitution with phenylalanine delayed but did not abolish phagosomal escape in J774-1 macrophages. Altogether our data demonstrated that the Y139 site of IglB plays a critical role in T6SS biogenesis, suggesting that sheath phosphorylation could participate to T6SS dynamics

    The Long-Baseline Neutrino Experiment: Exploring Fundamental Symmetries of the Universe

    The preponderance of matter over antimatter in the early Universe, the dynamics of the supernova bursts that produced the heavy elements necessary for life and whether protons eventually decay --- these mysteries at the forefront of particle physics and astrophysics are key to understanding the early evolution of our Universe, its current state and its eventual fate. The Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed plan for a world-class experiment dedicated to addressing these questions. LBNE is conceived around three central components: (1) a new, high-intensity neutrino source generated from a megawatt-class proton accelerator at Fermi National Accelerator Laboratory, (2) a near neutrino detector just downstream of the source, and (3) a massive liquid argon time-projection chamber deployed as a far detector deep underground at the Sanford Underground Research Facility. This facility, located at the site of the former Homestake Mine in Lead, South Dakota, is approximately 1,300 km from the neutrino source at Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino charge-parity symmetry violation and mass ordering effects. This ambitious yet cost-effective design incorporates scalability and flexibility and can accommodate a variety of upgrades and contributions. With its exceptional combination of experimental configuration, technical capabilities, and potential for transformative discoveries, LBNE promises to be a vital facility for the field of particle physics worldwide, providing physicists from around the globe with opportunities to collaborate in a twenty to thirty year program of exciting science.     Gameplay as a source of intrinsic motivation in a randomized controlled trial of auditory training

    Background: Previous studies of frequency discrimination training (FDT) for tinnitus used repetitive task-based training programmes relying on extrinsic factors to motivate participation. Studies reported limited improvement in tinnitus symptoms. Purpose: To evaluate FDT exploiting intrinsic motivations by integrating training with computer-gameplay. Methods: Sixty participants were randomly assigned to train on a conventional taskbased training, or one of two interactive game-based training platforms over six weeks. Outcomes included assessment of motivation, tinnitus handicap, and performance on tests of attention. Results: Participants reported greater intrinsic motivation to train on the interactive game-based platforms, yet compliance of all three groups was similar (~70%) and changes in self-reported tinnitus severity were not significant. There was no difference between groups in terms of change in tinnitus severity or performance on measures of attention. Conclusion: FDT can be integrated within an intrinsically motivating game. Whilst this may improve participant experience, in this instance it did not translate to additional compliance or therapeutic benefit
