    A Bioeconomic Model for Management of Orange Roughy Stocks

    The paper reports the results of a bioeconomic analysis of the exploitation of a recently discovered orange roughy stock located off Tasmania. The parameters of the model are based on the experience derived from the orange roughy fisheries in New Zealand where stocks have been heavily exploited. The model is used to predict the open-access equilibrium stock, and to calculate the stock which maximizes the net present value and the stock level consistent with the F,,, Rule. Assuming a linear approach path, the net present value of the fishery at each of these stocks is calculated. The results are used to estimate the benefit of management and the cost of a conservative stock policy.     Alanine-scanning mutagenesis of protein mannosyl-transferase from Streptomyces coelicolor reveals strong activity-stability correlation

    In Actinobacteria, protein O-mannosyl transferase (Pmt)-mediated protein O-glycosylation has an important role in cell envelope physiology. In S. coelicolor, defective Pmt leads to increased susceptibility to cell wall-targeting antibiotics, including vancomycin and β-lactams, and resistance to phage ϕC31. The aim of this study was to gain a deeper understanding of the structure and function of S. coelicolor Pmt. Sequence alignments and structural bioinformatics were used to identify target sites for an alanine-scanning mutagenesis study. Mutant alleles were introduced into pmt-deficient S. coelicolor strains using an integrative plasmid and scored for their ability to complement phage resistance and antibiotic hypersusceptibility phenotypes. Twenty-three highly conserved Pmt residues were each substituted for alanine. Six mutant alleles failed to complement the pmt▬ strains in either assay. Mapping the six corresponding residues onto a homology model of the three-dimensional structure of Pmt, indicated that five are positioned close to the predicted catalytic DE motif. Further mutagenesis to produce more conservative substitutions at these six residues produced Pmts that invariably failed to complement the DT1025 pmt▬ strain, indicating that strict residue conservation was necessary to preserve function. Cell fractionation and Western blotting of strains with the non-complementing pmt alleles revealed undetectable levels of the enzyme in either the membrane fractions or whole cell lysates. Meanwhile for all of the strains that complemented the antibiotic hypersusceptibility and phage resistance phenotypes, Pmt was readily detected in the membrane fraction. These data indicate a tight correlation between the activity of Pmt and its stability or ability to localize to the membrane

    Jellyfication of marine ecosystems as a likely consequence of overfishing small pelagic fishes: Lessons from the Benguela

    Changes in two contrasting ecosystems of the Benguela upwelling region, one dominated at mid-trophic level by jellyfishes (Namibia, northern Benguela ecosystem, where small pelagic fish abundance has been severely depleted) and one still dominated by small pelagic fishes (South Africa, southern Benguela) were compared in an effort to determine ecosystem trajectories under different exploitation regimes. The role of small pelagic fishes (clupeoids) was highlighted in the context of their importance in maintaining interactions in marine ecosystems. In particular, we examined trophic cascades and possible irreversible changes that promote the proliferation of jellyfishes in marine systems. We found that the presence of large populations of small pelagic fishes has a fundamental role in preserving beneficial trophic interactions in these marine ecosystems. The implications of trophic cascades, such as those observed in the northern Benguela, for ecosystem-based management were apparent. In addition, this comparison provides contrasting case studies to inform the development of management scenarios that avoid ecosystem shifts that affect predators and reduce the value of fisheries production.Web of Scienc

    Risk of major cardiovascular events in patients with psoriasis receiving biologic therapies: a prospective cohort study

    Background: The cardiovascular safety profile of biologic therapies used for psoriasis is unclear. Objectives: To compare the risk of major cardiovascular events (CVEs; acute coronary syndrome, unstable angina, myocardial infarction and stroke) in patients with chronic plaque psoriasis treated with adalimumab, etanercept or ustekinumab in a large prospective cohort. Methods: Prospective cohort study examining the comparative risk of major CVEs was conducted using the British Association of Dermatologists Biologics and Immunomodulators Register. The main analysis compared adults with chronic plaque psoriasis receiving ustekinumab with tumour necrosis‐α inhibitors (TNFi: etanercept and adalimumab), whilst the secondary analyses compared ustekinumab, etanercept or methotrexate against adalimumab. Hazard ratios (HRs) with 95% confidence intervals (CIs) were calculated using overlap weights by propensity score to balance baseline covariates among comparison groups. Results: We included 5468 biologic‐naïve patients subsequently exposed (951 ustekinumab; 1313 etanercept; and 3204 adalimumab) in the main analysis. The secondary analyses also included 2189 patients receiving methotrexate. The median (p25–p75) follow‐up times for patients using ustekinumab, TNFi, adalimumab, etanercept and methotrexate were as follows: 2.01 (1.16–3.21), 1.93 (1.05–3.34), 1.94 (1.09–3.32), 1.92 (0.93–3.45) and 1.43 (0.84–2.53) years, respectively. Ustekinumab, TNFi, adalimumab, etanercept and methotrexate groups had 7, 29, 23, 6 and 9 patients experiencing major CVEs, respectively. No differences in the risk of major CVEs were observed between biologic therapies [adjusted HR for ustekinumab vs. TNFi: 0.96 (95% CI 0.41–2.22); ustekinumab vs. adalimumab: 0.81 (0.30–2.17); etanercept vs. adalimumab: 0.81 (0.28–2.30)] and methotrexate against adalimumab [1.05 (0.34–3.28)]. Conclusions: In this large prospective cohort study, we found no significant differences in the risk of major CVEs between three different biologic therapies and methotrexate. Additional studies, with longer term follow‐up, are needed to investigate the potential effects of biologic therapies on incidence of major CVEs

