The Role of Food Parenting Skills and the Home Food Environment in Childrenâs Weight Gain and Obesity
The DONE framework: Creation, evaluation, and updating of an interdisciplinary, dynamic framework 2.0 of determinants of nutrition and eating.
The question of which factors drive human eating and nutrition is a key issue in many branches of science. We describe the creation, evaluation, and updating of an interdisciplinary, interactive, and evolving "framework 2.0" of Determinants Of Nutrition and Eating (DONE). The DONE framework was created by an interdisciplinary workgroup in a multiphase, multimethod process. Modifiability, relationship strength, and population-level effect of the determinants were rated to identify areas of priority for research and interventions. External experts positively evaluated the usefulness, comprehensiveness, and quality of the DONE framework. An approach to continue updating the framework with the help of experts was piloted. The DONE framework can be freely accessed (http://uni-konstanz.de/DONE) and used in a highly flexible manner: determinants can be sorted, filtered and visualized for both very specific research questions as well as more general queries. The dynamic nature of the framework allows it to evolve as experts can continually add new determinants and ratings. We anticipate this framework will be useful for research prioritization and intervention development
Socio-cultural determinants of physical activity across the life course: a 'Determinants of Diet and Physical Activity' (DEDIPAC) umbrella systematic literature review
Objective
Regular physical activity (PA) reduces the risk of disease and premature death. Knowing factors associated with PA might help reducing the disease and economic burden caused by low activity. Studies suggest that socio-cultural factors may affect PA, but systematic overviews of findings across the life course are scarce. This umbrella systematic literature review (SLR) summarizes and evaluates available evidence on socio-cultural determinants of PA in children, adolescents, and adults.
Methods
This manuscript was drafted following the recommendations of the âPreferred Reporting Items for Systematic reviews and Meta-Analysesâ (PRISMA) checklist. The MEDLINE, Web of Science, Scopus, and SPORTDiscus databases were searched for SLRs and meta-analyses (MAs) on observational studies published in English that assessed PA determinants between January 2004 and April 2016. The methodological quality was assessed and relevant information on socio-cultural determinants and any associations with PA was extracted. The available evidence was evaluated based on the importance of potential determinants and the strength of the evidence.
Results
Twenty SLRs and three MAs encompassing 657 eligible primary studies investigated potential socio-cultural PA determinants, with predominantly moderate methodological quality. Twenty-nine potential PA determinants were identified that were primarily assessed in children and adolescents and investigated the micro-environmental home/household level. We found probable evidence that receiving encouragement from significant others and having a companion for PA were associated with higher PA in children and adolescents, and that parental marital status (living with partner) and experiencing parental modeling were not associated with PA in children. Evidence for the other potential determinants was limited, suggestive, or non-conclusive. In adults, quantitative and conclusive data were scarce.
Conclusions
A substantial number of SLRs and MAs investigating potential socio-cultural determinants of PA were identified. Our data suggest that receiving social support from significant others may increase PA levels in children and adolescents, whereas parental marital status is not a determinant in children. Evidence for other potential determinants was limited. This was mainly due to inconsistencies in results on potential socio-cultural determinants of PA across reviews and studies
Behavioural Susceptibility Theory: Professor Jane Wardle and the Role of Appetite in Genetic Risk of Obesity
Purpose of Review: There is considerable variability in human body weight, despite the ubiquity of the 'obesogenic' environment. Human body weight has a strong genetic basis and it has been hypothesised that genetic susceptibility to the environment explains variation in human body weight, with differences in appetite being implicated as the mediating mechanism; so-called 'behavioural susceptibility theory' (BST), first described by Professor Jane Wardle. This review summarises the evidence for the role of appetite as a mediator of genetic risk of obesity. Recent Findings: Variation in appetitive traits is observable from infancy, drives early weight gain and is highly heritable in infancy and childhood. Obesity-related common genetic variants identified through genome-wide association studies show associations with appetitive traits, and appetite mediates part of the observed association between genetic risk and adiposity. Emotional over- and under-eating in early childhood are learned not inherited
Emotional overeating (EOE) has been associated with increased obesity risk, while emotional undereating (EUE) may be protective. Interestingly, EOE and EUE tend to correlate positively, but it is unclear whether they reflect different aspects of the same underlying trait, or are distinct behaviours with different aetiologies. Data were from 2054 five-year-old children from the Gemini twin birth cohort, including parental ratings of child EOE and EUE using the Child Eating Behaviour Questionnaire. Genetic and environmental influences on variation and covariation in EUE and EOE were established using a bivariate Twin Model. Variation in both behaviours was largely explained by aspects of the environment completely shared by twin pairs (EOE: Câ=â90%, 95% CI: 89%-92%; EUE: Câ=â91%, 95% CI: 90%-92%). Genetic influence was low (EOE: Aâ=â7%, 95% CI: 6%-9%; EUE: Aâ=â7%, 95% CI: 6%-9%). EOE and EUE correlated positively (râ=â0.43, pâ<â0.001), and this association was explained by common shared environmental influences (BivCâ=â45%, 95% CI: 40%-50%). Many of the shared environmental influences underlying EUE and EOE were the same (rCâ=â0.50, 95% CI: 0.44, 0.55). Childhood EOE and EUE are etiologically distinct. Using formative research to develop the healthy eating component of the CHANGE! school-based curriculum intervention
Background: Childhood obesity is a significant public health concern. Many intervention studies have attempted
to combat childhood obesity, often in the absence of formative or preparatory work. This study describes the
healthy eating component of the formative phase of the Childrenâs Health Activity and Nutrition: Get Educated!
