Ultrathin ceramic membranes as scaffolds for functional cell coculture models on a biomimetic scale
Epithelial tissue serves as an interface between biological compartments. Many in vitro epithelial cell models have been developed as an alternative to animal experiments to answer a range of research questions. These in vitro models are grown on permeable two-chamber systems; however, commercially available, polymer-based cell culture inserts are around 10âÎŒm thick. Since the basement membrane found in biological systems is usually less than 1âÎŒm thick, the 10-fold thickness of cell culture inserts is a major limitation in the establishment of realistic models. In this work, an alternative insert, accommodating an ultrathin ceramic membrane with a thickness of only 500ânm (i.e., the Silicon nitride Microporous Permeable Insert [SIMPLI]-well), was produced and used to refine an established human alveolar barrier coculture model by both replacing the conventional inserts with the SIMPLI-well and completing it with endothelial cells. The structuralâfunctional relationship of the model was evaluated, including the translocation of gold nanoparticles across the barrier, revealing a higher translocation if compared to corresponding polyethylene terephthalate (PET) membranes. This study demonstrates the power of the SIMPLI-well system as a scaffold for epithelial tissue cell models on a truly biomimetic scale, allowing construction of more functionally accurate models of human biological barriers
Consequences of the Timing of Menarche on Female Adolescent Sleep Phase Preference
Most parents experience their children's puberty as a dramatic change in family life. This is not surprising considering the dynamics of physical and psychosocial maturation which occur during adolescence. A reasonable question, particularly from the parents' perspective, is: when does this vibrant episode end and adulthood finally start? The aim of the present study was to assess the relationship between puberty and the changes in sleep phase preferences during female maturation and adulthood by a cross-sectional survey. The results from 1'187 females aged 5 to 51 years based on self-report measures of sleep preferences on weekdays and on free days as well as the occurrence of menarche, show that in contrast to prepubertal children, adolescent females exhibit a striking progression in delaying their sleep phase preference until 5 years after menarche. Thereafter, the sleep phase preference switches to advancing. The current study provides evidence that a clear shift in sleep-wake cycles temporally linked to menarche heralds the beginning of âadult-likeâ sleep-wake behaviour in women and can be used as a (chrono)biological marker for the onset of adulthood
AirâLiquid Interface In Vitro Models for Respiratory Toxicology Research: Consensus Workshop and Recommendations
In vitro airâliquid interface (ALI) cell culture models can potentially be used to assess inhalation toxicology endpoints and are usually considered, in terms of relevancy, between classic (i.e., submerged) in vitro models and animal-based models. In some situations that need to be clearly defined, ALI methods may represent a complement or an alternative option to in vivo experimentations or classic in vitro methods. However, it is clear that many different approaches exist and that only very limited validation studies have been carried out to date. This means comparison of data from different methods is difficult and available methods are currently not suitable for use in regulatory assessments. This is despite inhalation toxicology being a priority area for many governmental organizations. In this setting, a 1-day workshop on ALI in vitro models for respiratory toxicology research was organized in Paris in March 2016 to assess the situation and to discuss what might be possible in terms of validation studies. The workshop was attended by major parties in Europe and brought together more than 60 representatives from various academic, commercial, and regulatory organizations. Following plenary, oral, and poster presentations, an expert panel was convened to lead a discussion on possible approaches to validation studies for ALI inhalation models. A series of recommendations were made and the outcomes of the workshop are reported
Carbon Dioxide Reduction in an Optimization Model. A Case Study for Switzerland
At the Earth Summit in Rio, Switzerland was among those countries that declared to stabilize their carbon dioxide emissions. In order to achieve such a target, the Swiss energy system is analyzed and, based on this analysis, a dynamic optimization model is developed to evaluate technological C02 reduction measures like fuel or technology substitutions. Specialties of the model are the inclusion of a general economic part in addition to the energy part and a modeling of stocks of capital equipment in the neo-Austrian tradition. The scenario results suggest that, even without technological progress, a stabilization of carbon dioxide emissions is feasible.

Same data, different conclusions: Radical dispersion in empirical results when independent analysts operationalize and test the same hypothesis
In this crowdsourced initiative, independent analysts used the same dataset to test two hypotheses regarding the effects of scientistsâ gender and professional status on verbosity during group meetings. Not only the analytic approach but also the operationalizations of key variables were left unconstrained and up to individual analysts. For instance, analysts could choose to operationalize status as job title, institutional ranking, citation counts, or some combination. To maximize transparency regarding the process by which analytic choices are made, the analysts used a platform we developed called DataExplained to justify both preferred and rejected analytic paths in real time. Analyses lacking sufficient detail, reproducible code, or with statistical errors were excluded, resulting in 29 analyses in the final sample. Researchers reported radically different analyses and dispersed empirical outcomes, in a number of cases obtaining significant effects in opposite directions for the same research question. A Boba multiverse analysis demonstrates that decisions about how to operationalize variables explain variability in outcomes above and beyond statistical choices (e.g., covariates). Subjective researcher decisions play a critical role in driving the reported empirical results, underscoring the need for open data, systematic robustness checks, and transparency regarding both analytic paths taken and not taken. Implications for organizations and leaders, whose decision making relies in part on scientific findings, consulting reports, and internal analyses by data scientists, are discussed
