    Two storage hexamerins from the beet armyworm Spodoptera exigua: Cloning, characterization and the effect of gene silencing on survival

    <p>Abstract</p> <p>Background</p> <p>In insects, hemocyanin superfamily proteins accumulate apparently to serve as sources of amino acids during metamorphosis, reproduction and development. Storage hexamerins are important members of the hemocyanin superfamily. Although insects possess storage hexamerins, very little is known about the character and specific functions of hexamerin 1 and storage protein 1 in insect development.</p> <p>Results</p> <p>To gain insight into the function of storage proteins in insects, cDNAs for two storage proteins were cloned from the fat body of <it>Spodoptera exigua</it>. <it>S. exigua </it>hexamerin 1 (<it>SeHex</it>) cDNA contained an open reading frame of 2124 nucleotides encoding a protein of 707 amino acids with a predicted molecular weight of 82.12 kDa. <it>S. exigua </it>storage protein 1 (<it>SeSP1</it>) cDNA contained an open reading frame of 2256 bp encoding a protein of 751 amino acids with a predicted molecular weight of ~88.84 kDa. Northern blotting analyses revealed that <it>SeHex </it>mRNA is expressed in the fat body, cuticle, midgut and Malpighian tubules and <it>SeSP1 </it>in fat body, Malpighian tubules and tracheae. <it>SeHex </it>and <it>SeSP1 </it>mRNAs were expressed in fat body at different levels from first instar larvae to pupae, with expression was much lower from first instar larvae to first-day fifth instar larvae. <it>SeHex </it>transcript expression was high in fat body of wandering larvae (pre-pupae) and steadily decreased to the seventh pupal day. <it>SeSP1 </it>transcript expression was high in fat body of wandering larvae, 2-day-old fifth instar larvae and 2-, 4- and 7-day-old pupae. <it>SeHex </it>and <it>SeSP1 </it>mRNAs levels were expressed lower than control on the condition of starvation at 12 h. Of insects injected with SeHex and SeSP1 dsRNA, 38.7% and 24.3% survived to 204 h after treatment, respectively. This was significantly lower than in the controls groups.</p> <p>Conclusions</p> <p>These findings provide new data on the tissue distribution, expression patterns and the function in starvation of storage proteins. RNA interference results revealed that storage protein genes are key in metamorphosis, reproduction and insect development. The results for <it>SeHex </it>and <it>SeSP1 </it>interference reveal that a potential method to control this pest is to disrupt the regulation of storage proteins.</p

    Representative sampling method for laboratory testing on shear strength of rock joints

    The direct shear test is commonly used to determine the shear strength of rock joints. The measured shear strength, however, varies greatly with specimen positions due to the heterogeneity of rock joints. As a result, selecting specimens that represent the overall properties of rock joints is usually difficult. This paper aims to investigate the heterogeneity of rock joints and propose a new sampling method for selecting representative specimens. The roughness and shear strength variations of specimens taken from different positions of a natural rock joint were analyzed, and it was discovered that the heterogeneity of rock joint roughness is responsible for the heterogeneity of shear strength. The limitations of the traditional sampling method based on visual judgment were extensively investigated, revealing that shear strength parameters acquired by the traditional method contain large coefficient of variation (COV) values. To acquire trustworthy shear strength parameters, we proposed a representative sampling method based on the maximum likelihood estimation of the overall properties of rock joints. The number of determined representative specimens increases with the increase of normal stress. Representative specimens determined under low normal stresses can likewise exhibit the overall properties of the rock joint when subjected to high normal stresses. The Mohr-Coulomb and a nonlinear criterion were used to validate the derived representative specimens, demonstrating that the proposed method can produce reliable shear strength parameters and shear strength envelopes. Particularly, the determined representative specimens could derive shear strength parameters with relative errors less than 10% and COV values less than 0.1. The proposed method provides a quantitative and reliable tool for determining representative specimens to obtain reliable shear strength of rock joints