(CHANGE!) project. The aim of the present study was to gather qualitative focus group and interview data
regarding healthy eating particularly in relation to enabling and influencing factors, barriers and knowledge in
children and adults (parents and teachers) from schools within the CHANGE! programme to provide populationspecific
evidence to inform the subsequent intervention design.
Methods: Semi-structured focus group interviews were conducted with children, parents and teachers across 11
primary schools in the Wigan borough of North West England. Sixty children (N = 24 boys), 33 parents (N = 4 male)
and 10 teachers (N = 4 male) participated in the study. Interview questions were structured around the PRECEDE
phases of the PRECEDE-PROCEED model. Interviews were transcribed verbatim and analysed using the pen-profiling
technique.
Results: The pen-profiles revealed that childrenâs knowledge of healthy eating was generally good, specifically
many children were aware that fruit and vegetable consumption was âhealthyâ (N = 46). Adultsâ knowledge was also
good, including restricting fatty foods, promoting fruit and vegetable intake, and maintaining a balanced diet. The
important role parents play in childrenâs eating behaviours and food intake was evident. The emerging themes
relating to barriers to healthy eating showed that external drivers such as advertising, the preferred sensory
experience of âunhealthyâ foods, and food being used as a reward may play a role in preventing healthy eating.
Conclusions: Data suggest that; knowledge related to diet composition was not a barrier per se to healthy eating,
and education showing how to translate knowledge into behavior or action is required. The key themes that
emerged through the focus groups and pen-profiling data analysis technique will be used to inform and tailor the
healthy eating component of the CHANGE! intervention study.

Background
Low levels of physical activity (PA) are a global concern and increasing PA engagement is becoming a priority in current public health policies. Despite the large number of studies and reviews available, the evidence regarding the behavioral determinants of PA is still inconclusive. Thus, the aim of this umbrella systematic literature review (SLR) was to summarize the evidence on the behavioral determinants of PA across the life course.
Methods
A systematic online search was conducted on MEDLINE, ISI Web of Science, Scopus, and SPORTDiscus databases. The search was limited to studies published in English from January, 2004 to April, 2016. SLRs and meta-analyses (MAs) of observational studies that investigated the behavioral determinants of PA were considered eligible. The extracted data were assessed based on the importance of the determinants, the strength of evidence, and the methodological quality. Results
Seventeen reviews on 35 behavioral determinants of PA were eligible for this umbrella SLR. Regardless of age, the most investigated determinants were those related with âscreen useâ and âsmokingâ. For youth, probable positive evidence emerged for âprevious PAâ and âindependent mobility and active transportâ among children and adolescents. For the adult population, âtransition to universityâ and âpregnancy/having a childâ showed probable negative associations.
Conclusions
Although the majority of the evidence was limited and most of the determinants were not associated with PA, this umbrella SLR provided a comprehensive overview of the associations between behavioral determinants and PA. Youth should be physically active in the early years and increase active transportation to/from school, independent mobility, and âfree-range activitiesâ without adult supervision, whilst adult PA behaviors are mostly influenced by the life events. Finally, more research is needed that incorporates prospective study designs, standardized definitions of PA, objective measurement methods of PA assessment, and the use of interactionist and mediational approaches for the evaluation of different behavioral determinants influencing PA behaviors
SPO11-Independent DNA Repair Foci and Their Role in Meiotic Silencing
In mammalian meiotic prophase, the initial steps in repair of SPO11-induced DNA double-strand breaks (DSBs) are required to obtain stable homologous chromosome pairing and synapsis. The X and Y chromosomes pair and synapse only in the short pseudo-autosomal regions. The rest of the chromatin of the sex chromosomes remain unsynapsed, contains persistent meiotic DSBs, and the whole so-called XY body undergoes meiotic sex chromosome inactivation (MSCI). A more general mechanism, named meiotic silencing of unsynapsed chromatin (MSUC), is activated when autosomes fail to synapse. In the absence of SPO11, many chromosomal regions remain unsynapsed, but MSUC takes place only on part of the unsynapsed chromatin. We asked if spontaneous DSBs occur in meiocytes that lack a functional SPO11 protein, and if these might be involved in targeting the MSUC response to part of the unsynapsed chromatin. We generated mice carrying a point mutation that disrupts the predicted catalytic site of SPO11 (Spo11YF/YF), and blocks its DSB-inducing activity. Interestingly, we observed foci of proteins involved in the processing of DNA damage, such as RAD51, DMC1, and RPA, both in Spo11YF/YFand Spo11 knockout meiocytes.
