    Generation and analysis of ESTs from strawberry (Fragaria xananassa) fruits and evaluation of their utility in genetic and molecular studies

    <p>Abstract</p> <p>Background</p> <p>Cultivated strawberry is a hybrid octoploid species (<it>Fragaria xananassa </it>Duchesne ex. Rozier) whose fruit is highly appreciated due to its organoleptic properties and health benefits. Despite recent studies on the control of its growth and ripening processes, information about the role played by different hormones on these processes remains elusive. Further advancement of this knowledge is hampered by the limited sequence information on genes from this species, despite the abundant information available on genes from the wild diploid relative <it>Fragaria vesca</it>. However, the diploid species, or one ancestor, only partially contributes to the genome of the cultivated octoploid. We have produced a collection of expressed sequence tags (ESTs) from different cDNA libraries prepared from different fruit parts and developmental stages. The collection has been analysed and the sequence information used to explore the involvement of different hormones in fruit developmental processes, and for the comparison of transcripts in the receptacle of ripe fruits of diploid and octoploid species. The study is particularly important since the commercial fruit is indeed an enlarged flower receptacle with the true fruits, the achenes, on the surface and connected through a network of vascular vessels to the central pith.</p> <p>Results</p> <p>We have sequenced over 4,500 ESTs from <it>Fragaria xananassa</it>, thus doubling the number of ESTs available in the GenBank of this species. We then assembled this information together with that available from <it>F. xananassa </it>resulting a total of 7,096 unigenes. The identification of SSRs and SNPs in many of the ESTs allowed their conversion into functional molecular markers. The availability of libraries prepared from green growing fruits has allowed the cloning of cDNAs encoding for genes of auxin, ethylene and brassinosteroid signalling processes, followed by expression studies in selected fruit parts and developmental stages. In addition, the sequence information generated in the project, jointly with previous information on sequences from both <it>F. xananassa </it>and <it>F. vesca</it>, has allowed designing an oligo-based microarray that has been used to compare the transcriptome of the ripe receptacle of the diploid and octoploid species. Comparison of the transcriptomes, grouping the genes by biological processes, points to differences being quantitative rather than qualitative.</p> <p>Conclusions</p> <p>The present study generates essential knowledge and molecular tools that will be useful in improving investigations at the molecular level in cultivated strawberry (<it>F. xananassa</it>). This knowledge is likely to provide useful resources in the ongoing breeding programs. The sequence information has already allowed the development of molecular markers that have been applied to germplasm characterization and could be eventually used in QTL analysis. Massive transcription analysis can be of utility to target specific genes to be further studied, by their involvement in the different plant developmental processes.</p

    Systemic lupus erythematosus (SLE) is a systemic autoimmune disease. Pleuropulmonary manifestations occur in almost 50% of patients during the course of the disease. Of all of the manifestations, acute lupus pneumonitis (ALP) is an unusual complication, the clinical presentation of which is characterized by fever, dyspnea, pleurisy, and cough with minimal expectoration. The mainstay of treatment is systemic corticosteroids and high-dose immunosuppressants. The objective of this article is to describe an infrequent complication with high mortality in patients with systemic lupus erythematosus.El lupus eritematoso sistémico (LES) es una enfermedad autoinmune sistémica. Las manifestaciones pleuropulmonares se presentan casi en un 50% de los pacientes en el transcurso de la enfermedad. De todas las manifestaciones, la neumonitis lúpica aguda (NLA) es una complicación inusual, cuya presentación clínica se caracteriza por fiebre, disnea, pleuritis y tos con mínima expectoración. La base del tratamiento son los corticoesteroides sistémicos e inmunodepresores a dosis altas.     Factores a considerar en la elección del modo de entrada para la internacionalización de la industria hotelera andaluza