    Method evaluation and risk assessment: A framework for evaluating management strategies for data-limited fisheries

    Fisheries managers are in need of quantitative tools to inform decisions regarding selection of robust management practices, prioritising research gaps and stocks to focus on, particularly where there are limited resources or data. To support these decisions, the use of Management Strategy Evaluation (MSE), that is, closed loop simulation-testing of management procedures, is widely regarded as best practice. However, applying MSE is time- and computationally intensive, and requires highly skilled expertise and processes for stakeholder input and peer review. For data- and capacity-limited fisheries, MSE may be particularly challenging to implement. Yet, these are the contexts where it is most critical to test assumptions, evaluate the implications of all sources of uncertainty and identify the most informative data sources. To facilitate wider use of MSE, the Method Evaluation and Risk Assessment (MERA) framework was developed as an accessible online interface, with quick processing time, focused on generic data-limited management procedures, but allowing progression to tailored and more data-rich methods. The framework links a quantitative questionnaire and data input standard to a flexible operating model with optional customisation via command line access to the back-end open-source R libraries. Here, we illustrate a case study application of MERA for the bocinegro (Pagrus pagrus, Sparidae) fishery in the Gulf of Cadiz, where in conjunction with fishery stakeholders, a custom management procedure was developed and tested and key research gaps and data collection priorities were identified.     Development and validation of a multivariable risk prediction model for serious infection in patients with psoriasis receiving systemic therapy

    BACKGROUND: Patients with psoriasis are often concerned about the risk of serious infection associated with systemic psoriasis treatments. OBJECTIVES: To develop and externally validate a prediction model for serious infection in patients with psoriasis within 1 year of starting systemic therapies. METHODS: The risk prediction model was developed using the British Association of Dermatologists Biologic Interventions Register (BADBIR), and the German Psoriasis Registry PsoBest was used as the validation dataset. Model discrimination and calibration were assessed internally and externally using the C-statistic, the calibration slope and the calibration in the large. RESULTS: Overall 175 (1·7%) out of 10 033 participants from BADBIR and 41 (1·7%) out of 2423 participants from PsoBest developed a serious infection within 1 year of therapy initiation. Selected predictors in a multiple logistic regression model included nine baseline covariates, and starting infliximab was the strongest predictor. Evaluation of model performance showed a bootstrap optimism-corrected C-statistic of 0·64 [95% confidence interval (CI) 0·60-0·69], calibration in the large of 0·02 (95% CI -0·14 to 0·17) and a calibration slope of 0·88 (95% CI 0·70-1·07), while external validation performance was poor, with C-statistic 0·52 (95% CI 0·42-0·62), calibration in the large 0·06 (95% CI -0·25 to 0·37) and calibration slope 0·36 (95% CI -0·24 to 0·97). CONCLUSIONS: We present the first results of the development of a multivariable prediction model. This model may help patients and dermatologists in the U.K. and the Republic of Ireland to identify modifiable risk factors and inform therapy choice in a shared decision-making process

    Measurement of the B0-anti-B0-Oscillation Frequency with Inclusive Dilepton Events

    The B0B^0-Bˉ0\bar B^0 oscillation frequency has been measured with a sample of 23 million \B\bar B pairs collected with the BABAR detector at the PEP-II asymmetric B Factory at SLAC. In this sample, we select events in which both B mesons decay semileptonically and use the charge of the leptons to identify the flavor of each B meson. A simultaneous fit to the decay time difference distributions for opposite- and same-sign dilepton events gives Δmd=0.493±0.012(stat)±0.009(syst)\Delta m_d = 0.493 \pm 0.012{(stat)}\pm 0.009{(syst)} ps1^{-1}.Comment: 7 pages, 1 figure, submitted to Physical Review Letter

    Next-to-Leading Order QCD Analysis of Polarized Deep Inelastic Scattering Data

    We present a Next-to-Leading order perturbative QCD analysis of world data on the spin dependent structure functions g1p,g1ng_1^p, g_1^n, and g1dg_1^d, including the new experimental information on the Q2Q^2 dependence of g1ng_1^n. Careful attention is paid to the experimental and theoretical uncertainties. The data constrain the first moments of the polarized valence quark distributions, but only qualitatively constrain the polarized sea quark and gluon distributions. The NLO results are used to determine the Q2Q^2 dependence of the ratio g1/F1g_1/F_1 and evolve the experimental data to a constant Q2=5GeV2Q^2 = 5 GeV^2. We determine the first moments of the polarized structure functions of the proton and neutron and find agreement with the Bjorken sum rule.Comment: 21 pages, 4 figures; final version to be published in Phys.