    Sex Steroids and Osteoarthritis: A Mendelian Randomization Study

    ObjectiveSex steroids are thought to contribute to the pathogenesis of osteoarthritis (OA). This study investigated the causal role of sex steroids in site- and sex-specific OA and risk of joint replacement surgery using the Mendelian randomization (MR) method.MethodsInstrumental variables for estradiol, dehydroepiandrosterone sulfate, testosterone (T), and dihydrotestosterone (DHT) were selected. We used the inverse variance weighting (IVW) approach as the main MR method to estimate causal effects based on the summary-level data for OA and joint replacement surgery from genome-wide association studies (GWAS).ResultsA positive causal association was observed between serum T level and risks of hip OA (odds ratio [OR]=1.558, 95% confidence interval [CI]: 1.193–2.034; P=0.001) and hip replacement (OR=1.013, 95% CI: 1.008–1.018; P=2.15×10−8). Serum DHT level was also positively associated with the risk of hip replacement (OR=1.011, 95% CI: 1.006–1.015; P=4.03×10−7) and had potential causality with hip OA (OR=1.398, 95% CI: 1.054–1.855; P=0.020).ConclusionsSerum T and DHT levels may play causal roles in the development of hip OA and contribute to the risk of hip replacement, although the underlying mechanisms require further investigation

    Paramagnetic behaviour of silver nanoparticles generated by decomposition of silver oxalate

    Silver oxalate Ag2C2O4, was already proposed for soldering applications, due to the formation when it is decomposed by a heat treatment, of highly sinterable silver nanoparticles. When slowly decomposed at low temperature (125 °C), the oxalate leads however to silver nanoparticles isolated from each other. As soon as these nanoparticles are formed, the magnetic susceptibility at room temperature increases from -3.14 10-7 emu.Oe-1.g-1 (silver oxalate) up to -1.92 10-7 emu.Oe-1.g-1 (metallic silver). At the end of the oxalate decomposition, the conventional diamagnetic behaviour of bulk silver, is observed from room temperature to 80 K. A diamagnetic-paramagnetic transition is however revealed below 80 K leading at 2 K, to silver nanoparticles with a positive magnetic susceptibility. This original behaviour, compared to the one of bulk silver, can be ascribed to the nanometric size of the metallic particles

    <p>As a chronic disease, osteoarthritis (OA) leads to the degradation of both cartilage and subchondral bone, its development being mediated by proinflammatory cytokines like interleukin-1β. In the present study, the anti-inflammatory effect of specnuezhenide (SPN) in OA and its underlying mechanism were studied in vitro and in vivo. The results showed that SPN decreases the expression of cartilage matrix-degrading enzymes and the activation of NF-κB and wnt/β-catenin signaling, and increases chondrocyte-specific gene expression in IL-1β-induced inflammation in chondrocytes. Furthermore, SPN treatment prevents the degeneration of both cartilage and subchondral bone in a rat model of OA. To the best of our knowledge, this study is the first to report that SPN decreases interleukin-1β-induced inflammation in rat chondrocytes by inhibiting the activation of the NF-κB and wnt/β-catenin pathways, and, thus, has therapeutic potential in the treatment of OA.</p

    Stevioside Prevents Wear Particle-Induced Osteolysis by Inhibiting Osteoclastogenesis and Inflammatory Response via the Suppression of TAK1 Activation