    El turismo es uno de los principales sectores que sustenta la economía española, y en particular la andaluza. Dentro de éste, la industria hotelera tiene el mayor peso dentro del sector. Debido a una serie de motivos, se hace necesario que las cadenas hoteleras andaluzas deban emprender su proceso de internacionalización. En este trabajo, hacemos una radiografía de la situación actual de las cadenas hoteleras andaluzas. A partir de ahí, y tras un estudio analítico de la internacionalización de la industria hotelera describimos los distintos modos de entrada en el exterior utilizados en la internacionalización de esta industria en particular, y los distintos factores que hay que considerar para la correcta elección de la modalidad de penetración en otros mercados. Nuestro principal objetivo, por tanto, es definir, a partir de las explicaciones tradicionalmente utilizadas en la elección del modo de entrada, un conjunto de factores que han de tenerse en consideración para decidir entre las diferentes opciones de penetración en mercados exteriores. La evidencia empírica la realizamos a partir de una nuestra de 122 establecimientos que las distintas cadenas hoteleras españolas tienen operando fuera de nuestro país. Con la aplicación de la Regresión Logística Ordinal (LOGIT model), y a partir del modelo de Contractor y Kundu (1998), pretendemos establecer qué factores inciden en la elección entre las distintas modalidades. Ésta vendrá determinada por variables relacionadas con el país de destino, variables específicas de la empresa, así como de una serie de factores relacionados con objetivos estratégicos. ABSTRACT The tourism is one of the principal sectors that sustains the Spanish economy, and especially the Andalusian one. Inside this one, the hotel industry has the major weight inside the sector. Due to a series of motives, it becomes necessary that the hotel Andalusian chains should tackle his process of internationalization. In this work, we do a X-ray photography of the current situation of the hotel Andalusian chains. From there, and after an analytical study of the internationalization of the hotel industry we describe the different manners of entry in the exterior used in the internationalization of this industry especially, and the different factors that it is necessary to consider for the correct choice of the modality of penetration other markets. Our principal aim, therefore, is to define, from the explanations traditionally used in the election of the way of entry, a set of factors that have to be had in consideration to decide between the different options of penetration foreign markets. We realize the empirical evidence from a sample of 122 establishments that the different hotel Spanish chains haveo operating out of our country. With the application of the Logistic Ordinal Regression (LOGIT model), and from the model of Contractor and Kundu (1998), we try to establish what factors affect in the choice between the different modalities.     [EN] Nutrient recovery technologies are rapidly expanding due to the need for the appropriate recycling of key elements from waste resources in order to move towards a truly sustainable modern society based on the Circular Economy. Nutrient recycling is a promising strategy for reducing the depletion of non-renewable resources and the environmental impact linked to their extraction and manufacture. However, nutrient recovery technologies are not yet fully mature, as further research is needed to optimize process efficiency and enhance their commercial applicability. This paper reviews state-of-the-art of nutrient recovery, focusing on frontier technological advances and economic and environmental innovation perspectives.     A genome-wide association study suggests the HLA Class II region as the major susceptibility locus for IgA vasculitis.

    The genetic component of Immunoglobulin-A (IgA) vasculitis is still far to be elucidated. To increase the current knowledge on the genetic component of this vasculitis we performed the first genome-wide association study (GWAS) on this condition. 308 IgA vasculitis patients and 1,018 healthy controls from Spain were genotyped by Illumina HumanCore BeadChips. Imputation of GWAS data was performed using the 1000 Genomes Project Phase III dataset as reference panel. After quality control filters and GWAS imputation, 285 patients and 1,006 controls remained in the datasets and were included in further analysis. Additionally, the human leukocyte antigen (HLA) region was comprehensively studied by imputing classical alleles and polymorphic amino acid positions. A linkage disequilibrium block of polymorphisms located in the HLA class II region surpassed the genome-wide level of significance (OR = 0.56, 95% CI = 0.46-0.68). Although no polymorphic amino acid positions were associated at the genome-wide level of significance, P-values of potential relevance were observed for the positions 13 and 11 of HLA-DRB1 (P = 6.67E-05, P = 1.88E-05, respectively). Outside the HLA, potential associations were detected, but none of them were close to the statistical significance. In conclusion, our study suggests that IgA vasculitis is an archetypal HLA class II disease

    Association of HLA-B*41:02 with Henoch-Schönlein Purpura (IgA Vasculitis) in Spanish individuals irrespective of the HLA-DRB1 status

    INTRODUCTION: A study was conducted to determine whether the human leukocyte antigen (HLA) B alleles are implicated in the susceptibility to Henoch-Schönlein purpura (HSP) in the largest series of Caucasian HSP patients ever assessed for genetic studies. METHODS: The study population was composed of 349 Spanish patients diagnosed with HSP fulfilling the American College of Rheumatology and the Michel et al. classification criteria, and 335 sex and ethnically matched controls. HLA-B phenotypes were determined by sequencing-based typing (SBT) and analyzed by chi-square or Fisher exact test. RESULTS: A statistically significant increase of HLA-B*41:02 allele in HSP patients when compared with controls was found (8.3% versus 1.5% respectively; P = 0.0001; OR (odds ratio) =5.76 [2.15-19.3]). These results remained statistically significant after adjusting for Bonferroni correction (P = 0.0028). An internal validation also confirmed the susceptibility effect on HSP associated with HLA-B*41:02 (OR = 5.70 [1.98-16.44]). Since a former study described an association between HLA-DRB1*01:03 and HSP susceptibility, we also evaluated the implication of HLA-B*41:02 independently of HLA-DRB1*01:03. Interestingly, the association remained statistically significant (P = 0.0004, OR = 4.97 [1.8-16.9]). No HLA-B association with specific HSP clinical features was found. CONCLUSIONS: Our study indicates that HLA-B*41:02 is associated with the susceptibility to HSP in Spanish patients irrespective of HLA-DRB1 status.This study was supported by a grant from ‘Fondo de Investigaciones Sanitarias’ PI12/00193 (Spain).     Mucosal Immune Defence Gene Polymorphisms as Relevant Players in the Pathogenesis of IgA Vasculitis?

