    Synthetic biology - the next phase of biotechnology and genetic engineering. TAB-Fokus

    Scientific and technological progress allows to genetically redesign natural organisms in ever more profound ways (synbio in the broader sense). In the long term, the aim is to create artificial biological systems (synbio in the narrow sense). Application areas are chemical and energy production, environmental protection as well as the medical sector. Considering the current, early state of research and development, it is impossible to forecast reliably which approaches of synbio will prevail against procedures that make use of the available biological diversity or that are limited to more subtle interventions. Risk assessment and the evaluation of substantially modified organisms will require the development and exploration of new methods and procedures. Opportunities for different societal groups to participate in the responsible development of synbio are ranging from stakeholder involvement in setting research agendas to DIY biology. Dealing with intellectual property within the framework of an increasingly digital economy will also represent a major challenge for the future use of synbio

    Wissenschaftliche und technologische Fortschritte ermöglichen einen zunehmend tiefer greifenden genetischen Umbau natürlicher Organismen (Synbio i. w. S.), perspektivisch wird die Schaffung künstlicher biologischer Systeme (Synbio i. e. S.) anvisiert. Anwendungsbereiche liegen in der Chemie- und Energieproduktion, in der Medizin und im Umweltschutz. Welche Ansätze der Synbio sich durchsetzen werden gegenüber Verfahren, die die vorhandene biologische Vielfalt nutzen oder sich auf subtile Eingriffe beschränken, ist beim heutigen Stand von Forschung und Entwicklung nicht verlässlich prognostizierbar. Für die Risikoabschätzung und -bewertung substanziell veränderter Organismen müssen neue Methoden und Verfahren entwickelt und erprobt werden. Die Partizipationsmöglichkeiten der Gesellschaft an einer verantwortungsvollen Weiterentwicklung der Synbio reichen von einer Stakeholderbeteiligung an der Forschungsprogrammatik bis zur DIY-Biologie. Der Umgang mit geistigem Eigentum unter den Bedingungen einer zunehmend digitalen Ökonomie stellt eine der zentralen Zukunftsherausforderungen auch für die Nutzung der Synbio dar

    Learning through social spaces: migrant women and lifelong learning in post-colonial London

    This article shows how migrant women engage in learning through social spaces. It argues that such spaces are little recognised, and that there are multiple ways in which migrant women construct and negotiate their informal learning through socialising with other women in different informal modes. Additionally, the article shows how learning is shaped by the socio-political, geographical and multicultural context of living in London, outlining ways in which gendered and racialised identities shape, construct and constrain participation in lifelong learning. The article shows that one way in which migrant women resist (post)colonial constructions of difference is by engaging in informal and non-formal lifelong learning, arguing that the benefits are (at least) two-fold. The women develop skills (including language skills) but also use their informal learning to develop what is referred to in this article as 'relational capital'. The article concludes that informal lifelong learning developed through social spaces can enhance a sense of belonging for migrant women

    The sustainability conundrum of fishmeal substitution by plant ingredients in shrimp feeds

    Aquaculture is central in meeting expanding global demands for shrimp consumption. Consequently, increasing feed use is mainly responsible for the overall environmental impact of aquaculture production. Significant amounts of fishmeal are included in shrimp diets, causing dependency on finite marine resources. Driven by economic incentives, terrestrial plant ingredients are widely viewed as sustainable alternatives. Incremental fishmeal substitution by plant ingredients in shrimp feed was modeled and effects on marine and terrestrial resources such as fish, land, freshwater, nitrogen, and phosphorus were assessed. We find that complete substitution of 20–30% fishmeal totals could lead to increasing demand for freshwater (up to 63%), land (up to 81%), and phosphorus (up to 83%), while other substitution rates lead to proportionally lower impacts. These findings suggest additional pressures on essential agricultural resources with associated socio-economic and environmental effects as a trade-off to pressures on finite marine resources. Even though the production of shrimp feed (or aquafeed in general) utilizes only a small percentage of the global crop production, the findings indicate that the sustainability of substituting fishmeal by plant ingredients should not be taken for granted, especially since aquaculture has been one of the fastest growing food sectors. Therefore, the importance of utilizing by-products and novel ingredients such as microbial biomass, algae, and insect meals in mitigating the use of marine and terrestrial resources is discussed

