    The role of Si impurities in the transient dopant segregation and precipitation in yttrium-doped alumina

    Y-doped alumina was sintered at 1500 degrees C for 10 h under ultra-clean experimental conditions without experiencing any abnormal grain growth. The yttrium was fairly homogeneously distributed at the grain boundaries, with a mean value of (Gamma) over bar (Y) = 5.5 at nm(-2). The Y-Al-O precipitates in the clean, Y2O3-doped alumina specimen were the YAP (YAlO3) phase, whereas only the YAG (Y3Al5O12) phase was present in the Y2O3-doped alumina samples contaminated with SiO2. The excess concentrations of Y and Si atoms at the grain boundaries that, at the same time, provoke the formation of structurally complex YAG precipitates and abnormal grain growth were both estimated to be at 4-5 at nm(-2). The compositions of the triple point pocket phases found in the region of the exaggeratedly grown alumina grains indicate the presence of alumino-silicate bulk liquids at the sintering temperature

    Extracellular riboflavin induces anaerobic biofilm formation in Shewanella oneidensis

    Background Some microorganisms can respire with extracellular electron acceptors using an extended electron transport chain to the cell surface. This process can be applied in bioelectrochemical systems in which the organisms produce an electrical current by respiring with an anode as electron acceptor. These organisms apply flavin molecules as cofactors to facilitate one-electron transfer catalyzed by the terminal reductases and in some cases as endogenous electron shuttles. Results In the model organism Shewanella oneidensis, riboflavin production and excretion trigger a specific biofilm formation response that is initiated at a specific threshold concentration, similar to canonical quorum-sensing molecules. Riboflavin-mediated messaging is based on the overexpression of the gene encoding the putrescine decarboxylase speC which leads to posttranscriptional overproduction of proteins involved in biofilm formation. Using a model of growth-dependent riboflavin production under batch and biofilm growth conditions, the number of cells necessary to produce the threshold concentration per time was deduced. Furthermore, our results indicate that specific retention of riboflavin in the biofilm matrix leads to localized concentrations, which by far exceed the necessary threshold value. Conclusion This study describes a new quorum-sensing mechanism in S. oneidensis. Biofilm formation of S. oneidensis is induced by low concentrations of riboflavin resulting in an upregulation of the ornithine-decarboxylase speC. The results can be applied for the development of strains catalyzing increased current densities in bioelectrochemical systems

    Efficient Bioelectrochemical Conversion of Industrial Wastewater by Specific Strain Isolation and Community Adaptation

    The aim of this study was the development of a specifically adapted microbial community for the removal of organic carbon from an industrial wastewater using a bioelectrochemical system. In a first step, ferric iron reducing microorganisms were isolated from the examined industrial wastewater. In a second step, it was tested to what extent these isolates or a cocultivation of the isolates with the exoelectrogenic model organism Geobacter sulfurreducens (G. sulfurreducens) were able to eliminate organic carbon from the wastewater. To establish a stable biofilm on the anode and to analyze the performance of the system, the experiments were conducted first under batch-mode conditions for 21 days. Since the removal of organic carbon was relatively low in the batch system, a similar experiment was conducted under continuous-mode conditions for 65 days, including a slow transition from synthetic medium to industrial wastewater as carbon and electron source and variations in the flow rate of the medium. The overall performance of the system was strongly increased in the continuous- compared to the batch-mode reactor and the highest average current density (1,368 mA/m2) and Coulombic efficiency (54.9%) was measured in the continuous-mode reactor inoculated with the coculture consisting of the new isolates and G. sulfurreducens. The equivalently inoculated batch-mode system produced only 82-fold lower current densities, which were accompanied by 42-fold lower Coulombic efficiencies

    Soluble versions of outer membrane cytochromes function as exporters for heterologously produced cargo proteins

    This study reveals that it is possible to secrete truncated versions of outer membrane cytochromes into the culture supernatant and that these proteins can provide a basis for the export of heterologously produced proteins. Different soluble and truncated versions of the outer membrane cytochrome MtrF were analyzed for their suitability to be secreted. A protein version with a very short truncation of the N-terminus to remove the recognition sequence for the addition of a lipid anchor is secreted efficiently to the culture supernatant, and moreover this protein could be further truncated by a deletion of 160 amino acid and still is detectable in the supernatant. By coupling a cellulase to this soluble outer membrane cytochrome, the export efficiency was measured by means of relative cellulase activity. We conclude that outer membrane cytochromes of S. oneidensis can be applied as transporters for the export of target proteins into the medium using the type II secretion pathway

    Maxwell consideration of polaritonic quasi-particle Hamiltonians in multi-level systems

