    Shade as enrichment : testing preferences for shelter in two model fish species

Zebrafish showed no preference for either shelter, whereas sticklebacks showed a preference for both shelter types over open areas and for shade over plants.     Pathogen-mediated selection for MHC variability in wild zebrafish

    ABSTRACT Hypothesis: Genes of the major histocompatibility complex (MHC) show high variability, which is believed to arise through pathogen-mediated selection. Organism: Zebrafish (Danio rerio) from seven independent natural populations. Location: Bangladesh: two sites in Khulna District in the River Ganges drainage, and five sites in Mymensingh District in the River Brahmaputra drainage. Methods: We investigated the relationship of parasite abundance and diversity with allelic variability of the MHC class II B gene in natural populations of zebrafish. A sample of 120 zebrafish was screened for metazoan parasites, and a subsample of 77 was assessed for immunogenetic variation. In total, 2475 metazoan parasites from 23 taxa were identified and 13 unique MHC class IIB alleles were isolated. Conclusion: We found no evidence for a heterozygote advantage, and overall there was lower than expected heterozygosity of MHC alleles. We identified lower parasite abundance and diversity with specific MHC alleles, and this pattern varied between geographic regions. Results suggest that MHC variability in zebrafish is maintained by frequency-dependent and fluctuating selection

    Genetic analysis of male reproductive success in relation to density in the zebrafish, Danio rerio

    BACKGROUND: We used behavioural and genetic data to investigate the effects of density on male reproductive success in the zebrafish, Danio rerio. Based on previous measurements of aggression and courtship behaviour by territorial males, we predicted that they would sire more offspring than non-territorial males. RESULTS: Microsatellite analysis of paternity showed that at low densities territorial males had higher reproductive success than non-territorial males. However, at high density territorial males were no more successful than non-territorials and the sex difference in the opportunity for sexual selection, based on the parameter I(mates), was low. CONCLUSION: Male zebrafish exhibit two distinct mating tactics; territoriality and active pursuit of females. Male reproductive success is density dependent and the opportunity for sexual selection appears to be weak in this species

    The Influence of Behavioral, Social, and Environmental Factors on Reproducibility and Replicability in Aquatic Animal Models

    The publication of reproducible, replicable, and translatable data in studies utilizing animal models is a scientific, practical, and ethical necessity. This requires careful planning and execution of experiments and accurate reporting of results. Recognition that numerous developmental, environmental, and test-related factors can affect experimental outcomes is essential for a quality study design. Factors commonly considered when designing studies utilizing aquatic animal species include strain, sex, or age of the animal; water quality; temperature; and acoustic and light conditions. However, in the aquatic environment, it is equally important to consider normal species behavior, group dynamics, stocking density, and environmental complexity, including tank design and structural enrichment. Here, we will outline normal species and social behavior of 2 commonly used aquatic species: zebrafish (Danio rerio) and Xenopus (X. laevis and X. tropicalis). We also provide examples as to how these behaviors and the complexity of the tank environment can influence research results and provide general recommendations to assist with improvement of reproducibility and replicability, particularly as it pertains to behavior and environmental complexity, when utilizing these popular aquatic models. © The Author(s) 2020.     The development and validation of a scoring tool to predict the operative duration of elective laparoscopic cholecystectomy

    Background: The ability to accurately predict operative duration has the potential to optimise theatre efficiency and utilisation, thus reducing costs and increasing staff and patient satisfaction. With laparoscopic cholecystectomy being one of the most commonly performed procedures worldwide, a tool to predict operative duration could be extremely beneficial to healthcare organisations. Methods: Data collected from the CholeS study on patients undergoing cholecystectomy in UK and Irish hospitals between 04/2014 and 05/2014 were used to study operative duration. A multivariable binary logistic regression model was produced in order to identify significant independent predictors of long (> 90 min) operations. The resulting model was converted to a risk score, which was subsequently validated on second cohort of patients using ROC curves. Results: After exclusions, data were available for 7227 patients in the derivation (CholeS) cohort. The median operative duration was 60 min (interquartile range 45–85), with 17.7% of operations lasting longer than 90 min. Ten factors were found to be significant independent predictors of operative durations > 90 min, including ASA, age, previous surgical admissions, BMI, gallbladder wall thickness and CBD diameter. A risk score was then produced from these factors, and applied to a cohort of 2405 patients from a tertiary centre for external validation. This returned an area under the ROC curve of 0.708 (SE = 0.013, p  90 min increasing more than eightfold from 5.1 to 41.8% in the extremes of the score. Conclusion: The scoring tool produced in this study was found to be significantly predictive of long operative durations on validation in an external cohort. As such, the tool may have the potential to enable organisations to better organise theatre lists and deliver greater efficiencies in care