    Hygienic quality of dehydrated aromatic herbs marketed in Southern Portugal

    Dehydrated aromatic herbs are highly valued ingredients, widely used at home level and by food processing industry, frequently added to a great number of recipes in the Mediterranean countries. Despite being considered low-moisture products and classified as GRAS, during pre and post-harvesting stages of production they are susceptible of microbial contamination. In Europe an increasing number of food recalls and disease outbreaks associated with dehydrated herbs have been reported in recent years. In this study the microbial quality of 99 samples of aromatic herbs (bay leaves, basil, coriander, oregano, parsley, Provence herbs, rosemary and thyme) collected from retails shops in the region of Algarve (Southern Portugal) was assessed. All the samples were tested by conventional methods and were assayed for the total count of aerobic mesophilic microorganisms, Salmonella spp., Escherichia coli, coagulase-positive staphylococci and filamentous fungi. Almost 50 % of the herbs did not exceed the aerobic mesophilic level of 104 CFU/g. The fungi count regarded as unacceptable (106 CFU/g) was not found in any of the tested herbs, while 84 % of the samples ranged from ≤102 to 104 CFU/g. No sample was positive for the presence of Salmonella spp., Escherichia coli and staphylococci. The results are in compliance with the European Commission criteria although they point out to the permanent need of surveillance on the good standards of handling/cooking practices as well as the importance of avoiding contamination at production, retailing and distribution. The microbiological hazards associated with the pathogenic and toxigenic microbiota of dried herbs remain as a relevant public health issue, due to the fact that they are added to foods not submitted to any following lethal procedure.     Comparative evaluation of direct plating and most probable number for enumeration of low levels of Listeria monocytogenes in naturally contaminated ice cream products

    AbstractA precise and accurate method for enumeration of low level of Listeria monocytogenes in foods is critical to a variety of studies. In this study, paired comparison of most probable number (MPN) and direct plating enumeration of L. monocytogenes was conducted on a total of 1730 outbreak-associated ice cream samples that were naturally contaminated with low level of L. monocytogenes. MPN was performed on all 1730 samples. Direct plating was performed on all samples using the RAPID'L.mono (RLM) agar (1600 samples) and agar Listeria Ottaviani and Agosti (ALOA; 130 samples). Probabilistic analysis with Bayesian inference model was used to compare paired direct plating and MPN estimates of L. monocytogenes in ice cream samples because assumptions implicit in ordinary least squares (OLS) linear regression analyses were not met for such a comparison. The probabilistic analysis revealed good agreement between the MPN and direct plating estimates, and this agreement showed that the MPN schemes and direct plating schemes using ALOA or RLM evaluated in the present study were suitable for enumerating low levels of L. monocytogenes in these ice cream samples. The statistical analysis further revealed that OLS linear regression analyses of direct plating and MPN data did introduce bias that incorrectly characterized systematic differences between estimates from the two methods

    Pion and proton showers in the CALICE scintillator-steel analogue hadron calorimeter

    Showers produced by positive hadrons in the highly granular CALICE scintillator-steel analogue hadron calorimeter were studied. The experimental data were collected at CERN and FNAL for single particles with initial momenta from 10 to 80 GeV/c. The calorimeter response and resolution and spatial characteristics of shower development for proton- and pion-induced showers for test beam data and simulations using Geant4 version 9.6 are compared.Comment: 26 pages, 16 figures, JINST style, changes in the author list, typos corrected, new section added, figures regrouped.     A meta-analysis of long-term effects of conservation agriculture on maize grain yield under rain-fed conditions

