    Model-based image analysis of a tethered Brownian fibre for shear stress sensing

    The measurement of fluid dynamic shear stress acting on a biologically relevant surface is a challenging problem, particularly in the complex environment of, for example, the vasculature. While an experimental method for the direct detection of wall shear stress via the imaging of a synthetic biology nanorod has recently been developed, the data interpretation so far has been limited to phenomeno-logical random walk modelling, small-angle approximation, and image analysis techniques which do not take into account the production of an image from a three-dimensional subject. In this report, we develop a mathematical and statistical framework to estimate shear stress from rapid imaging sequences based firstly on stochastic modelling of the dynamics of a tethered Brownian fibre in shear flow, and secondly on a novel model-based image analysis, which reconstructs fibre positions by solving the inverse problem of image formation. This framework is tested on experimental data, providing the first mechanistically rational analysis of the novel assay. What follows further develops the established theory for an untethered particle in a semi-dilute suspension, which is of relevance to, for example, the study of Brownian nanowires without flow, and presents new ideas in the field of multi-disciplinary image analysis

    IL-21 receptor expression in human tendinopathy

    The pathogenetic mechanisms underlying tendinopathy remain unclear, with much debate as to whether inflammation or degradation has the prominent role. Increasing evidence points toward and early inflammatory infiltrate and associated inflammatory cytokine production in human and animal models of tendon disease. The IL-21/IL-21R axis is a proinflammatory cytokine complex that has been associated with chronic inflammatory diseases including rheumatoid arthritis and inflammatory bowel disease. This project aimed to investigate the role and expression of the cytokine/receptor pair IL-21/IL-21R in human tendinopathy. We found significantly elevated expression of IL-21 receptor message and protein in human tendon samples but found no convincing evidence of the presence of IL-21 at message or protein level. The level of expression of IL-21R message/protein in human tenocytes was significantly up regulated by proinflammatory cytokines (TNFα/IL-1β) in vitro. These findings demonstrate that IL-21R is present in early human tendinopathy mainly expressed by tenocytes and macrophages. Despite a lack of IL-21 expression these data again suggest that early tendinopathy has an inflammatory/cytokine phenotype, which may provide novel translational targets in the treatment of tendinopathy

    Identifying people at higher risk of melanoma across the U.K.: a primary-care-based electronic survey

    BACKGROUND: Melanoma incidence is rising rapidly worldwide among white populations. Defining higher-risk populations using risk prediction models may help targeted screening and early detection approaches. OBJECTIVES: To assess the feasibility of identifying people at higher risk of melanoma using the Williams self-assessed clinical risk estimation model in U.K. primary care. METHODS: We recruited participants from the waiting rooms of 22 general practices covering a total population of > 240 000 in three U.K. regions: Eastern England, North East Scotland and North Wales. Participants completed an electronic questionnaire using tablet computers. The main outcome was the mean melanoma risk score using the Williams melanoma risk model. RESULTS: Of 9004 people approached, 7742 (86%) completed the electronic questionnaire. The mean melanoma risk score for the 7566 eligible participants was 17·15 ± 8·51, with small regional differences [lower in England compared with Scotland (P = 0·001) and Wales (P < 0·001), mainly due to greater freckling and childhood sunburn among Scottish and Welsh participants]. After weighting to the age and sex distribution, different potential cut-offs would allow between 4% and 20% of the population to be identified as higher risk, and those groups would contain 30% and 60%, respectively of those likely to develop melanoma. CONCLUSIONS: Collecting data on the melanoma risk profile of the general population in U.K. primary care is both feasible and acceptable for patients in a general practice setting, and provides opportunities for new methods of real-time risk assessment and risk stratified cancer interventions

    Deep learning optimized single-pixel LiDAR

    Interest in autonomous transport has led to a demand for 3D imaging technologies capable of resolving fine details at long range. Light detection and ranging (LiDAR) systems have become a key technology in this area, with depth information typically gained through time-of-flight photon-counting measurements of a scanned laser spot. Single-pixel imaging methods offer an alternative approach to spot-scanning, which allows a choice of sampling basis. In this work, we present a prototype LiDAR system, which compressively samples the scene using a deep learning optimized sampling basis and reconstruction algorithms. We demonstrate that this approach improves scene reconstruction quality compared to an orthogonal sampling method, with reflectivity and depth accuracy improvements of 57% and 16%, respectively, for one frame per second acquisition rates. This method may pave the way for improved scan-free LiDAR systems for driverless cars and for fully optimized sampling to decision-making pipelines

    Fatigue in Medical Residents Leads to Reactivation of Herpes Virus Latency

    The main objective of this study was to detect fatigue-induced clinical symptoms of immune suppression in medical residents. Samples were collected from the subjects at rest, following the first night (low-stress), and the last night (high-stress) of night float. Computerized reaction tests, Epworth Sleepiness Scale, and Wellness Profile questionnaires were used to quantify fatigue level. DNA of human herpes viruses HSV-1, VZV, EBV, as well as cortisol and melatonin concentrations, were measured in saliva. Residents at the high-stress interval reported being sleepier compared to the rest interval. EBV DNA level increased significantly at both stress intervals, while VZV DNA level increased only at low-stress. DNA levels of HSV-1 decreased at low-stress but increased at high-stress. Combined assessment of the viral DNA showed significant effect of stress on herpes virus reactivation at both stress intervals. Cortisol concentrations at both stress intervals were significantly higher than those at rest

    The effects of barbell load on countermovement vertical jump power and net impulse

