    Accounting for Research Quality: Research Audits and the Journal Rankings Debate

    The question of whether and how research quality should be measured, and the consequences of research audits such as the UK's Research Excellence Framework (REF) – formerly the RAE – are considered in relation to the role of journal ratings such as the Association of Business Schools Academic Journal Quality Guide (the ABS Guide). Criticism of the ABS Guide has distracted attention from the results of successive RAEs, where the panel for Business and Management has been one of the most selective in its allocation of the highest grades, especially when compared with the neighbouring field of Economics. If the ABS Guide had been used to grade outputs submitted for Business and Management in the RAE 2008 then many more outputs would have received the highest grades, especially in accounting where outputs from journals such as Critical Perspectives on Accounting, which are highly rated in the ABS Guide, appear to have been downgraded by the RAE panel. The alleged bias against accounting in the ABS Guide rests on a particular interpretation of citation impact factors for journals, and a narrow definition of subject fields. Critics of the ABS Guide would be better advised to direct their attention to scrutinizing the results of the REF and considering whether it provides an adequate research ranking for UK business schools. 15% of all full time students in the UK study business and management, including accounting and finance, but only 6.7% of the full time equivalent research active staff submitted in the RAE 2008 were in business and management, or accounting and finance. Research audits are therefore forcing the separation of teaching from research in UK business schools. With an estimated ratio of 71 full time students per research active faculty member in UK business schools, it may be time to consider a more appropriate, inclusive, and economical form of ranking for research in business and management

    Genetic Variation in OAS1 Is a Risk Factor for Initial Infection with West Nile Virus in Man

    West Nile virus (WNV) is a re-emerging pathogen that can cause fatal encephalitis. In mice, susceptibility to WNV has been reported to result from a single point mutation in oas1b, which encodes 2′–5β€² oligoadenylate synthetase 1b, a member of the type I interferon-regulated OAS gene family involved in viral RNA degradation. In man, the human ortholog of oas1b appears to be OAS1. The β€˜A’ allele at SNP rs10774671 of OAS1 has previously been shown to alter splicing of OAS1 and to be associated with reduced OAS activity in PBMCs. Here we show that the frequency of this hypofunctional allele is increased in both symptomatic and asymptomatic WNV seroconverters (Caucasians from five US centers; total nβ€Š=β€Š501; ORβ€Š=β€Š1.6 [95% CI 1.2–2.0], Pβ€Š=β€Š0.0002 in a recessive genetic model). We then directly tested the effect of this SNP on viral replication in a novel ex vivo model of WNV infection in primary human lymphoid tissue. Virus accumulation varied markedly among donors, and was highest for individuals homozygous for the β€˜A’ allele (P<0.0001). Together, these data identify OAS1 SNP rs10774671 as a host genetic risk factor for initial infection with WNV in humans

    Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage. We did subgroup analyses of the RESTART trial to explore whether these brain imaging features modify the effects of antiplatelet therapy

    Assessing past water availability using phytoliths from the C4 plant Sorghum bicolor: an experimental approach

    Water availability and water management systems were critical for the success of past agricultural societies. One way to determine past water availability is through phytolith ratios as demonstrated by research conducted on modern C3 plants. In order to determine if phytolith ratios in C4 plants are similarly affected by plant water availability, the C4 plant Sorghum bicolor was experimentally grown at three different crop growing stations over a two year period in Jordan. The husks, leaves and stems of the plants grown under the 0% and 100% irrigation regimes were processed and analysed for their sensitive to fixed form phytolith ratio. These results were then compared to results of those conducted using C3 plants. Our results showed that while there were differences in ratios between growing years and the crop growing site, the greatest difference in the ratios was in irrigation regime. Our results also showed, however, that the ratio of sensitive to fixed forms for the samples taken from the husks was far higher than the ratios found in the leaves and stems and far higher than those found in previous studies on C3 grasses. We suggest that if this method is to be used to interpret archaeological phytolith assemblages, an assessment of the likely taxa and plant part composition of the assemblage should first be undertaken through phytolith and macro-botanical analysis

    Transient exposure to low levels of insecticide affects metabolic networks of honeybee larvae

