    Movement of plasma membrane proteins of Ehrlich ascites tumor cells in relation to cap formation induced by concanavalin A: a study on the non-capped areas.

    In order to get precise information about the movement of plasma membrane proteins in cap formation, cyto- and bio-chemical analyses were made of the plasma membranes from non-capped areas of Ehrlich ascites tumor cells (EATCs) exposed to concanavalin A (Con A). Blebs formed by treatment with cytochalasin B (CB) of the non-capped areas of cells having a cap were isolated and used as the plasma membranes from non-capped areas (ConA-CB-bleb fraction). This bleb fraction was compared with a bleb fraction prepared from cells without ConA-treatment (CB-bleb fraction). Cytochemical analysis of ConA-CB-bleb fraction revealed a decreased in conA binding sites (ConA-BS) compared to the CB-bleb fraction. SDS polyacrylamide slab gel electrophoresis also revealed a decrease in the major components of ConA-BS of the ConA-CB-bleb fraction. The minor components of ConA-BS showed no distinct quantitative difference between the ConA-CB-bleb and CB-bleb fractions. NA+ K+-adenosine triphosphatase (ATPase), 5' nucleotidase (5'ND) and gamma-glutamyl transpeptidase (gamma-GTP) did not show any decrease in activity in the ConA-CB-bleb fraction, but the activity of D+-stimulated phosphatase (K-Pase) was decreased. The findings indicate that there are two types of plasma membrane glycoproteins in EATCs; one includes those participating in cap formation due to ConA, e.g. the major components of ConA-BS and K-Pase, and the other, those not participating in such cap formation, e.g. some minor components of ConA-BS, ATPase, 5'ND and gamma-GTP, which keep their places without moving.</p

    Validation of a dietary balance score

    This study assessed the validity of dietary balance scores (DBSs) by investigating the association between DBSs and nutrient adequacy (NA) in two Japanese populations. The participants were 65 community-dwelling Japanese from Tokushima Prefecture and 2,330 community-dwelling Japanese from Aichi Prefecture. Based on food frequency questionnaires or 3-day dietary records, we obtained 18 food groups. The NA score integrates nine beneficial nutrients and two nutrients that should be limited. We calculated four different DBSs: DBS1 consisted of five food groups (score range : 0–20), DBS2 consisted of nine food groups (score range : 0–36), DBS3 consisted of eight food groups (score range : 0–32), and DBS4 consisted of 10 food groups (score range : 0–40). Both the Spearman rank correlation coefficient with NA and the area under the receiver operating characteristic curve (AUC-ROC) for the nine beneficial nutrients were then estimated to test the performance of each DBS in predicting nutrient intake. The results showed that DBS1 and DBS4 were positively correlated with NA, while the AUC-ROC showed that DBS4 could moderately discriminate individuals with adequate intake levels of all nine nutrients. These findings suggest DBSs (especially DBS4) are useful in assessing dietary balance in middle-aged and older community-dwelling Japanese

    Organ distribution of asialo-red blood cell ghosts: an attempt at targeting to the liver.

    We investigated the organ distribution of four types of red blood cells (RBC) preparations: native RBC, asialo-RBC, native ghosts and asialo-ghosts. Intravenously injected asialo-ghosts were rapidly removed from the blood stream and accumulated mainly in the liver 120 min after the injection. Our results suggest that asialo-ghosts are a simple and effective carrier for targeting of drugs to the liver.</p

    A scanning electron microscopic study of the two-step effect of cytochalasin B on Ehrlich ascites tumor cells.

    The effect of cytochalasin B (CB) on the surface structure of Ehrlich ascites tumor cells was investigated using the scanning electron microscope. The effect occurs in two steps: formation of zeiotic knobs on the cell surface and subsequent grouping of the knobs at one pole of the cell. The early step of zeiotic knob formation occurs at low concentrations of CB (0.5-1 microgram/ml) at 37 degrees C and at high concentrations of the drug (5-10 microgram/ml) at low temperature but within 1 min at 37 degrees C. This step is only partially inhibited by 5 x 10(-3) M sodium azide. The subsequent grouping of zeiotic knobs lasts for more than 2 min at 37 degrees C and occurs only in the case of high concentrations of CB. It is inhibited by sodium azide and is often associated with grouping of the microvilli, which are then lost from all of the cell surface except the area of knob-grouping.</p