    The aim of this study was to examine the effects of barbell load on countermovement vertical jump (CMJ) power and net impulse within a theoretically valid framework, cognisant of the underpinning force, temporal, and spatial components. Twenty-four resistance-trained rugby union athletes (average ± SD: age: 23.1 ± 3.4 yrs; height: 1.83 ± 0.05 m; body mass: 91.3 ± 10.5 kg) performed maximal CMJ under five experimental conditions in a randomised, counterbalanced order: unloaded, and with additional loads of 25, 50, 75 and 100% of body mass (BM). Peak power and average power were maximised during the unloaded condition, both decreasing significantly (p < 0.05) as load increased. Net impulse was maximised with 75% of BM, which was significantly greater (p < 0.05) than the unloaded and 100% of BM conditions. Net mean force and mean velocity were maximised during the unloaded condition and decreased significantly (p < 0.05) as load increased, whereas phase duration increased significantly (p < 0.05) as load increased. As such, the interaction between barbell load and the underpinning force, time and displacement components should be considered by strength and conditioning coaches when prescribing barbell loads

    Testing Inflation with Large Scale Structure: Connecting Hopes with Reality

    The statistics of primordial curvature fluctuations are our window into the period of inflation, where these fluctuations were generated. To date, the cosmic microwave background has been the dominant source of information about these perturbations. Large scale structure is however from where drastic improvements should originate. In this paper, we explain the theoretical motivations for pursuing such measurements and the challenges that lie ahead. In particular, we discuss and identify theoretical targets regarding the measurement of primordial non-Gaussianity. We argue that when quantified in terms of the local (equilateral) template amplitude fNLlocf_{\rm NL}^{\rm loc} (fNLeqf_{\rm NL}^{\rm eq}), natural target levels of sensitivity are ΔfNLloc,eq.1\Delta f_{\rm NL}^{\rm loc, eq.} \simeq 1. We highlight that such levels are within reach of future surveys by measuring 2-, 3- and 4-point statistics of the galaxy spatial distribution.     A Mid-Infrared Census of Star Formation Activity in Bolocam Galactic Plane Survey Sources

    We present the results of a search for mid-infrared signs of star formation activity in the 1.1 mm sources in the Bolocam Galactic Plane Survey (BGPS). We have correlated the BGPS catalog with available mid-IR Galactic plane catalogs based on the Spitzer Space Telescope GLIMPSE legacy survey and the Midcourse Space Experiment (MSX) Galactic plane survey. We find that 44% (3,712 of 8,358) of the BGPS sources contain at least one mid-IR source, including 2,457 of 5,067 (49%) within the area where all surveys overlap (10 deg < l < 65 deg). Accounting for chance alignments between the BGPS and mid-IR sources, we conservatively estimate that 20% of the BPGS sources within the area where all surveys overlap show signs of active star formation. We separate the BGPS sources into four groups based on their probability of star formation activity. Extended Green Objects (EGOs) and Red MSX Sources (RMS) make up the highest probability group, while the lowest probability group is comprised of "starless" BGPS sources which were not matched to any mid-IR sources. The mean 1.1 mm flux of each group increases with increasing probability of active star formation. We also find that the "starless" BGPS sources are the most compact, while the sources with the highest probability of star formation activity are on average more extended with large skirts of emission. A subsample of 280 BGPS sources with known distances demonstrates that mass and mean H_2 column density also increase with probability of star formation activity.Comment: 20 pages, 12 figures, 3 tables.     Ovarian cancer symptom awareness and anticipated delayed presentation in a population sample

    Background: While ovarian cancer is recognised as having identifiable early symptoms, understanding of the key determinants of symptom awareness and early presentation is limited. A population-based survey of ovarian cancer awareness and anticipated delayed presentation with symptoms was conducted as part of the International Cancer Benchmarking Partnership (ICBP). Methods: Women aged over 50 years were recruited using random probability sampling (n = 1043). Computer-assisted telephone interviews were used to administer measures including ovarian cancer symptom recognition, anticipated time to presentation with ovarian symptoms, health beliefs (perceived risk, perceived benefits/barriers to early presentation, confidence in symptom detection, ovarian cancer worry), and demographic variables. Logistic regression analysis was used to identify the contribution of independent variables to anticipated presentation (categorised as < 3 weeks or ≥ 3 weeks). Results: The most well-recognised symptoms of ovarian cancer were post-menopausal bleeding (87.4%), and persistent pelvic (79.0%) and abdominal (85.0%) pain. Symptoms associated with eating difficulties and changes in bladder/bowel habits were recognised by less than half the sample. Lower symptom awareness was significantly associated with older age (p ≤ 0.001), being single (p ≤ 0.001), lower education (p ≤ 0.01), and lack of personal experience of ovarian cancer (p ≤ 0.01). The odds of anticipating a delay in time to presentation of ≥ 3 weeks were significantly increased in women educated to degree level (OR = 2.64, 95% CI 1.61 – 4.33, p ≤ 0.001), women who reported more practical barriers (OR = 1.60, 95% CI 1.34 – 1.91, p ≤ 0.001) and more emotional barriers (OR = 1.21, 95% CI 1.06 – 1.40, p ≤ 0.01), and those less confident in symptom detection (OR = 0.56, 95% CI 0.42 – 0.73, p ≤ 0.001), but not in those who reported lower symptom awareness (OR = 0.99, 95% CI 0.91 – 1.07, p = 0.74). Conclusions: Many symptoms of ovarian cancer are not well-recognised by women in the general population. Evidence-based interventions are needed not only to improve public awareness but also to overcome the barriers to recognising and acting on ovarian symptoms, if delays in presentation are to be minimised