    The survival of a species depends on its capacity to adjust to changing environmental conditions, and new stressors. Such new, anthropogenic stressors include the neonicotinoid class of crop-protecting agents, which have been implicated in the population declines of pollinating insects, including honeybees (Apis mellifera). The low-dose effects of these compounds on larval development and physiological responses have remained largely unknown. Over a period of 15 days, we provided syrup tainted with low levels (2 Β΅g/Lβˆ’1) of the neonicotinoid insecticide imidacloprid to beehives located in the field. We measured transcript levels by RNA sequencing and established lipid profiles using liquid chromatography coupled with mass spectrometry from worker-bee larvae of imidacloprid-exposed (IE) and unexposed, control (C) hives. Within a catalogue of 300 differentially expressed transcripts in larvae from IE hives, we detect significant enrichment of genes functioning in lipid-carbohydrate-mitochondrial metabolic networks. Myc-involved transcriptional response to exposure of this neonicotinoid is indicated by overrepresentation of E-box elements in the promoter regions of genes with altered expression. RNA levels for a cluster of genes encoding detoxifying P450 enzymes are elevated, with coordinated downregulation of genes in glycolytic and sugar-metabolising pathways. Expression of the environmentally responsive Hsp90 gene is also reduced, suggesting diminished buffering and stability of the developmental program. The multifaceted, physiological response described here may be of importance to our general understanding of pollinator health. Muscles, for instance, work at high glycolytic rates and flight performance could be impacted should low levels of this evolutionarily novel stressor likewise induce downregulation of energy metabolising genes in adult pollinators

    Illumination of Parainfluenza Virus Infection and Transmission in Living Animals Reveals a Tissue-Specific Dichotomy

    The parainfluenza viruses (PIVs) are highly contagious respiratory paramyxoviruses and a leading cause of lower respiratory tract (LRT) disease. Since no vaccines or antivirals exist, non-pharmaceutical interventions are the only means of control for these pathogens. Here we used bioluminescence imaging to visualize the spatial and temporal progression of murine PIV1 (Sendai virus) infection in living mice after intranasal inoculation or exposure by contact. A non-attenuated luciferase reporter virus (rSeV-luc(M-F*)) that expressed high levels of luciferase yet was phenotypically similar to wild-type Sendai virus in vitro and in vivo was generated to allow visualization. After direct intranasal inoculation, we unexpectedly observed that the upper respiratory tract (URT) and trachea supported robust infection under conditions that result in little infection or pathology in the lungs including a low inoculum of virus, an attenuated virus, and strains of mice genetically resistant to lung infection. The high permissivity of the URT and trachea to infection resulted in 100% transmission to naΓ―ve contact recipients, even after low-dose (70 PFU) inoculation of genetically resistant BALB/c donor mice. The timing of transmission was consistent with the timing of high viral titers in the URT and trachea of donor animals but was independent of the levels of infection in the lungs of donors. The data therefore reveals a disconnect between transmissibility, which is associated with infection in the URT, and pathogenesis, which arises from infection in the lungs and the immune response. Natural infection after transmission was universally robust in the URT and trachea yet limited in the lungs, inducing protective immunity without weight loss even in genetically susceptible 129/SvJ mice. Overall, these results reveal a dichotomy between PIV infection in the URT and trachea versus the lungs and define a new model for studies of pathogenesis, development of live virus vaccines, and testing of antiviral therapies

    Characterizing Emerging Canine H3 Influenza Viruses.

    The continual emergence of novel influenza A strains from non-human hosts requires constant vigilance and the need for ongoing research to identify strains that may pose a human public health risk. Since 1999, canine H3 influenza A viruses (CIVs) have caused many thousands or millions of respiratory infections in dogs in the United States. While no human infections with CIVs have been reported to date, these viruses could pose a zoonotic risk. In these studies, the National Institutes of Allergy and Infectious Diseases (NIAID) Centers of Excellence for Influenza Research and Surveillance (CEIRS) network collaboratively demonstrated that CIVs replicated in some primary human cells and transmitted effectively in mammalian models. While people born after 1970 had little or no pre-existing humoral immunity against CIVs, the viruses were sensitive to existing antivirals and we identified a panel of H3 cross-reactive human monoclonal antibodies (hmAbs) that could have prophylactic and/or therapeutic value. Our data predict these CIVs posed a low risk to humans. Importantly, we showed that the CEIRS network could work together to provide basic research information important for characterizing emerging influenza viruses, although there were valuable lessons learned