    Aseptic loosening and periprosthetic osteolysis are the leading causes of total joint arthroplasty failure, which occurs as a result of chronic inflammatory response and enhanced osteoclast activity. Here we showed that stevioside, a natural compound isolated from Stevia rebaudiana, exhibited preventative effects on titanium particle-induced osteolysis in a mouse calvarial model. Further histological assessment and real-time PCR analysis indicated that stevioside prevented titanium particle-induced osteolysis by inhibiting osteoclast formation and inflammatory cytokine expression in vivo. In vitro, we found that stevioside could suppress RANKL-induced osteoclastogenesis and titanium particle-induced inflammatory response in a dose-dependent manner. Mechanistically, stevioside achieved these effects by disrupting the phosphorylation of TAK1 and subsequent activation of NF-κB/MAPKs signaling pathways. Collectively, our data suggest that stevioside effectively suppresses osteoclastogenesis and inflammatory response both in vitro and in vivo, and it might be a potential therapy for particle-induced osteolysis and other osteolytic diseases

    HemoglobinA1c Is a Risk Factor for Changes of Bone Mineral Density: A Mendelian Randomization Study

    BackgroundAs a valuable blood glucose measurement, HemoglobinA1c (HbA1c) is of great clinical value for diabetes. However, in previous observational studies, studies on its effect on bone mineral density (BMD) have different results. This study aimed to use Mendelian randomization (MR) to assess the effect of HbA1c on bone mineral density and fracture risk, and try to further explore whether this association was achieved through glycemic or non-glycemic factors.MethodsTake HbA1c measurement as exposure, and BMD estimated from quantitative heel ultrasounds (eBMD) and bone fractures as outcomes. Two-Sample MR Analysis was conducted to assess the causal effect of HbA1C on heel BMD and risk fracture. Then, we performed the analysis using two subsets of these variants, one related to glycemic measurement and the other to erythrocyte indices.ResultsGenetically increased HbA1C was associated with the lower heel eBMD [odds ratio (OR) 0.91 (95% CI 0.87, 0.96) per %-unit, P = 3 × 10−4(IVW)]. Higher HbA1C was associated with lower heel eBMD when using only erythrocytic variants [OR 0.87 (0.82, 0.93), P=2× 10−5(IVW)]; However, when using only glycemic variants, this casual association does not hold. In further MR analysis, we test the association of erythrocytic traits with heel eBMD.ConclusionOur study revealed the significant causal effect of HbA1c on eBMD, and this causal link might achieve through non-glycemic pathways (erythrocytic indices)

    Existence and stability of traveling wave fronts in reaction advection diffusion equations with nonlocal delay

    AbstractThis paper is concerned with the existence, uniqueness and globally asymptotic stability of traveling wave fronts in the quasi-monotone reaction advection diffusion equations with nonlocal delay. Under bistable assumption, we construct various pairs of super- and subsolutions and employ the comparison principle and the squeezing technique to prove that the equation has a unique nondecreasing traveling wave front (up to translation), which is monotonically increasing and globally asymptotically stable with phase shift. The influence of advection on the propagation speed is also considered. Comparing with the previous results, our results recovers and/or improves a number of existing ones. In particular, these results can be applied to a reaction advection diffusion equation with nonlocal delayed effect and a diffusion population model with distributed maturation delay, some new results are obtained

    Analysis of Rabies in China: Transmission Dynamics and Control

    Human rabies is one of the major public-health problems in China. The number of human rabies cases has increased dramatically in the last 15 years, partially due to the poor understanding of the transmission dynamics of rabies and the lack of effective control measures of the disease. In this article, in order to explore effective control and prevention measures we propose a deterministic model to study the transmission dynamics of rabies in China. The model consists of susceptible, exposed, infectious, and recovered subpopulations of both dogs and humans and describes the spread of rabies among dogs and from infectious dogs to humans. The model simulations agree with the human rabies data reported by the Chinese Ministry of Health. We estimate that the basic reproduction number for the rabies transmission in China and predict that the number of the human rabies is decreasing but may reach another peak around 2030. We also perform some sensitivity analysis of in terms of the model parameters and compare the effects of culling and immunization of dogs. Our study demonstrates that (i) reducing dog birth rate and increasing dog immunization coverage rate are the most effective methods for controlling rabies in China; and (ii) large scale culling of susceptible dogs can be replaced by immunization of them