    ITGAM&ndash;ITGAX (rs11150612, rs11574637), VAV3 rs17019602, CARD9 rs4077515, DEFA (rs2738048, rs10086568), and HORMAD2 rs2412971 are mucosal immune defence polymorphisms, that have an impact on IgA production, described as risk loci for IgA nephropathy (IgAN). Since IgAN and Immunoglobulin-A vasculitis (IgAV) share molecular mechanisms, with the aberrant deposit of IgA1 being the main pathophysiologic feature of both entities, we assessed the potential influence of the seven abovementioned polymorphisms on IgAV pathogenesis. These seven variants were genotyped in 381 Caucasian IgAV patients and 997 matched healthy controls. No statistically significant differences were observed in the genotype and allele frequencies of these seven polymorphisms when the whole cohort of IgAV patients and those with nephritis were compared to controls. Similar genotype and allele frequencies of all polymorphisms were disclosed when IgAV patients were stratified according to the age at disease onset or the presence/absence of gastrointestinal or renal manifestations. Likewise, no ITGAM&ndash;ITGAX and DEFA haplotype differences were observed when the whole cohort of IgAV patients, along with those with nephritis and controls, as well as IgAV patients, stratified according to the abovementioned clinical characteristics, were compared. Our results suggest that mucosal immune defence polymorphisms do not represent novel genetic risk factors for IgAV pathogenesis

    Impact of the 13-valent conjugated pneumococcal vaccine on the direct costs of invasive pneumococcal disease requiring hospital admission in children aged < 5 years. A prospective study

    The lack of invasive pneumococcal disease (IPD) cost studies may underestimate the effect of pneumococcal polysaccharide conjugated vaccines (PCV). The objective of this study was to estimate the direct costs of hospitalized IPD cases. A prospective study was made in children aged <5 years diagnosed with IPD in two high-tech hospitals in Catalonia (Spain) between 2007-2009 (PCV7 period) and 2012-2015 (PCV13 period). Costs were calculated according to 2014 Catalan Health Service rates using diagnostic-related groups. In total, 319 and 154 cases were collected, respectively. Pneumonia had the highest cost (65.7% and 62.0%, respectively), followed by meningitis (25.8% and 26.1%, respectively). During 2007-2015, the costs associated with PCV7 serotypes (Pearson coeffcient (Pc) = 0.79; p = 0.036) and additional PCV13 serotypes (Pc = 0.75; p = 0.05) decreased, but those of other serotypes did not (Pc = 0.23 p = 0.62). The total mean cost of IPD increased in the PCV13 period by 31.4% (¿3016.1 vs. ¿3963.9), mainly due to ICU stay (77.4%; ¿1051.4 vs. ¿1865.6). During the PCV13 period, direct IPD costs decreased due to a reduction in the number of cases, but cases were more severe and had a higher mean cost. During 2015, IPD costs increased due to an increase in the costs associated with non-PCV13 serotypes and serotype 3 and this requires further investigation

    Maximizing resource recovery from urban wastewater through an innovative facility layout

    [EN] This research work proposes an innovative layout for urban wastewater treatment based on anaerobic technology, microalgal cultivation and membrane technology. The proposed Water Resource Recovery Facility (WRRF) system can treat urban wastewater efficiently, complying with legal discharge limits and allowing for resource recovery, i.e. energy, nutrients and reclaimed water. In addition, the proposed layout produces less solid wastes than a conventional wastewater treatment plant (WWTP) and it is possible to recover energy as biogas, not only from the original wastewater sources but also from the biomass generated in the WRRF system

    Role of the IL33 and IL1RL1 pathway in the pathogenesis of Immunoglobulin A vasculitis

    Cytokines signalling pathway genes are crucial factors of the genetic network underlying the pathogenesis of Immunoglobulin-A vasculitis (IgAV), an inflammatory vascular condition. An influence of the interleukin (IL)33- IL1 receptor like (IL1RL)1 signalling pathway on the increased risk of several immune-mediated diseases has been described. Accordingly, we assessed whether the IL33-IL1RL1 pathway represents a novel genetic risk factor for IgAV. Three tag polymorphisms within IL33 (rs3939286, rs7025417 and rs7044343) and three within IL1RL1 (rs2310173, rs13015714 and rs2058660), that also were previously associated with several inflammatory diseases, were genotyped in 380 Caucasian IgAV patients and 845 matched healthy controls. No genotypes or alleles differences were observed between IgAV patients and controls when IL33 and IL1RL1 variants were analysed independently. Likewise, no statistically significant differences were found in IL33 or IL1RL1 genotype and allele frequencies when IgAV patients were stratified according to the age at disease onset or to the presence/absence of gastrointestinal (GI) or renal manifestations. Similar results were disclosed when IL33 and IL1RL1 haplotypes were compared between IgAV patients and controls and between IgAV patients stratified according to the clinical characteristics mentioned above.