    Conservation agriculture involves reduced tillage, permanent soil cover and crop rotations to enhance soil fertility and to supply food from a dwindling land resource. Recently, conservation agriculture has been promoted in Southern Africa, mainly for maize-based farming systems. However, maize yields under rain-fed conditions are often variable. There is therefore a need to identify factors that influence crop yield under conservation agriculture and rain-fed conditions. Here, we studied maize grain yield data from experiments lasting 5 years and more under rain-fed conditions. We assessed the effect of long-term tillage and residue retention on maize grain yield under contrasting soil textures, nitrogen input and climate. Yield variability was measured by stability analysis. Our results show an increase in maize yield over time with conservation agriculture practices that include rotation and high input use in low rainfall areas. But we observed no difference in system stability under those conditions. We observed a strong relationship between maize grain yield and annual rainfall. Our meta-analysis gave the following findings: (1) 92% of the data show that mulch cover in high rainfall areas leads to lower yields due to waterlogging; (2) 85% of data show that soil texture is important in the temporal development of conservation agriculture effects, improved yields are likely on well-drained soils; (3) 73% of the data show that conservation agriculture practices require high inputs especially N for improved yield; (4) 63% of data show that increased yields are obtained with rotation but calculations often do not include the variations in rainfall within and between seasons; (5) 56% of the data show that reduced tillage with no mulch cover leads to lower yields in semi-arid areas; and (6) when adequate fertiliser is available, rainfall is the most important determinant of yield in southern Africa. It is clear from our results that conservation agriculture needs to be targeted and adapted to specific biophysical conditions for improved impact

    Hadron shower decomposition in the highly granular CALICE analogue hadron calorimeter

    The spatial development of hadronic showers in the CALICE scintillator-steel analogue hadron calorimeter is studied using test beam data collected at CERN and FNAL for single positive pions and protons with initial momenta in the range from 10 to 80 GeV/c. Both longitudinal and radial development of hadron showers are parametrised with two-component functions. The parametrisation is fit to test beam data and simulations using the QGSP_BERT and FTFP_BERT physics lists from Geant4 version 9.6. The parameters extracted from data and simulated samples are compared for the two types of hadrons. The response to pions and the ratio of the non-electromagnetic to the electromagnetic calorimeter response, h/e, are estimated using the extrapolation and decomposition of the longitudinal profiles.Comment: 38 pages, 19 figures, 5 tables; author list changed; submitted to JINS

    A critical experimental study of the classical tactile threshold theory

    <p>Abstract</p> <p>Background</p> <p>The tactile sense is being used in a variety of applications involving tactile human-machine interfaces. In a significant number of publications the classical threshold concept plays a central role in modelling and explaining psychophysical experimental results such as in stochastic resonance (SR) phenomena. In SR, noise enhances detection of sub-threshold stimuli and the phenomenon is explained stating that the required amplitude to exceed the sensory threshold barrier can be reached by adding noise to a sub-threshold stimulus. We designed an experiment to test the validity of the classical vibrotactile threshold. Using a second choice experiment, we show that individuals can order sensorial events below the level known as the classical threshold. If the observer's sensorial system is not activated by stimuli below the threshold, then a second choice could not be above the chance level. Nevertheless, our experimental results are above that chance level contradicting the definition of the classical tactile threshold.</p> <p>Results</p> <p>We performed a three alternative forced choice detection experiment on 6 subjects asking them first and second choices. In each trial, only one of the intervals contained a stimulus and the others contained only noise. According to the classical threshold assumptions, a correct second choice response corresponds to a guess attempt with a statistical frequency of 50%. Results show an average of 67.35% (STD = 1.41%) for the second choice response that is not explained by the classical threshold definition. Additionally, for low stimulus amplitudes, second choice correct detection is above chance level for any detectability level.</p> <p>Conclusions</p> <p>Using a second choice experiment, we show that individuals can order sensorial events below the level known as a classical threshold. If the observer's sensorial system is not activated by stimuli below the threshold, then a second choice could not be above the chance level. Nevertheless, our experimental results are above that chance level. Therefore, if detection exists below the classical threshold level, then the model to explain the SR phenomenon or any other tactile perception phenomena based on the psychophysical classical threshold is not valid. We conclude that a more suitable model of the tactile sensory system is needed.</p
