Satisfaction with body image for early adolescent females: The impact of pubertal timing within different school environments
During adolescence dramatic physical changes take place which the individual must incorporate into his or her evolving body image. The impact of different school environments on this incorporation process is explored using data on 225 White females from a longitudinal study. Differences in physical characteristics between early, middle, and late developers were assessed each year. The effects of pubertal timing on satisfaction with body image dimensions and self-esteem were then explored for sixth- and seventh-graders within different school environments. Reference group theory was used to examine three alternative hypotheses. Early versus late onset of menarche had different effects on certain aspects of satisfaction with body image, depending on the school environment. Results support the strength of the cultural ideal of thinness for women, but no other hypothesis had consistent support. Physical attractiveness as a mediator of the impact of early pubertal changes for girls
The present study examined the effects of early pubertal development and physical attractiveness on the popularity, body image, and self-esteem of over 200 sixth-grade girls. Two rival hypotheses were explored. The first suggests that physically attractive girls, because of their more favorable social environment, will exhibit fewer psychosocial difficulties than unattractive girls during pubertal development. The second hypothesis argues that attractive girls will exhibit greater difficulty during pubertal transition because their self-image is more intimately connected with their physical appearance. Although there were no significant interactions between attractiveness and pubertal development for either popularity or body image, the second hypothesis was supported with respect to self-esteem. Specifically, developing attractive girls exhibited lower self-esteem than their unattractive counterparts. Genome-wide Analyses Identify KIF5A as a Novel ALS Gene
To identify novel genes associated with ALS, we undertook two lines of investigation. We carried out a genome-wide association study comparing 20,806 ALS cases and 59,804 controls. Independently, we performed a rare variant burden analysis comparing 1,138 index familial ALS cases and 19,494 controls. Through both approaches, we identified kinesin family member 5A (KIF5A) as a novel gene associated with ALS. Interestingly, mutations predominantly in the N-terminal motor domain of KIF5A are causative for two neurodegenerative diseases: hereditary spastic paraplegia (SPG10) and Charcot-Marie-Tooth type 2 (CMT2). In contrast, ALS-associated mutations are primarily located at the C-terminal cargo-binding tail domain and patients harboring loss-of-function mutations displayed an extended survival relative to typical ALS cases. Taken together, these results broaden the phenotype spectrum resulting from mutations in KIF5A and strengthen the role of cytoskeletal defects in the pathogenesis of ALS.Peer reviewe

Background
The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd.
Methods
We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background.
Results
First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001).
Conclusions
In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival
Transition Into Adolescence: A Longitudinal Study, 1974-1979
This longitudinal study evaluates the transition from childhood into early and middle adolescence and how it is affected by the type of school a child attends, onset of puberty, and gender.
The study was conducted between 1974 and 1979 and had two major phases. The first phase examined the transition from late childhood into early adolescence using a 2-year longitudinal design. Students were followed from sixth grade in elementary school into the two types of contrasting seventh grade settings: (1) where the children attended the same school from kindergarten through eighth grade; (2) where the child moved from a kindergarten through sixth grade school onto a junior high school. The second phase of the study, which took place over a second 2-year period (ninth and tenth grade), was designed to study the transition into senior high school and to contrast the effects of a double transition in school environment versus a single change in school environments; that is to contrast students who experienced both a junior and senior high school transition to those who moved directly from elementary school to senior high school.

Understanding donors' motivations: A study of unrelated bone marrow donors
Medical advances in bone marrow transplantation techniques and immunosuppressive medications have dramatically increased the number of such transplants performed each year, and consequently, the demand for bone marrow from unrelated donors. Although physiological aspects of bone marrow donation have been thoroughly investigated, very few studies have examined psychosocial factors that may impact individuals' donation decisions and outcomes. To examine one particular set of donor psychosocial issues, this study investigated motives for bone marrow donation among 343 unrelated bone marrow donors who donated through the National Marrow Donor Program. Six distinct types of donor motives were identified from open-ended questionnaire responses. Donors most frequently reported motives reflecting some awareness of both the costs (to themselves) and potential benefits (to themselves and the recipient) of donation. A desire to act in accordance with social or religious precepts, expected positive feelings about donating, empathy for the recipient, and the simple desire to help another person were also commonly cited reasons for donating. Among a series of donor background characteristics, donors' gender was the variable most strongly associated with motive type; women were most likely to cite expected positive feelings, empathy, and the desire to help someone. Central study findings indicated that donor motives predicted donors reactions to donation even after the effects of donor background characteristics (including gender) were controlled. Donors who reported exchange motives (weighing costs and benefits) and donors who reported simple (or idealized) helping motives experienced the donation as less positive in terms of higher predonation ambivalence and negative postdonation psychological reactions than did remaining donors. Donors who reported positive feeling and empathy motives had the most positive donation reactions in terms of lower ambivalence, and feeling like better persons postdonation. These findings add substantially to the body of work concerning medical volunteerism generally, and also have important practical implications for the recruitment and education of potential bone marrow donors.bone marrow donation altruism

Colorectal Cancer Screening: Language is a Greater Barrier for Latino Men than Latino Women
Colorectal cancer screening (CRC) disparities between non-Latino Whites and Latinos remain, and may have increased. The goal of this analysis was to examine the association between Latino race/ethnicity, gender, and English-proficiency and CRC screening. Analysis of the CDC’s BRFSS 2008 survey. We estimated crude and adjusted screening rates and odds ratios of respondents’ reported CRC test receipt stratified by self-reported Latino ethnicity, gender, and limited English proficiency (LEP) as determined by language of survey response (English vs Spanish). Of 99,883 respondents included in the study populations, LEP Latino men had the lowest adjusted screening rates (48.2 %) which were lower that all other Latinos subgroups including Latina women with LEP (56.2 %). Compared to non-Latino White men, LEP Latino men were 0.47 times as likely to report receiving CRC screening tests (AOR 0.47; 95 % CI 0.35–0.63). Disparities in CRC screening are most dramatic for LEP Latino men
