    Loci Controlling Resistance to High Plains Virus and Wheat Streak Mosaic Virus in a B73 Ă— Mo17 Population of Maize

    High Plains disease has the potential to cause significant yield loss in susceptible corn (Zea mays L.) and wheat (Triticum aestivum L.) genotypes, especially in the central and western USA. The primary causal agent, High Plains virus (HPV), is vectored by wheat curl mite (WCM; Aceria tossicheila Keifer), which is also the vector of wheat streak mosaic virus (WSMV). In general, the two diseases occur together as a mixed infection in the field. The objective of this research was to characterize the inheritance of HPV and WSMV resistance using B73 (resistant to HPV and WSMV) Ă— Mo17 (moderately susceptible to HPV and WSMV) recombinant inbred lines. A population of 129 recombinant inbred lines scored for 167 molecular markers was used to evaluate resistance to WSMV and to a mixed infection of WSMV and HPV. Loci conferring resistance to systemic movement of WSMV in plants mapped to chromosomes 3, 6, and 10, consistent with the map position of wsm2, wsm1, and wsm3, respectively. Major genes for resistance to systemic spread of HPV in doubly infected plants mapped to chromosomes 3 and 6, coincident or tightly linked with the WSMV resistance loci. Analysis of doubly infected plants revealed that chromosome 6 had a major effect on HPV resistance, consistent with our previous analysis of B73 Ă— W64A and B73 Ă— Wf9 populations. Quantitative trait loci (QTL) affecting resistance to localized symptom development mapped to chromosomes 4 (umc66), 5 (bnl5.40), and 6 (umc85), and accounted for 24% of the phenotypic variation. Localized symptoms may reflect the amount of mite feeding or the extent of virus spread at the point of infection. Identification of cosegregating markers may facilitate selection for HPV and WSMV resistance in corn breeding programs

    Imaging of the unstable plaque: how far have we got?

    Rupture of unstable plaques may lead to myocardial infarction or stroke and is the leading cause of morbidity and mortality in western countries. Thus, there is a clear need for identifying these vulnerable plaques before the rupture occurs. Atherosclerotic plaques are a challenging imaging target as they are small and move rapidly, especially in the coronary tree. Many of the currently available imaging tools for clinical use still provide minimal information about the biological characteristics of plaques, because they are limited with respect to spatial and temporal resolution. Moreover, many of these imaging tools are invasive. The new generation of imaging modalities such as magnetic resonance imaging, nuclear imaging such as positron emission tomography and single photon emission computed tomography, computed tomography, fluorescence imaging, intravascular ultrasound, and optical coherence tomography offer opportunities to overcome some of these limitations.     A broadband FFT spectrometer for radio and millimeter astronomy

    The core architecture, tests in the lab and first results of a Fast Fourier Transform (FFT) spectrometer are described. It is based on a commercially available fast digital sampler (AC240) with an on-board Field Programmable Gate Array (FPGA). The spectrometer works continuously and has a remarkable total bandwidth of 1 GHz, resolved into 16384 channels. The data is sampled with 8 bits, yielding a dynamic range of 48 dB. An Allan time of more than 2000 s and an SFDR of 37 dB were measured.     Anti-tumour therapeutic efficacy of OX40L in murine tumour model

    OX40 ligand (OX40L), a member of TNF superfamily, is a co-stimulatory molecule involved in T cell activation. Systemic administration of mOX40L fusion protein significantly inhibited the growth of experimental lung metastasis and subcutaneous (s.c.) established colon (CT26) and breast (4T1) carcinomas. Vaccination with OX40L was significantly enhanced by combination treatment with intra-tumour injection of a disabled infectious single cycle-herpes simplex virus (DISC-HSV) vector encoding murine granulocyte macrophage-colony stimulating factor (mGM-CSF). Tumour rejection in response to OX40L therapy required functional CD4+ and CD8+ T cells and correlated with splenocyte cytotoxic T lymphocytes (CTLs) activity against the AH-1 gp70 peptide of the tumour associated antigen expressed by CT26 cells. These results demonstrate the potential role of the OX40L in cancer immunotherapy

    Genetic Correlates of Individual Differences in Sleep Behavior of Free-Living Great Tits (Parus major)

    Within populations, free-living birds display considerable variation in observable sleep behaviors, reflecting dynamic interactions between individuals and their environment. Genes are expected to contribute to repeatable between-individual differences in sleep behaviors, which may be associated with individual fitness. We identified and genotyped polymorphisms in nine candidate genes for sleep, and measured five repeatable sleep behaviors in free-living great tits (Parus major), partly replicating a previous study in blue tits (Cyanistes caeruleus). Microsatellites in the CLOCK and NPAS2 clock genes exhibited an association with sleep duration relative to night length, and morning latency to exit the nest box, respectively. Furthermore, microsatellites in the NPSR1 and PCSK2 genes associated with relative sleep duration and proportion of time spent awake at night, respectively. Given the detection rate of associations in the same models run with random markers instead of candidate genes, we expected two associations to arise by chance. The detection of four associations between candidate genes and sleep, however, suggests that clock genes, a clock-related gene, or a gene involved in the melanocortin system, could play key roles in maintaining phenotypic variation in sleep behavior in avian populations. Knowledge of the genetic architecture underlying sleep behavior in the wild is important because it will enable ecologists to assess the evolution of sleep in response to selection

    The Origin of Cornbelt Maize: The Isozyme Evidence

    Historical records show t hat the Midwestern dent corns of the United States originated from hybridization of two landraces, Northern Flint and Southern Dent. We examined the origin of Southern and Midwestern Dents by means of isozyme electrophoresis. Isozyme genotypes were determined for 23 loci in 12 plants each of 32 accessions of Southern Dent. Previously published isozyme data for maize landraces of Mexico and North America and for U.S. Midwestern Dents were included for comparative purposes. The data show that Northern Flint and Southern Dent are among the isozymically most divergent maize landraces. Nei’s genetic identities between populations of these two landraces are very low for conspecific populations (ca. 0.80). Southern Dent of the southeastern U.S. appears closely related to similar dent corns of southern Mexico, supporting a previously published hypothesis that U.S. Southern Dent is largely derived from the dent corns of southern Mexico. The Midwestern Dents, which resulted from crosses of Southern Dent and Northern Flint, are much more like Southern Dent than Northern Flint in their isozyme profile. Similarly, public inbreds show greater affinity to Southern Dent with the exception of sweet corn lines, which resemble Northern Flint in their isozyme allele frequencies. North American public inbreds do not contain appreciable isozymic variation beyond that found in Northern Flint and Southern Dent
