    The Prosody of Directive Speech Acts: Pragmastylistic Aspect (On the Material of the Modern German Language)

    The article represents the results of a phonetic experiment devoted to the study of pragmatically, and the stylistically determined manifestation of directive speech acts in the modern German language. The experiment allowed establishing the prosodic invariant of the directive speech act and describing variant patterns realized in diverse situational conditions which demonstrated a different degree of categoricalness and revealed their differential character on the level of prosodic expressive means. The research showed that pragmatic identification and stylistic differentiation of directive speech acts is realized utilizing all prosodic parameters - pitch, intensity and tempo. The complex analysis of pitch, intensity and tempo characteristics specific for situational variants of the directive illocutive allowed defining the level of involvement of these prosodic means in the functional realization of directive speech impact

    The article discusses the application of intelligent transport systems for railway transport. The purpose of developing intelligent transport systems on railway transport is to strictly comply with the requirements for ensuring transport safety, reducing the level of environmental impact, significantly improving the efficiency of production activities. The software makes automatic accounting, control and analysis of fuel and energy resources consumption on the basis of the obtained data. At the same time, the dynamics of changes in fuel consumption indicators, actual and estimated costs of diesel fuel are visually reflected in real time on the monitor screen. If necessary, the system will help to identify the reasons for non-matching of these parameters and quickly contact the locomotive team to provide qualified assistance in their elimination. It is important that the hardware structure and the structure of the system software expand functionality, providing continuous operation and repair work, increasing the number of measured parameters, creating a closed fuel consumption control system in a locomotive economy.     Diabetes mellitus in combination with COVID-19: modern views on therapy

    Diabetic patients are in the spotlight from the early stages of a pandemic, as growing epidemiological data show they are at higher risk for severe clinical outcomes from COVID-19. As the global COVID-19 pandemic continues to evolve, it is also becoming increasingly apparent that the interactions between COVID-19 and diabetes mellitus (DM) are complex pathophysiological mechanisms. The outcome of COVID-19 is more severe in people with DM, which has the potential to accelerate the onset of acute metabolic complications of DM such as diabetic ketoacidosis and hyperglycemia. These mechanisms underlying these associations remain unclear, but they likely include the angiotensin converting enzyme receptor 2, a binding site for SARS-CoV-2, which is expressed in key metabolic organs such as in the pancreas, in particular in Ξ²-cells. The potential Ξ²-cell tropism of SARS-CoV-2 can damage cells and impair insulin secretion, causing hyperglycemia and ketoacidosis. Understanding the bidirectional interaction between DM and COVID-19 will be critical to protecting and treating people with DM. Current epidemiological data on COVID-19 do not support the hypothesis that diabetic patients are at increased risk of infection compared to the general population. To date, it has been established that decompensated DM is an independent factor that aggravates the course of coronavirus infection and significantly increases the risk of a fatal outcome of the disease. The review provides a brief summary of the evolution of pathogenetic and clinical aspects for understanding the mechanisms of this pathological tandem, as well as therapeutic strategies for treating patients with COVID-19 and DM. As the incidence of DM continues to rise globally, more than ever, diabetes prevention and control must be a priority for health systems around the world

    DEVELOPMENT OF A TECHNICAL AND TECHNOLOGICAL SOLUTION FOR THE PRODUCTION OF CARROT NECTAR

    The modern food market forms a need for two trends for the development, production and sale of products: taking into account the demand of consumers and taking into account lack of irreplaceable (functional) micronutrients in the diet of different population groups. It is advisable to develop technical and technological solutions for innovative ideas at a higher institution, which are tested in business incubators (small innovative enterprises) with the interaction of scientists and production workers. The study objects included: the carrots of different varieties of the crop of 2015-2017 grown in the climatic conditions of the Kemerovo region; the syrup based on Lesovichok plant raw materials; pectolytic, amylolytic and cytolytic enzymatic preparations. Modern research methods have been used in the study. The antioxidant activity was determined using the coulometric method; the content of dry soluble substances - using the refractometric method; the size of carrot particles in the semi-finished products and the finished product - by means of microscopy using a CT-2200 electron microscope; the strength characteristics of raw materials - using Structurometer St-1. As a result of the studies, a product with a high antioxidant activity - carrot nectar developed using hybrid technologies - has been obtained. On the basis of the results of the study of the structural and mechanical properties of carrots, two-stage grinding has been proposed with the subsequent biotechnological processing of raw materials (semi-finished products), which provides specified consumer properties.     Registration of atmospheric neutrinos with the Baikal neutrino telescope

    We present first neutrino induced events observed with a deep underwater neutrino telescope. Data from 70 days effective life time of the BAIKAL prototype telescope NT-96 have been analyzed with two different methods. With the standard track reconstruction method, 9 clear upward muon candidates have been identified, in good agreement with 8.7 events expected from Monte Carlo calculations for atmospheric neutrinos. The second analysis is tailored to muons coming from close to the opposite zenith. It yields 4 events, compared to 3.5 from Monte Carlo expectations. From this we derive a 90 % upper flux limit of 1.1 * 10^-13 cm^-2 sec^-1 for muons in excess of those expected from atmospheric neutrinos with zenith angle > 150 degrees and energy > 10GeV.Comment: 20 pages, 11 figure

    Diffeomorphism Invariance in the Hamiltonian formulation of General Relativity

    It is shown that when the Einstein-Hilbert Lagrangian is considered without any non-covariant modifications or change of variables, its Hamiltonian formulation leads to results consistent with principles of General Relativity. The first-class constraints of such a Hamiltonian formulation, with the metric tensor taken as a canonical variable, allow one to derive the generator of gauge transformations, which directly leads to diffeomorphism invariance. The given Hamiltonian formulation preserves general covariance of the transformations derivable from it. This characteristic should be used as the crucial consistency requirement that must be met by any Hamiltonian formulation of General Relativity.Comment: 13 page

    Cohesin mutations alter DNA damage repair and chromatin structure and create therapeutic vulnerabilities in MDS/AML

    The cohesin complex plays an essential role in chromosome maintenance and transcriptional regulation. Recurrent somatic mutations in the cohesin complex are frequent genetic drivers in cancer including myelodysplatic syndromes (MDS) and acute myeloid leukemia (AML). Here, using genetic dependency screens of STAG2-mutant AML, we identified DNA damage repair and replication as genetic dependencies in cohesin-mutant cells. We demonstrated increased levels of DNA damage and sensitivity of cohesin-mutant cells to PARP inhibition. We developed a mouse model of MDS in which Stag2 mutations arise as clonal secondary lesions in the background of clonal hematopoiesis driven by Tet2 mutations, and demonstrated selective depletion of cohesin-mutant cells with PARP inhibition in vivo. Finally, we demonstrated a shift from STAG2- to STAG1-containing cohesin complexes in cohesin-mutant cells, which is associated with longer DNA loop extrusion, more intermixing of chromatin compartments, and increased interaction with PARP and RPA proteins.