    We address the problem of the correct description of light-matter coupling for excitons and cavity photons in the case of systems with multiple photon modes or excitons, respectively. In the literature, two different approaches for the phenomenological coupling Hamiltonian can be found: Either one single Hamiltonian with a basis whose dimension equals the sum of photonic modes and excitonic resonances is used. Or a set of independent Hamiltonians, one for each photon mode, is chosen. Both are usually used equivalently for the same kind of multi-photonic systems which cannot be correct. However, identifying the suitable Hamiltonian is difficult when modeling experimental data. By means of numerical transfer matrix calculations, we demonstrate the scope of application of each approach: The first one holds only for the coupling of a single photon state to several excitons, while in the case of multiple photon modes, separate Hamiltonians must be used for each photon mode

    Angle-dependence of the Hall effect in HgBa2CaCu2O6 thin films

    Superconducting compounds of the family Hg-Ba-Ca-Cu-O have been the subject of intense study since the current record-holder for the highest critical temperature of a superconductor belongs to this class of materials. Thin films of the compound with two adjacent copper-oxide layers and a critical temperature of about 120 K were prepared by a two-step process that consists of the pulsed-laser deposition of precursor films and the subsequent annealing in mercury-vapor atmosphere. Like some other high-temperature superconductors, Hg-Ba-Ca-Cu-O exhibits a specific anomaly of the Hall effect, a double-sign change of the Hall coefficient close to the superconducting transition. We have investigated this phenomenon by measurements of the Hall effect at different angles between the magnetic field direction and the crystallographic c-axis. The results concerning the upper part of the transition, where the first sign change occurs, are discussed in terms of the renormalized fluctuation model for the Hall conductivity, adapted through the field rescaling procedure in order to take into account the arbitrary orientation of the magnetic field.Comment: to be published in Phys.     Feasibility study for the development of plutonium reference materials for age dating in nuclear forensics

    Isotopic reference materials certified for the age of nuclear material (uranium, plutonium) are needed in the fields of nuclear forensics and environmental measurements. Therefore a feasibility study for the development of plutonium reference materials for age dating has been started recently at the Institute for Reference Materials and Measurements (EC-JRC-IRMM). The "age" of the material is defined as the time that has passed since the last chemical separation of the mother and daughter isotopes (e.g. 241Pu and 241Am). Assuming that the separation has been complete and all the daughter isotopes have been removed from the original material during this last separation, the age of the material can be determined by measuring the ratio of daughter and mother radio-nuclides, e.g. 241Am/241Pu. At a given time after the last separation and depending on the half lives of the radio-nuclides involved, a certain amount of the daughter radionuclide(s) will be present. For the determination of the unknown age of a material different "clocks" can be used; "clocks" are pairs of mother and daughter radio-nuclides, such as 241Am/241Pu, 238Pu/234U, 239Pu/235U, 240Pu/236U, and possibly 242Pu/238U. For the age estimation of a real sample, such as material seized in nuclear forensics investigations or dust samples in environmental measurements, it is advisable to use more than one clock in order to ensure the reliability of the results and to exclude the possibility that the sample under question is a mixture of two or more materials. Consequently, a future reference material certified for separation date should ideally be certified for more than one "clock" or several reference materials for different "clocks" should be developed. The first step of this study is to verify the known separation dates of different plutonium materials of different ages and isotopic compositions by measuring the mother (238Pu, 239Pu, 240Pu, 241Pu, 242Pu) and daughter (234U, 235U, 236U, 238U and 241Am) isotope abundances and to estimate the achievable uncertainties of the calculated ages. Thermal ionisation mass spectrometry (TIMS) is applied for the measurement of the plutonium and uranium isotope ratios after chemical separation of the plutonium and uranium fractions from the sample matrix, while 241Am is measured by gamma-ray spectrometry.     Galectin-4 Controls Intestinal Inflammation by Selective Regulation of Peripheral and Mucosal T Cell Apoptosis and Cell Cycle

    Galectin-4 is a carbohydrate-binding protein belonging to the galectin family. Here we provide novel evidence that galectin-4 is selectively expressed and secreted by intestinal epithelial cells and binds potently to activated peripheral and mucosal lamina propria T-cells at the CD3 epitope. The carbohydrate-dependent binding of galectin-4 at the CD3 epitope is fully functional and inhibited T cell activation, cycling and expansion. Galectin-4 induced apoptosis of activated peripheral and mucosal lamina propria T cells via calpain-, but not caspase-dependent, pathways. Providing further evidence for its important role in regulating T cell function, galectin-4 blockade by antisense oligonucleotides reduced TNF-alpha inhibitor induced T cell death. Furthermore, in T cells, galectin-4 reduced pro-inflammatory cytokine secretion including IL-17. In a model of experimental colitis, galectin-4 ameliorated mucosal inflammation, induced apoptosis of mucosal T-cells and decreased the secretion of pro-inflammatory cytokines. Our results show that galectin-4 plays a unique role in the intestine and assign a novel role of this protein in controlling intestinal inflammation by a selective induction of T cell apoptosis and cell cycle restriction. Conclusively, after defining its biological role, we propose Galectin-4 is a novel anti-inflammatory agent that could be therapeutically effective in diseases with a disturbed T cell expansion and apoptosis such as inflammatory bowel disease