    Vanishing Parotid Tumors on MR Imaging

    【Background】 Of all parotid gland tumors, only oncocytoma has been reported to appear isointense to the parotid gland, namely vanishing, on fat-saturated T2 and T1 postcontrast gadolinium-enhanced magnetic resonance imaging (MRI). The purpose of this study was to evaluate vanishing of parotid tumors on conventional MRI with and/or without postcontrast gadolinium-enhancement and on diffusion weighted imaging (DWI). 【Methods】 In 8 of 51 patients, ten parotid gland tumors had homogeneously enhanced lesions and were retrospectively analysed. Comparisons of signal intensity between those parotid tumors and parotid glands and evaluations of vanishing were performed on T1-weighted imaging (T1WI), T2-weighted imaging (T2WI), fat-suppressed T2WI (FS-T2WI), postcontrast gadolinium-enhanced T1WI (CE-T1WI) and fat-suppressed CET1WI (FS-CE-T1WI), DWI as well as apparent diffusion coefficient (ADC). 【Results】 Ten parotid gland tumors consisted of five Warthin tumors, two pleomorphic adenomas, two parotid carcinomas (small cell carcinoma and adenoid cystic carcinoma) and one oncocytoma. All tumors showed hypointensity on T1WI and hyperintensity on DWI. Nine of ten tumors showed vanishing on the other MR sequences. All Warthin tumors showed vanishing on FST2WI, FS-CE-T1WI and the ADC map. One oncocytoma showed vanishing on FS-T2WI and the ADC map and hyperintensity on FS-CE-T1WI. All pleomorphic adenomas showed vanishing on T2WI and CE-T1WI. One adenoid cystic carcinoma showed vanishing only on CE-T1WI. 【Conclusion】 Vanishing of parotid tumors can be observed not only on FS-T2WI and FS-CE-T1WI but also on T2WI, CE-T1WI and ADC mapping

    Secular evolution of viscous and self-gravitating circumstellar discs

    We add the effect of turbulent viscosity via the \alpha-prescription to models of the self-consistent formation and evolution of protostellar discs. Our models are non-axisymmetric and carried out using the thin-disc approximation. Self-gravity plays an important role in the early evolution of a disc, and the later evolution is determined by the relative importance of gravitational and viscous torques. In the absence of viscous torques, a protostellar disc evolves into a self-regulated state with disk-averaged Toomre parameter Q \sim 1.5-2.0, non-axisymmetric structure diminishing with time, and maximum disc-to-star mass ratio \xi = 0.14. We estimate an effective viscosity parameter \alpha_eff associated with gravitational torques at the inner boundary of our simulation to be in the range 10^{-4}-10^{-3} during the late evolution. Addition of viscous torques with a low value \alpha = 10^{-4} has little effect on the evolution, structure, and accretion properties of the disc, and the self-regulated state is largely preserved. A sequence of increasing values of \alpha results in the discs becoming more axisymmetric in structure, being more gravitationally stable, having greater accretion rates, larger sizes, shorter lifetimes, and lower disc-to-star mass ratios. For \alpha=10^{-2}, the model is viscous-dominated and the self-regulated state largely disappears by late times. (Abridged)Comment: 13 pages, 11 figures, accepted for publication in MNRA

    "Pseudo-cap" formation in Ehrlich ascites tumor cells induced by cytochalasin B.

    Cytochalasin B (CB) treatment induces or accelerates the capping phenomenon in some cells. In Ehrlich ascites tumor cells (EATC) CB treatment apparently induced the capping of Con A binding sites as observed under a fluorescent microscope. However, electron microscopic examinations revealed that the CB treatment did not induce a rearrangement of Con A binding sites, but rather it only induced a change in cell shape. On the contrary, CB treatment inhibited the capping phenomenon induced by treatment with Con A. Electron microscopic observations may give exact information on the distribution of lectin binding sites.</p

    Sub-Keplerian accretion onto circumstellar disks

    Models of the formation, evolution and photoevaporation of circumstellar disks are an essential ingredient in many theories of the formation of planetary systems. The ratio of disk mass over stellar mass in the circumstellar phase of a disk is largely determined by the angular momentum of the original cloud core from which the system was formed. While full 3D or 2D axisymmetric hydrodynamical models of accretion onto the disk automatically treat all aspects of angular momentum, this is not so trivial for 1D and semi-2D viscous disk models. Since 1D and semi-2D disk models are still very useful for long-term evolutionary modelling of disks with relatively little numerical effort, we investigate how the 2D nature of accretion affects the formation and evolution of the disk in such models. A proper treatment of this problem requires a correction for the sub-Keplerian velocity at which accretion takes place. We develop an update of our semi-2D time-dependent disk evolution model to properly treat the effects of sub-Keplerian accretion. The new model also accounts for the effects of the vertical extent of the disk on the accretion streamlines from the envelope. The disks produced with the new method are smaller than those obtained previously, but their mass is mostly unchanged. The new disks are a few degrees warmer in the outer parts, so they contain less solid CO. Otherwise, the results for ices are unaffected. The 2D treatment of the accretion results in material accreting at larger radii, so a smaller fraction comes close enough to the star for amorphous silicates to be thermally annealed into crystalline form. The lower crystalline abundances thus predicted correspond more closely to observed abundances than did earlier model predictions. We argue that thermal annealing followed by radial mixing must be responsible for at least part of the observed crystalline material.Comment: Accepted by A&A; 10 pages, 6 figure
