Production of human recombinant proapolipoprotein A-I in Escherichia coli: purification and biochemical characterization
A human liver cDNA library was used to isolate a clone coding for apolipoprotein A-I (Apo A-I). The clone
carries the sequence for the prepeptide (18 amino acids), the propeptide (6 amino acids), and the mature protein
(243 amino acids). A coding cassette for the proapo A-I molecule was reconstructed by fusing synthetic
sequences, chosen to optimize expression and specifying the amino-terminal methionine and amino acids -6
to +14, to a large fragment of the cDNA coding for amino acids 15-243. The module was expressed in
pOTS-Nco, an Escherichia coli expression vector carrying the regulatable X P^ promoter, leading to the production
of proapolipoprotein A-I at up to 10% of total soluble proteins. The recombinant polypeptide was
purified and characterized in terms of apparent molecular mass, isoelectric point, and by both chemical and
enzymatic peptide mapping. In addition, it was assayed in vitro for the stimulation of the enzyme lecithin:
cholesterol acyltransferase. The data show for the first time that proapo A-I can be produced efficiently in
E. coli as a stable and undegraded protein having physical and functional properties indistinguishable from
those of the natural product
Modification of Amorphous Mesoporous Zirconia Nanoparticles with Bisphosphonic Acids: A Straightforward Approach for Tailoring the Surface Properties of the Nanoparticles
The use of readily prepared bisphosphonic acids obtained in few steps through a thio-Michael addition of commercially available thiols on tetraethyl vinylidenebisphosphonate enables the straightforward surface modification of amorphous mesoporous zirconia nanoparticles. Simple stirring of the zirconia nanoparticles in a buffered aqueous solution of the proper bisphosphonic acid leads to the surface functionalization of the nanoparticles with different kinds of functional groups, charge and hydrophobic properties. Formation of both chemisorbed and physisorbed layers of the bisphosphonic acid take place, observing after extensive washing a grafting density of 1.1 molecules/nm2 with negligible release in neutral or acidic pH conditions, demonstrating stronger loading compared to monophosphonate derivatives. The modified nanoparticles were characterized by IR, XPS, ζ-potential analysis to investigate the loading of the bisphosphonic acid, FE-SEM to investigate the size and morphologies of the nanoparticles and 31P and 1H MAS NMR to investigate the coordination motif of the phosphonate units on the surface. All these analytical techniques demonstrated the strong affinity of the bisphosphonic moiety for the Zr(IV) metal centers. The functionalization with bisphosphonic acids represents a straightforward covalent approach for tailoring the superficial properties of zirconia nanoparticles, much straightforward compared the classic use of trisalkoxysilane or trichlorosilane reagents typically employed for the functionalization of silica and metal oxide nanoparticles. Operation of Energy MicroGrids
Integrated Operation is a strategy to operate the Energy MicroGrid as an integrated system. In this strategy, a CHP plant, an electric water heater, and thermal storage were operated to support both electricity and heat networks of the Energy MicroGrid. An Integrated Optimal Power Flow was formulated and was used to investigate the advantages of this Integrated Operation. Simulations indicated that Integrated Operation minimised generation curtailment and load shedding. MARTA: A high-energy cosmic-ray detector concept with high-accuracy muon measurement
A new concept for the direct measurement of muons in air showers is
presented. The concept is based on resistive plate chambers (RPCs), which can
directly measure muons with very good space and time resolution. The muon
detector is shielded by placing it under another detector able to absorb and
measure the electromagnetic component of the showers such as a water-Cherenkov
detector, commonly used in air shower arrays. The combination of the two
detectors in a single, compact detector unit provides a unique measurement that
opens rich possibilities in the study of air showers.Comment: 11 page
Azimuthal asymmetry in the risetime of the surface detector signals of the Pierre Auger Observatory
The azimuthal asymmetry in the risetime of signals in Auger surface detector
stations is a source of information on shower development. The azimuthal
asymmetry is due to a combination of the longitudinal evolution of the shower
and geometrical effects related to the angles of incidence of the particles
into the detectors. The magnitude of the effect depends upon the zenith angle
and state of development of the shower and thus provides a novel observable,
, sensitive to the mass composition of cosmic rays
above eV. By comparing measurements with predictions from
shower simulations, we find for both of our adopted models of hadronic physics
(QGSJETII-04 and EPOS-LHC) an indication that the mean cosmic-ray mass
increases slowly with energy, as has been inferred from other studies. However,
the mass estimates are dependent on the shower model and on the range of
distance from the shower core selected. Thus the method has uncovered further
deficiencies in our understanding of shower modelling that must be resolved

Evidence for a mixed mass composition at the `ankle' in the cosmic-ray spectrum
We report a first measurement for ultra-high energy cosmic rays of the
correlation between the depth of shower maximum and the signal in the water
Cherenkov stations of air-showers registered simultaneously by the fluorescence
and the surface detectors of the Pierre Auger Observatory. Such a correlation
measurement is a unique feature of a hybrid air-shower observatory with
sensitivity to both the electromagnetic and muonic components. It allows an
accurate determination of the spread of primary masses in the cosmic-ray flux.
Up till now, constraints on the spread of primary masses have been dominated by
systematic uncertainties. The present correlation measurement is not affected
by systematics in the measurement of the depth of shower maximum or the signal
in the water Cherenkov stations. The analysis relies on general characteristics
of air showers and is thus robust also with respect to uncertainties in
hadronic event generators. The observed correlation in the energy range around
the `ankle' at differs significantly from
expectations for pure primary cosmic-ray compositions. A light composition made
up of proton and helium only is equally inconsistent with observations. The
data are explained well by a mixed composition including nuclei with mass . Scenarios such as the proton dip model, with almost pure compositions, are
thus disfavoured as the sole explanation of the ultrahigh-energy cosmic-ray