    Background Some high-income countries have deployed fourth doses of COVID-19 vaccines, but the clinical need, effectiveness, timing, and dose of a fourth dose remain uncertain. We aimed to investigate the safety, reactogenicity, and immunogenicity of fourth-dose boosters against COVID-19.Methods The COV-BOOST trial is a multicentre, blinded, phase 2, randomised controlled trial of seven COVID-19 vaccines given as third-dose boosters at 18 sites in the UK. This sub-study enrolled participants who had received BNT162b2 (Pfizer-BioNTech) as their third dose in COV-BOOST and randomly assigned them (1:1) to receive a fourth dose of either BNT162b2 (30 µg in 0·30 mL; full dose) or mRNA-1273 (Moderna; 50 µg in 0·25 mL; half dose) via intramuscular injection into the upper arm. The computer-generated randomisation list was created by the study statisticians with random block sizes of two or four. Participants and all study staff not delivering the vaccines were masked to treatment allocation. The coprimary outcomes were safety and reactogenicity, and immunogenicity (antispike protein IgG titres by ELISA and cellular immune response by ELISpot). We compared immunogenicity at 28 days after the third dose versus 14 days after the fourth dose and at day 0 versus day 14 relative to the fourth dose. Safety and reactogenicity were assessed in the per-protocol population, which comprised all participants who received a fourth-dose booster regardless of their SARS-CoV-2 serostatus. Immunogenicity was primarily analysed in a modified intention-to-treat population comprising seronegative participants who had received a fourth-dose booster and had available endpoint data. This trial is registered with ISRCTN, 73765130, and is ongoing.Findings Between Jan 11 and Jan 25, 2022, 166 participants were screened, randomly assigned, and received either full-dose BNT162b2 (n=83) or half-dose mRNA-1273 (n=83) as a fourth dose. The median age of these participants was 70·1 years (IQR 51·6–77·5) and 86 (52%) of 166 participants were female and 80 (48%) were male. The median interval between the third and fourth doses was 208·5 days (IQR 203·3–214·8). Pain was the most common local solicited adverse event and fatigue was the most common systemic solicited adverse event after BNT162b2 or mRNA-1273 booster doses. None of three serious adverse events reported after a fourth dose with BNT162b2 were related to the study vaccine. In the BNT162b2 group, geometric mean anti-spike protein IgG concentration at day 28 after the third dose was 23 325 ELISA laboratory units (ELU)/mL (95% CI 20 030–27 162), which increased to 37 460 ELU/mL (31 996–43 857) at day 14 after the fourth dose, representing a significant fold change (geometric mean 1·59, 95% CI 1·41–1·78). There was a significant increase in geometric mean anti-spike protein IgG concentration from 28 days after the third dose (25 317 ELU/mL, 95% CI 20 996–30 528) to 14 days after a fourth dose of mRNA-1273 (54 936 ELU/mL, 46 826–64 452), with a geometric mean fold change of 2·19 (1·90–2·52). The fold changes in anti-spike protein IgG titres from before (day 0) to after (day 14) the fourth dose were 12·19 (95% CI 10·37–14·32) and 15·90 (12·92–19·58) in the BNT162b2 and mRNA-1273 groups, respectively. T-cell responses were also boosted after the fourth dose (eg, the fold changes for the wild-type variant from before to after the fourth dose were 7·32 [95% CI 3·24–16·54] in the BNT162b2 group and 6·22 [3·90–9·92] in the mRNA-1273 group).Interpretation Fourth-dose COVID-19 mRNA booster vaccines are well tolerated and boost cellular and humoral immunity. Peak responses after the fourth dose were similar to, and possibly better than, peak responses after the third dose

    Though the zebrafish is an important laboratory model organism, little is known about its natural ecology, and few studies have been conducted on wild populations. It is a small, shoaling cyprinid, native to the floodplains of the Indian subcontinent. A survey conducted in Bangladesh showed that it is most abundant in shallow lakes, ponds and ditches, often associated with rice cultivation. Length-frequency analysis indicates that the zebrafish is an annual species, recruitment being linked to the monsoon season. Its diet, based on gut content analysis, consists primarily of zooplankton and insects. Zebrafish are group spawners and egg scatterers, though females are choosy with respect to sites for oviposition and males defend territories around such sites. The reproductive success of both females and males is affected by density; females produce smaller clutches at higher densities; at low densities territorial males achieve greater reproductive success than non-territorial males, but at higher densities they are no more successful than non-territorials. Female mating preferences, when assessed in the absence of male-male competition, do not correspond to male dominance. The opportunity for selection appears to be weak in zebrafish. Zebrafish show innate and learned preferences for visual cues, both in social and foraging contexts. They shoal preferentially with their own colour pattern, but when individuals are cross-reared with groups of an alternative colour pattern, they prefer to shoal with fish of the appearance with which they were reared; there appears to be a strong learned component to species recognition. In a foraging context, zebrafish display both innate and learned colour preferences. When fish were reared on diets consisting of different colours, each group respond most strongly to red, although there was also an effect of conditioning.     Hypothesis: The genetic benefits of female mating preferences can be additive (good alleles'), non-additive ('compatible alleles') or both, and the importance of each may be population dependent. Organism: Threespine stickleback (Gasterosteus aculeatus) from two independent natural populations. Location: Two adjacent sites in Scotland, one a pond with high stickleback density and high parasite prevalence, the other a river with low stickleback density and fish predators. Methods: In vitro fertilizations in a North Carolina Type II breeding design to estimate the contribution of additive and non-additive genetic variation to early offspring survival. Results: In both populations we detected significant additive maternal effects. These effects were largely explained by variance in egg size, though not entirely so. In the pond population there were significant male additive genetic effects on offspring survival, implicating a role for 'good alleles', but not non-additive effects. In the river population there were significant additive and non-additive effects, implicating a role both for 'good alleles' and genetic compatibility. Different populations of the same species may vary in the mechanism by which sexual selection operates.</p
